Alterations affecting the status of robustness and health can bring about physiological changes including hematological parameters in pigs. To identify quantitative trait loci (QTL) associated with 8 hematological phenotypes (one leukocyte trait, six erythrocyte traits, and one platelet trait), we performed a genome-wide association study using the Porcine SNP 60K BeadChip in an intercross population between Landrace and Korean native pigs. A total of 36,740 SNPs from 816 F2 offspring were analysed for each blood related traits after filtering by quality control. Data were analysed genome-wide rapid association using the mixed model and regression (GRAMMAR) approach. A total of 257 significant SNPs (P<1.36x10-6) on SSC3, 6, 8, 13, and 17 were detected for blood related traits in this study. Interestingly, the genomic region between 17.9 and 130 Mb on SSC8 was found to be significantly associated with RBC, MCV, and MCH. Our results include 5 significant SNPs within five candidate genes (KIT, IL15, TXK, ARAP2, and ERG) for hematopoiesis. Further validation of these identified SNPs could give valuable information for understanding the variation of hematological traits in swine.

Actinomycosis is a chronic, progressive, Suppurative, granulomatous disease. Definite diagnosis will generally be based upon histologic identification of actinomycotic granules or culture of the Actinomyces, or both. The diagnosis of abdomino-pelvic actinomycosis is often difficult to make. Since abdominal actinomycosis is indeed an uncommonly encountered clinical entity today, many clinicians fail to consider actinomycosis as a possibility. Actinomyces is anaerobic. It is not usually isolated from routine cultures and anaerobic cultures are difficult to obtain. It may be impossible to clinically distinguish abdominal actinomycosis from other diseases because of the variable forms of presentation which may be recognized in patients, including fever, anorexia, nausea and vomiting. Especially in Korea, the clinical presentation of actinomycosis often resembles intestinal tuberculosis, which should not usually be treated surgically. We experienced a case of Abdomino-pelvic Actinomycosis in which it was difficult to make a early diagnosis.

In this study, the masking effect of olfactory stimulus on the awakening state due to sound stimuli while driving using Graphic Driving Simulator was observed through the response of autonomic nervous system. The test was conducted for 11 males in their twenties. The siren of ambulance car was presented to them as auditory stimulus for 30 secs while driving in a situation of high way in the condition of both peppermint and control, respectively, and LF/HF ratio of HRV (Heart Rate Variability), the activity index of sympathetic nerve, and GSR (Galvanic Skin Response) response were examined. The test was proceeded in the order of three stages, that is, sound stimuli (test 1), driving performance, and sound stimuli (test 2), and fragrance stimulus, driving performance, and sound stimuli (test 3), and the physiological signal of GSR, HRV was measured in the whole stages. As a result of test, comparing the results of before and after auditory stimulus test (1) (p < 0.01), test (2) (p < 0.05), and test (3) (p < 0.01), driving performance test (2) (p < 0.01), test (3) (p < 0.01), and olfactory stimulus test (3) (p < 0.05), respectively, GSR response increased, showing significant difference in all the tests. It indicates that when auditory stimulus was presented to the subjects, they were in the awakening state as sympathetic nervous system got activated. As a result of comparing auditory stimulus while driving before and after presenting olfactory stimulus, there was no significant difference in GSR response. The LF/HF ratio of HRV increased, showing a significant difference only in test (2) (p < 0.05), and in driving performance test (2) (p < 0.05) in auditory stimulus, however, it showed no significant difference in olfactory stimulus. As a result of comparing auditory stimulus while driving before and after presenting olfactory stimulus, there was a decrease, showing significant difference (p < 0.05) in LF/HF ratio of HRV. That is, it means that the activation of sympathetic nervous system decreased, and that parasympathetic nervous system got activated. From these results, it was observed that while driving, the awakening level due to auditory stimulus was settled with olfactory stimulus. In conclusion, it was drawn that while driving, olfactory stimulus could have the masking effect on auditory stimulus.

Previously, we reported a quantitative trait locus (QTL) that affect total teat number (TTN) on pig chromosome 7 (SSC7) in a large F2 intercross population between Landrace and Korean native pigs. The aim of this study was to refine the QTL associated with TTN and to identify positional candidate gene(s) within the refined genomic region. TTN was recorded in 1,105 F2 progeny. All experimental animals were genotyped using 998 informative single nucleotide polymorphism (SNP) markers located on SSC7. A haplotype-based linkage and association analysis using the PHASEBOOK programme was applied to perform high-resolution QTL analysis. Additionally, linear mixed-effect models were used to assess the effect of a positional candidate gene on TTN and other economically important traits [i.e., thoracic vertebrae number (THO), carcass body length (CBL) and weight (CW), back fat thickness (BFT) and intramuscular fat content (IMF) in loin muscles]. Joint linkage and association analysis refined the critical region to a 1.07 Mb region that included a novel positional candidate gene, BRMS1L, that encodes the breast cancer metastasis-suppressor 1-like protein, which could possibly be implicated in normal mammary gland development. Significant association of an SNP marker (g.-1087 G>A) in the 5’-flanking region of BRMS1L with TTN (P=1.10x10-8), THO (P=5.80x10-4), and CBL (P=0.038) was observed. Based on these data, we propose BRMS1L as a positional candidate gene for TTN in pigs. After validation of the association in other independent populations and further functional studies, these results could be useful in optimizing breeding programmes that improve TTN and other economically important traits in swine
Phylogenetic analysis of the maternal lineages presented in extant Jeju native pigs

An intensive analysis was conducted of the maternal lineages of the Jeju native pigs (JNPs). A total of 100 mtDNA sequences from Asian wild boars (AWB), European wild boars (EWB), Asian domestic pigs (ADP), European domestic pigs (EDP), and JNPs were used for the phylogeny and network analyses. Two distinct JNP groups were found JNPA and JNPE in the Asian and European cluster. The maternal lineage of JNPE was the closest to that of EWB and a clear haplogroup sharing an identical haplotype (hap16) among 15 individuals of JNPE and 2 individuals of EWB was identified. However, except for hap18, no EDP shared any identical haplotypes with JNPE, suggesting that no obvious maternal contribution of EDP has occurred in JNPE in recent years. The possible existence of an additional and unknown path of maternal lineage from EDP into JNPE could therefore be postulated, in addition to those from AWB and ADP into the JNPA groups. Thus, JNPE appeared to have a pure maternal lineage that had no recent contact with EDP, and both the JNPA groups and JNPE are pure Jeju native pigs.

Allergic diseases such as asthma, allergic rhinitis, and atopic dermatitis are heterogeneous diseases characterized by multiple symptoms and phenotypes. Recent advancements in genetic study enabled us to identify disease associated genetic factors. Numerous genome-wide association studies (GWAS) have revealed multiple associated loci for allergic diseases. However, the majority of previous studies have been conducted in populations of European ancestry. Moreover, the associations of single nucleotide polymorphisms (SNPs) with allergic diseases have not been studied amongst the large-scale general Korean population. Herein, we performed the replication study to validate the previous variants, known to be associated with allergic diseases, in the Korean population. In this study, we categorized three allergic related phenotypes, one allergy and two asthma related phenotypes, based on self-reports of physician diagnosis and their symptoms from 8,842 samples. As a result, we found nominally significant associations of 6 SNPs with at least one allergic related phenotype in the Korean population. [BMB reports 2012; 45(5): 305-310]

본 연구에서는 지그비 기반의 컴퓨팅 기술이 혼합된, 난방 보일러 제어시스템을 제안한다. 지그비 RF 기술과 임베디드 하드웨어 기술의 조합을 통하여, 주택 및 아파트 등에서 각 방별로 온도 및 습도를 취합하여 난방을 관리할 수 있다. 또한 제안된 시스템은 체감온도 기반으로 난방을 관리하여 인간에게 쾌적한 환경을 제공하기 위한 최적의 선택이라 할 수 있다.

Lightning currents are one of major influences on the dynamic performance of counterpoise. High lightning current causes the dynamic grounding performance of counterpoise due to very fast fronted pulses. The previous analysis has often been based on quasi-static approximation that can not be applicable to very fast fronted pulses. To extend the analysis for fast fronted pulses in this paper, the full-wave analysis method based on the rigorous electromagnetic-field theory approach is used and the effects of the ionization of the soil are disregarded. Based on the simulation results, the empirical formulas applicable for slow and fast fronted lightning current pulses are reviewed; therefore, the validity of the theoretical approach is verified through comparison between the calculated and measured results.
Phaeodactylum tricornutum의 (E)-4-Hydroxy-3-methylbut-2-enyl Diphosphate Reductase 유전자의 형질전환

Phaeodactylum tricornutum is a model diatom that its genomic information and biological tools are well established. In this study, a gene encoding (E)-4-hydroxy-3-methylbut-2-enyl diphosphate reductase (PtHDR), a terminal enzyme of the methylerythritol phosphate pathway regulating chlorophyll and carotenoid biosynthesis, was isolated from P. tricornutum. The isolated gene was cloned into pPha-T1 vector containing fcpA promoter to prepare pPha-T1-HDR plasmid. As a positive control, pPha-T1-eGFP plasmid was constructed with egfp gene. Stable nuclear transformation was carried out with these plasmids by particle bombardment method and zeocin resistant colonies of P. tricornutum were selected on f/2 agar plate. In result, transformation efficiency was evaluated according to the amount of plasmid DNA coated with gold particles. Integration of introduced plasmids was confirmed with genomic DNA of each transformant by polymerase chain reaction. The eGFP fluorescence was visible in the cytoplasm, indicating that eGFP was successively expressed in P. tricornutum system. The transcript level of exogenous Pthdr gene was evaluated with the obtained transformants. The results presented here demonstrated that introduction of Pthdr gene into P. tricornutum chromosome succeeded and expression of PtHDR was enhanced under the fcpA promoter.
