Interleukin-21 is a common γ-chain cytokine that controls the immune responses of B cells, T cells, and natural killer cells. Targeting IL-21 to strengthen the immune system is promising for the development of vaccines as well as anti-infection and anti-tumor therapies. However, the practical application of IL-21 is limited by the high production cost. In this study, we improved IL-21 production by codon optimization and selection of appropriate signal peptide in CHO-K1 cells. Codon-optimized or non-optimized human IL-21 was stably transfected into CHO-K1 cells. IL-21 expression was 10-fold higher for codon-optimized than non-optimized IL-21. We fused five different signal peptides to codon-optimized mature IL-21 and evaluated their effect on IL-21 production. The best result (a 3-fold increase) was obtained using a signal peptide derived from human azurocidin. Furthermore, codon-optimized IL-21 containing the azurocidin signal peptide promoted IFN-γ secretion and STAT3 phosphorylation in NK-92 cells similar to codon-optimized IL-21 containing original signal peptide. Collectively, these results indicate that codon optimization and azurocidin signal peptides provide an efficient approach for the high-level production of IL-21 as a biopharmaceutical.

Background : In recent years, gonorrhea has been panedemic and remains one of the most commom STDs in the world, especially in developing countries. Objective & Methods: For the detection of a more effective therapeutic regimen and assessing the prevalence of PPNG, we have been trying to study the patients who have visited the VD Clinic of Choong-Ku Public Health Center in Seoul since 1980 by means of the chromogenic cephalosporin method. Results: In 1996, 139 strains of N. gonorrhoeae were isolated, among which 53(39.0%) were PPNG. Conclusion: Our results suggests that after a peak of 74.3% in 1993, the prevalence of PPNG in Seoul is gradually declining.

In recent years, gonorrhea has been pandemic and remains one of the most common STDs in the world, especially in developing countries. For the detection of a more effective therapeutic regimen and assessing the prevalence of Penicillinase Producing Neisseria gonorrhoeae(PPNG), we have been trying to study the patients who have visited the Venereal Disease Clinic of Choong-Ku Public Health Center in Seoul since 1980 by menas of the chromogenic cephalosporin method. In 1998, 93 strians of N. genorrhoeae were isolated, among which 60(64.5%) were PPNG. The prevalence of PPNG in Seoul, which had been decreased to 39% in 1996 after a peak of 74.3% in 1993, is increased to 64.5% in 1998.

In this study, caissons for accelerating consolidation with many filtering-drains on their back sides, for the purpose of inducing lateral dissipation of excess pore water pressure caused by new backfill, have been proposed. And to analyze the effect of accelerating consolidation by using proposed caissons, the analytical solution for two-dimensional consolidation equation is derived. In an isotropic clay, this study shows consolidation times at ξ = 0.1 to reach the average degrees of consolidation of 50% and 90% when using caissons with drains are taken about 7 and 3.5 times less than those when using the conventional caissons, respectively.

The feasibility of caissons for accelerating consolidation (C.A.C.) constructed in marine clays with anisotropic permeability has been presented in this paper. By considering anisotropic permeability, this study provides analytical solutions for excessive pore water pressures of two-dimensional consolidation equation and average degrees of consolidation with varying time factors. With varying anisotropic permeability ratios of 5, 10, and 50, consolidation times required for reaching the average degree of consolidation of 90% for C.A.C. at the section of ξ= 0.1 are shorter than those for conventional caissons by 8, 13, and 47 times, respectively. C.A.C. is more effective in reducing consolidation time as anisotropic permeability ratio increases. Also plots for average degrees of consolidation versus the width to depth ratios of analyzed sections and permeability ratios, which will enable engineers to determine easily average consolidation settlements, are given in this study.

The subchronic toxicity study of rHuEPO, a newly developed recombinant erythropoietin, was investigated for 13 weeks in Beagle dogs intravenously treated with doses of 100,500 and 2,500 IU/㎏/day. There were no significant changes in body weight, food intake, physical and opthalmic examination, urine analysis, etc. Any toxic response was not observed except for enlarged spleen and extramedullary hematopoiesis. These results indicate that the no-observed adverse effect level (NOAEL) of rHuEPO is 100 IU/㎏ in Beagle dogs.

Background: Pulmonary tuberculosis is a great public health problem in Korea. Early detection of Mycobacterium tuberculosis (M. tuberculosis) in clinical samples becomes more and more important in the control of tuberculosis. Recently, molecular methods have been applied to early detection of M. tuberculosis. Methods: To elucidate the effectiveness of nested polymerase chain reaction (PCR) in early diagnosis of pulmonary tuberculosis, we peformed Acid-fast bacilli (AFB) stain, AFB culture, and nested PCR on 84 sputa. Results: 8 (9.5%) specimens were positive by AFB stain, 17 (20.2%) by AFB culture, and 15 (17.9%) by nested PCR. Using AFB culture as standard method, the sensitivity of AFB stain and nested PCR was 47.1% and 82.4%, respectively. The specificity of AFB stain and nested PCR was 100% and 98.5%, respectively. Conclusions: Nested PCR was more sensitive than AFB stain and had shorter processing time(24-48hrs) than AFB culture. So it may be effective to use nested PCR in order to detect M. tuberculosis when AFB smear Is negative.
단열양생을 이용한 고강도 콘크리트 압축강도 추정에 관한 실험적 연구

The present study is a basic experiment on the estimation of the compressive strength of high strength concrete, aiming at estimating the compression strength of mass test pieces of high strength concrete by giving the temperature hysteresis of the mass test pieces to managerial test pieces. Different from ordinary concrete, high strength concrete generally shows the high rise of temperature caused by hydration heat inside the concrete. It is known that, in mass concrete, thermal stress occurs due to the difference in temperature between the inside and the outside, which causes a significant difference in compression strength between structure beams and managerial test pieces. It is also reported that there is a large difference between the compression strength of cylindrical managerial test pieces of standard underwater curing and the strength of structure beam concrete. Thus, this study made concrete test pieces in an optimal mix ratio for each strength level, and also created adiabatic curing tank and managerial test pieces. Then it carried out comparative analysis in relation to core strength and suggested equipment and a technique that can control the strength of high strength concrete mass more conveniently and accurately.

To develop new pharmacological charactization of isoquinoline derivatives, we have synthesized 3-benzyl-9,10-dimethoxy-6,7,11b,12-tetrahydropyridazino [4',5': 5,6] [1,4] -oxazino [3,4-а]isoqinolin-4(3H) -one. We have developed a convenient cyclization of novel isoquinoline derivatives starting from 3,4-dimethoxy phenethylamine using boron trifluoride diethyl etherate.
