        Crystal Structure Control of ZnxCd1-xS Alloyed Nanocrystals and Structural Dependent Fluorescence Properties

        Crystal structure control is so important to the fluorescence properties that each element should be considered carefully. In conventional synthesis of ZnxCd1-xS alloyed nanocrystals (NCs), most of studies focus on ligand–surface interaction on the formation of either zinc blende or wurtzite ZnxCd1-xS nanocrystals, instead of the reactant source. In this work, mixed crystal phase was found easily in ZnxCd1-xS alloyed NCs when reaction proceeded at high Zn/Cd source ratio condition. Therefore, we regulate the Zn/Cd ratio to obtain relative pure zinc-blende structure, and study the influence of structure change on the fluorescence properties. Further, we have proposed a two-step ZnS coating method to acquire ZnxCd1-xS/ZnS NCs with separated crystal-phase between core and shell. Compared with maximum QY of 81% for ZnxCd1-xS/ZnS NCs synthesized by conventional one-step coating method, the performance of the optimized NCs has significantly improved with maximum QY of 93%.

        Cloning and Sequence Analysis of Wild Argali ISG15 cDNA

        The complete coding sequence of Wild Argali ISG15 cDNA was generated by rapid amplification of cDNA ends. The ISG15 cDNA was 642 bp with an open reading frame of 474 bp, which encoded a 17.47 kDa protein composed of 157 amino acids. Its amino acid sequence shared 97.9%, 80.8%, 91.4%, 94.3%, 78.3% identity with those of ISG15cDNA from Ovis aries (accession no. NM001009735.1), Capra hircus (accession no. HQ329186.1), Bos taurus (accession no. BC102318.1), Bubalus bubalis (accession no. HM543269.1), and Sus scrofa (accession no. EU647216.1), respectively. The entire coding sequence was inserted into the pET-28a vector and expressed in E. coli. The recombinant protein corresponded to the expected molecular mass of 25 kDa as judged by SDS-PAGE, and it was detected in the bacterial inclusion bodies. The expressed protein could be purified by $Ni^{2+}$ chelate affinity chromatography and the results from the lymphocyte proliferation test showed that the product could stimulate lymphocyte proliferation very well (p<0.05), which further confirmed its biological activity.

        Cloning and sequence analysis of Wild Argali short palate, lung and nasal epithelium clone 1 cDNA

        Objective: Experiments were conducted to clone the sequence of Wild Argali short palate, lung and nasal epithelium clone 1 (SPLUNC1) cDNA, and to lay the foundation for further study the biological function of Wild Argali SPLUNC1. Methods: The complete sequence of Wild Argali SPLUNC1 cDNA was generated by rapid amplification of cDNA ends. The entire coding sequence was inserted into the pPIC9K vector and expressed in Pichia pastoris (P. pastoris) GS115. The recombinant SPLUNC1 protein was detected by Western blot and purified by Ni2+ chelate affinity chromatography. The test of effect of the protein on Mycoplasma ovipneumoniae (MO) was performed with real-time polymerase chain reaction. Results: The Wild Argali SPLUNC1 cDNA was 1,076 bp with an open reading frame of 768 bp, which encoded a 26.49 kDa protein composed of 255 amino acids. Its amino acid sequence shared 98.4%, 96.9%, 94.5%, 90.2%, 80.8%, 78.4%, 78.3%, 72.5%, 72.3%, 68.8% identity with those of SPLUNC1 cDNA from Ovis aries (accession no. NP_001288334.1), Capra hircus (accession no. XP_005688516.1), Pantholops hodgsonii (accession no. XP_005979709.1), Bos taurus (accession no. NP_776851.1), Felis catus (accession no. XP_006929910.1), Homo sapiens (accession no. NP_001230122.1), Sus scrofa (accession no. NP_001005727.1), Chinchilla lanigera (accession no. NP_001269294.1), Mus musculus (accession no. NP_035256.2), and Rattus norvegicus (accession no. NP_742028.1), respectively. The recombinant protein corresponded to the expected molecular mass of 25.47 kDa as judged by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and it was detected in the supernatant of P. pastoris, and it could be purified. The results from the test of inhibition effect of argali recombinant SPLUNC1 protein on MO showed that the product could inhibit MO very well (p<0.01). Conclusion: The amino acid sequence of Wild Argali SPLUNC1 was different from other organisms. The recombinant SPLUNC1 protein has good biological activity.

        Motives of cheating among secondary students: the role of self-efficacy and peer influence

        A survey research study was conducted with a sample of 100 secondary students from a local secondary school about the motives of cheating. The primary focus of this study was the interplay among variables of self-efficacy, peer influence and cheating. The results showed that students with low self-efficacy were more likely to cheat than those who perceived themselves as efficacious. It was further found that peers played a significant role in discouraging cheating by expressing disapproval and informing teachers of dishonest behaviour.

        Cyclic AMP Responsive Element Binding Protein 3-like 4/AarF Domain Containing Kinase 5 Axis Facilitates Proliferation, Migration and Invasion of Lung Adenocarcinoma Cells by Modulating the TGFβ Pathway

        Cyclic AMP responsive element binding protein 3-like 4 (CREB3L4) has been reported as a transcription factor showing high expression in various cancers, whereas the mechanism of CREB3L4 in lung adenocarcinoma (LUAD) progression remains unclear yet. Expression levels of CREB3L4 and its downstream target gene AarF Domain Containing Kinase 5 (ADCK5) in LUAD tissues were analyzed by bioinformatics methods and were detected by real-time quantitative polymerase chain reaction at cellular level. The signaling pathways in which the downregulated ADCK5 enriched were analyzed via Gene Set Enrichment Analysis. The regulatory relationship between CREB3L4 and ADCK5 was identified by ChIP and dual luciferase reporter assay. CCK-8 and colony formation assays were utilized to evaluate LUAD cell proliferation. Transwell was utilized to measure migration and invasion of LUAD cells. Western blot was employed to check expression changes of CREB3L4, ADCK5, TGFβ pathway proteins and Epithelial- Mesenchymal Transition related proteins. Compared with paracancer tissues, CREB3L4 and ADCK5 were highly expressed in LUAD tissues, while silencing CREB3L4 could dramatically hinder invasion, proliferation and migration of LUAD cells. ADCK5 was the downstream target gene of CREB3L4, and CREB3L4 promoted the transcription of ADCK5. ADCK5 was markedly enriched in the TGFβ pathway, which stimulated the malignant development of LUAD cells by activating this pathway. In addition, the rescue assay verified that CREB3L4 regulated the TGFβ pathway by activating ADCK5, thereby accelerating LUAD cell malignant phenotypes. This study revealed the mechanism of CREB3L4/ADCK5 axis facilitating malignant phenotypes of LUAD cells, and bred new insights into the LUAD treatment.

        Ectopic expression of grape hyacinth R3 MYB repressor MaMYBx affects anthocyanin and proanthocyanidin biosynthesis and epidermal cell differentiation in Arabidopsis

        Single-repeat R3 MYB transcription factors regulate multiple developmental and metabolic pathways in plants. The anthocyanin-related R3 MYB repressor MaMYBx was previously identified in grape hyacinth (Muscari spp.), and its ectopic expression reduced the accumulation of anthocyanin in tobacco petals. However, whether the function of MaMYBx is conserved across plant species remains unclear. Here, we report that ectopic expression of MaMYBx regulates anthocyanin and proanthocyanidin (PA) biosynthesis and epidermal cell differentiation in Arabidopsis. MaMYBx overexpression in Arabidopsis decreased the accumulation of anthocyanin and PA by downregulating the late structural genes (AtDFR, AtANS, AtANR, and AtGST), some key regulatory factors of MBW complexes (AtPAP1, AtTT8, and AtGL3) and an R3 MYB gene (AtCPC) involved in flavonoid biosynthesis. In addition, MaMYBx also modulated trichome formation and root hair differentiation by downregulating MBW component genes (AtGL1, AtWER, and AtGL3) and their target gene AtGL2 and by upregulating several R3 MYB genes (AtTRY and AtTCL1). We further found that MaMYBx transgenic plants accumulated less anthocyanin than did wild-type plants under salt, osmotic pressure, cold, and nitrogen stresses. Moreover, the effect was even greater, and significantly less anthocyanin accumulated. In conclusion, these results implied functional conservation of MaMYBx in anthocyanin and PA biosynthesis pathways of Arabidopsis. Therefore, MaMYBx likely represents a new potential gene for the molecular breeding of horticulture and ornamental plants via the regulation of anthocyanin and PA.

        Low-Temperature Synthesis of Highly Efficient, Deep-Red Zn-Cu-In-Se/ZnSe Fluorescence Quantum Dots

        We report a facile synthesis method on CuInSe2 (CISe)-based quantum dots (QDs) by using tri-n-octylphosphine selenium (TOPSe) as selenide precursor, with assistance of oleylamine (OAm) and n-dodecanethiol (DDT). We demonstrate that the OAm and DDT jointly contribute to the formation of the low-temperature-decomposable metal-sulfide clusters, and promote the QD nucleation at relatively low temperature range of 180–200 ℃. Furthermore, to improve fluorescence property, Zn-doping and ZnSe coating are simultaneously carried out. The obtained deep-red ZnCISe/ZnSe QDs possess higher quantum yield of 65% at wavelength of 670 nm, which is in the best performance range ever reported. Then, we investigate the improvement mechanism, where the sufficient Zn replacement of In sites is the crucial factor. This modified core–shell structure provides two benefits, on the one hand, the enhancement on intrinsic defect-related recombination, and the other hand, the improved core–shell interface that reduces the nonradiative recombination.

        Combination of High-Density Lipoprotein Cholesterol and Lipoprotein(a) as a Predictor of Collateral Circulation in Patients With Severe Unilateral Internal Carotid Artery Stenosis or Occlusion

        Background and Purpose Collateral circulation is considered an important factor affecting the risk of stroke, but the factors that affect collateral circulation remain unclear. This study was performed to identify the factors associated with collateral circulation, especially blood lipids. Methods The study involved patients who had undergone digital subtraction angiography and were confirmed as having severe unilateral stenosis or occlusion of the internal carotid artery (ICA). We classified the collateral circulation status of each patient as good (Grade 3 or 4) or poor (Grade 0, 1, or 2) according to the grading system of the American Society of Interventional and Therapeutic Neuroradiology/American Society of Interventional Radiology. We collected data on patients’ characteristics and identified the factors that affect collateral circulation. Results This study included 212 patients. The multivariate logistic regression analysis showed that the high-density lipoprotein cholesterol (HDL-C) concentration and a complete anterior half of the circle of Willis were independent protective factors for good collateral circulation, whereas elevated lipoprotein(a) [Lp(a)] and serum creatinine concentrations were independent risk factors for good collateral circulation. The area under the receiver operating characteristics curve (AUC) was 0.68 (95% confidence interval [CI], 0.61–0.76) for HDL-C and 0.69 (95% CI, 0.62–0.76) for Lp(a). A binary logistic regression model analysis of the joint factor of HDL-C and Lp(a) yielded an AUC of 0.77 (95% CI, 0.71–0.84). Conclusions In patients with severe unilateral ICA stenosis or occlusion, the combination of HDL-C and Lp(a) is a useful predictor of collateral circulation.

        Facile Preparation of Novel Hierarchically Porous Carbon at the Molecular Level for Supercapacitor Electrode Application

        Hierarchically porous carbons are obtained by a facile process of carbonizing the precursor of diblock copolymer xPS-b-polymethylmethacrylate (PMMA). In the diblock copolymer, there is a carbon source of cross-linked polystyrene (xPS) and a sacrificial block of PMMA. The atom transfer radical polymerization was used to synthesize the diblock copolymer with controlled molecular weight and narrow polydispersity in dimethylformamide at 120 ℃. The obtained hierarchically porous carbons (HPCs) exhibit remarkable electrochemical properties, such as higher specific capacitance of 204 Fg -1 at the current density of 0.5Ag -1 in 6 mol L -1 aqueous KOH electrolyte. In addition, it also presents an excellent rate performance of 64% capacitance retention as the current density increases to 10Ag -1 and a superior cycling stability of 98% capacitance retention after 5000 cycles at the current density of 1Ag -1.
