        혈관평활근세포에서 산화에너지대사 억제에 의한 아밀로이드전구단백질 대사의 변화

        Background: A reduction in the activity of cytochrome c oxidase(COX) has been recently identified in mitochondria from platelets and postmortem brain tissue of AD patients Sodium azide (NaN₃). a COX inhibitor, is an effective chemical agent producing energy shortage and oxidative stress both in vitro and in vivo system Furthermore it has been suggested that vascular compromise could be either directly involved AD pathogenesis or indirectly associated with triggering pathogenetic events leading to AD This study was performed to investigate amyloid precursor protein (APP) metabolism by inhibition of mitochondrial energy metabolism in cultured vascular smooth muslce cells (VSMCs) Materials and Methods: VSMCs isolated from the aorta of seven weeks old Spraque-Dawley rat were treated with NaN₃in a low concentration (100-500μM) or in a high concentration (1-100mM) Cellular proliferation and viability were determined by MTS (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenly)-2H-tetrazolium) assay Cellular APP was detected with N-terminal specific antibody 22C11. Celldeath was determined by observation of morphology and terminal deoxynucleotidyl transferase-mediated dUTP-fluorescein nick end labeling stain (TUNEL) We used ginkgo biloba extract(EGb761) and melatonin as anti-oxidants to investigate the mechanism of latered APP metabolism Results: The viability of VSMCs was increased after treatment with 1 mM and 10mM NaN₃(p<0.05) unitl 3 hr and then dimnished Many TUNEL positive cells were found in 10mM and 100mM treatment group. but were not apoptotic in nature 22C11 immunoreactivity was not changed at 3 hr, 6hr, 12hr Anti-oxidants reduced cellular proliferation (p<0.05). but did not block TUNEL positivities and did not influence the 22C11 immunoreactivity In a low concentration NaN₃ treatment group the viability of VSMCs was increased concentration dependently(p<0.05) Immunoblot with 22C11 showed the concentration dependent decrease at 145 kDa, 125 kDa. and high molecular weight range (>160kDa) TUNEL staining showed DNA fragmentations and condensations of nuclear chromatin suggesting apoptosis After treatment with anti-oxidants, the cellular proliferation was more decreased (p<0.05), and TUNEL positive cell deaths were blocked Immunoreactivities of 125 kDa (immature APP). 145 kDa (mature APP). and higher molecular weight bands were recovered below 400μM of NaN₃ Immunoreactivity of 145 kDa was recovered in 100 μM NaN₃ treated group Conclusions: The presumed mechanism of low concentration COX inhibitor is the overproduction of reactive oxygen species resulting from a depression of the mitochondiral electron transport chain. whereas potential consequence of high concentration COX inhibitor might be related to decreassion of ATP synthesis and bioenergetic impairment Reactive oxygen radicals in response to low concentration COX inhibitor alter the processing of APP in VSMCs This investigation demonstrated analtered APP metabolism as a peripheral marker of AD Therefore VSMCs treated with low concentration COX inhibitor could be concsidered as a novel in vitro model of AD.

      • Al₂O₃ TiO₂ 담체종류별 Pt, PtRu 촉매의 산화특성

        The volatile organic compounds (VOCs) have been recognized as major contributor to air pollution. The catalytic oxidation is one of the most important processes for VOCs destruction clue to getting high efficiency at low temperature. In this study, mono metallic Pt and bimetallic Pt-Ru were supported to TiO₂ and γ-Al₂O₃. In order to distribute metals uniformly, Xylene, toluene and MEK were used as reactants. The mono metallic or bimetallic catalysts were prepared by the excess wetness impregnation method and were characterized by XRD, XPS, TEM and BET analysis. As a result, Pt-Ru bimetallic catalysts were showed higher conversion than Pt mono metallic catalyst. Pt-Ru bimetallic catalyst was showed highest conversion on the γ-Al₂O₃support. In the VOCs oxidation, Pt-Ru bimetallic catalyst had multi point active sites, so it improved the range of Pt metal state. Therefore, bimetallic catalysts were higher conversion of VOCs than mono metallic ones.

      • Efeect of Necropsy Days on Number of Clonorchis sinensis and Plaque Forming Cells after Challenge in Hamsters

        Challenge 感染된 햄스터에서 屠殺日이 肝吸蟲과 PFC의 數에 미치는 影響을 決定하기 위해 肝吸蟲 成蟲의 代謝産物과 Freund's incomplete adjuvant로 感作된 golden 햄스터의 腹腔渗出細胞와 血淸을 recipient 햄스터의 腹腔內에 注入하였다. 第1次 感作한지 7日 後에 肝吸蟲 被囊幼蟲 20個씩 經口感染시킨 다음, 50日, 70日, 90日 그리고 110日에 各各 屠殺하였다. Challenge感染 後, EpG는 對照群에서 感作群보다 하루 일찍 나타났고, 對照群과 感作群 모두 漸次로 增加되다가 40日에서 65日사이에 最高値를 나타낸 후 漸次 減少하였다. 그러나, 感作群 사이에 EpG의 有意的 差는 認定할 수가 없었다. 蟲體回收率은 感作群과 對照群 사이에 有意的 差를 認定할 수 있었다. 蟲體回收率은 Challenge感染후, 약 70日 前後에 減少되었다. Plaque 形成細胞는 感作後 50日과 70日의 感作群에서 檢出되었으나, 그 以後의 感作群 및 對照群에서는 檢出되지 않았다. 以上의 成績으로 미루어 보아 golden 햄스터의 適切한 屠殺日은 Challenge感染 後 50日에서 70日이 適當함을 알 수 있었다. OBJECTIVE, MATERIALS AND METHODS: To determine the appropriate necropsy day in the passive transfer of immunity against Clonorchis sinensis in golden hamsters, the donor hamsters were sensitized with two injections of the admixture of the metabolic products of C. sinensis and Freund's incomplete adjuvant into footpads at 2 weeks' intervals. The peritoneal cells and sera of sensitized and control donor hamsters were injected intraperitoneally to the recipient hamsters, seven days after primary sensitiza-tion. Four groups of the recipient and control hamsters were challenged with 20 metacercariae of C. sinensis, and killed on the 50th, 70th, 90th and 110th day after challenge infection. RESULTS: The eggs of control hamsters appeared one day earlier than those of the recipient groups, and the eggs per gram of feces in both recipient and control groups increased stepwise and continued at high levels from the 40th to 65th day and then decreased irregularily after challenge infection. There was no significant difference in EpG of feces between the sensitized recipient and control groups, but there was significant difference in the worm burden of C. sinensis between the recipient and control groups. The recovery number of the fluke in the recipient and control hamsters decreased about the 70th day after challenge infection. CONCLUSIONS: These results indicate in part that the appropriate necropsy day of golden hamsters would be estimated around the 50th day after challenge infection of C. sinensis.

        중증 외상환자에서 염기 결핍수치의 유용성

        Background: This study's objective was to determine the prognostic value of the base deficit measured in the emergency department(ED) and to determine whether base deficit can provide information not provided by advanced injury scoring system. Methods: This study was a retrospective analysis of data collected for two years. Thirty-two severe trauma patients who were admitted to the Emergency Department of Korea University Hospital were included in this study. The patients were divided into two groups: the normal base deficit group(-3 mmol/L to 3 mmol/L) and the elevated base deficit group(>3 mmol/L). The base-deficit value, age, sex, head injury, organ failure rate, and survival were considered, and the also Revised Trauma Score(RTS), Acute Physiology And Chronic Health Evaluation(APACHE II), and Injury Severity Score(ISS) were measured. Result: The elevated base-deficit group showed a lower survival rate and a higher organ failure rate compared to the normal base deficit group. Logistic Regression showed a strong association between base deficit and mortality rate. Among the previous injury scoring Systems(RTS, APACHE II, ISS), base deficit had the strongest correlation with RTS. By using base deficit value and the RTS together, we obtained a higher positive predictive value than that obtained by using base deficit or RTS alone. Conclusion: The admission value of the base deficit in the ED is a useful tool in predicting the outcome in severe trauma patients, and it can be an adjunct to previous injury scoring systems. As an advanced injury scoring System is developed in the future, the base deficit may have some significant role.

      • 梁江 淡水魚에서 肝吸蟲 被襄幼蟲의 寄生狀

        1986年 6月부터 同年 9月까지 慶北 淸道郡과 慶南 密陽郡의 境界를 貫流하는 梁江에서 投網 및 사발모지等으로 14種의 淡水魚를 採集하여 肝吸蟲 被囊幼蟲의 寄生率과 寄生程度를 魚種別로 調査하였다. 6種의 淡水魚, 큰납지리, 칼납자루, 몰개, 모래주사, 납지리 및 돌고기에서 肝吸蟲 被囊幼蟲을 찾아볼 수 있었으며 그 寄生率은 돌고기 100%, 몰개 67.5%, 칼납자루 50.0%, 납지리 44.0%, 모래주사 37.5%, 큰납지리 11.1%의 順位였다. 寄生程度의 標議로서 魚肉 1g當 肝吸蟲 被囊幼蟲의 平均數는 몰개 48.3個로서 가장 많았고 큰납지리 0.3個로서 가장 적었으며 칼납자루, 모래주사, 납지리 및 돌고기는 1.4∼14.0個로서 中間置를 나타내었다. 梁江의 淡水魚에서의 肝吸蟲 被囊幼蟲의 寄生率과 寄生程度는 淸道川과 東倉川의 그 率과 程度의 中間置를 나타내었다. From June to September, 1986, fresh-water fish were collected in the river Yang running from the North-west parts of Chongdo county (Kun), Kyungpook Province, to Milyang county, Kyungnam Province, Korea. Fourteen species of fish were collected by throwing a cast net and using glass bowl containing crushed oil-cake as bait. They were examined for the presence and the intensity of infestation with the metacercaria of Clonorchis sinensis. Six species of fish were infested with the metacercaria of C. sinensis. The infestation rate for the metacercaria was highest in Pungtungia herzi 100%; followed by Gnathopogon atromaculatus 67.5%, Acheilognathus limbata 50.0%, Paracheilognathus rhombea 44.4%, Microphysogobio koreensis 37.5%, and Acanthorhodeus taenianalis 11.1%, in decreasing order. In the intensity of infestation with the cysts, G. atromaculatus was most heavily infested and the mean number of metacercaria per gram of flesh was 48.3, A. limbata, M. koreensis, P. rhombea and P. herzi were infested intermediately with from 1.4 to 14.0. Whereas, A. taenianalis was infested with the least number 0.3. It is indicated that the infestation rates and intensity of infestation with the metacercaria of C. sinensis from fresh-water fish caught in the river Yang were intermediate level compared with the results reported by Kang et al. (1984) in the river Chongdo and by Kim and Choi (1985) in the river Dongchang.

        Anti-osteoarthritis effects of Pomegranate, Eucommiae cortex and Achyranthis radix extracts on the primary cultured rat articular chondrocytes

        Objectives : The objective of present study is to evaluate anti-arthritic effects of dried pomegranate concentrate powders (PCP), Eucommiae Cortex aqueous (EC) and ethanolic (ECe) extracts, Achyranthis Radix aqueous (AR) and ethanolic (ARe) extracts on the primary cultured rat articular chondrocytes. Methods : MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium Bromide) assay was performed cytotoxic effect of test substances. In addition, anti-inflammatory effects were also observed on the lipopolysaccaride (LPS) treated chondrocytes through prostaglandin $E_2\;(PGE_2)$ production and 5-lipoxygenase (LPO) activities, and inhibitory effects on metalloproteinase (MMP)-2 and MMP-9 activities were observed on the recombinant human interleukin $(rhIL)-1{\alpha}$ treated chondrocytes with their extracellular matrix (ECM) related mRNA expressions - collagen type II, SOX9 and aggrecan. Results : As results, ECe and ARe showed obvious cytotoxicity against primary cultured rat articular chondrocytes at a dose level of 10 mg/ml, respectively. However, no obvious cytotoxic effects of PCP, EC and AR were demonstrated at a dose level of 10 mg/ml, on the primary cultured rat articular chondrocytes. In addition, treatment of LPS $50{\mu}g/ml$ induced significant increases of $PGE_2$ contents and 5-LPO activities indicating inflammatory responses of the primary cultured rat articular chondrocytes, and also decreases of cell viabilities, increases of MMP-2 and MMP-9 activities with decreases of extracellular matrix (ECM) related collagen type II, SOX9 and aggrecan mRNA expressions were observed by treatment of $rhIL-1{\alpha}$ 50 ng/ml, suggesting damages on the primary cultured rat articular chondrocytes and related ECM degradations. However, these inflammatory responses and related ECM degradations were inhibited by pretreatment of all test substances, in order of PCP > ECe > ARe > EC > AR, and $rhIL-1{\alpha}$ induced chondrocytes deaths are inhibited by treatment in order of PCP > EC > AR > ECe > ARe. Conclusions : Taken together, it is expected that mixed formulation of PCP as main components with appropriate proportion of EC and AR as additional components will be achieved a potent alternative medicinal food for osteoarthritis.

        High tibial osteotomy (HTO) is an effective surgical technique that reduces pain and delays medial cartilage damage as a part of varus malalignment treatment for young, active patients. Lateral closed wedge HTO has been a popular choice, but with advances of surgical technique and fixation, medial open wedge HTO has become a popular choice of surgical treatment for most knee varus deformity. In the present review, we present the major issues to be considered during rehabilitation after medial open wedge HTO and propose a phased rehabilitation program.

        Impact of White Matter Changes on Activities of Daily Living in Mild to Moderate Dementia

        <P>The association between white matter changes and activities of daily living (ADL) in a large, well-defined cohort of patients with mild-to-moderate dementia (either Alzheimer’s disease or subcortical vascular dementia) were investigated. A total of 289 patients were divided into three groups (140 mild, 99 moderate, and 50 severe) depending on the degree of white matter changes as indicated on brain magnetic resonance image scans. Further, we analyzed the three groups’ performances on basic and instrumental ADL. The degree of white matter changes was associated with greater age, hypertension, previous history of stroke, higher Hachinski Ischemic Score, worse global cognitive and functional status, and an increased impairment of basic ADL and instrumental ADL. The increased impairment with regard to the severe group’s performance on both the basic and instrumental ADL remained significant after adjustment for age and hypertension. Tasks involving physical activities were most significant. This was the first study investigating the association between white matter changes and ADL in a large, well-defined dementia cohort. The present study suggests that severe white matter changes may be associated with higher impairment on both basic and instrumental ADL.</P><P>Copyright © 2010 S. Karger AG, Basel</P>
