        Bone Marrow T Cells are Superior to Splenic T Cells to Induce Chimeric Conversion After Non-Myeloablative Bone Marrow Transplantation

        Background/Aims: The bone marrow functions not only as the primary B-lymphocyte-producing organ but also as a secondary lymphoid organ for CD4 and CD8 cell responses and a site of preferential homing and persistence for memory T cells. Bone marrow T (BM-T) cells are distinguished from peripheral blood T cells by surface phenotype, cytokine secretion profile, and immune functions. In this study, we evaluated the alloreactive potential of donor lymphocyte infusion (DLI) using BM-T cells in mixed chimerism compared to that using spleen T (SP-T) cells. Methods: Cells were prepared using established procedures. BM-T cells were obtained as a by-product of T-cell depletion in BM grafting and then cryopreserved for subsequent DLI. We performed DLI using BM-T cells in allogeneic mixed chimera mice on post-BMT day 21. Results: When the same dose of T cells, 5-10×105 (Thy1.2+), fractionated from BM and spleen were administered into mixed chimeras, the BM-T group showed complete chimeric conversion, with self-limited graftversus-host disease (GVHD) and no pathological changes. However, the SP-T group showed persistent mixed chimerism, with pathological signs of GVHD in the liver and intestine. Conclusions: Our results suggest that DLI using BM-T cells, even in small numbers, is more potent at inducing chimeric conversion in mixed chimerism than DLI using SP-T cells. Further study is needed to determine whether cryopreserved BM-T cells are an effective cell source for DLI to consolidate donor-dominant chimerism in clinical practice without concerns about GVHD.         Protective effect of chorioamnionitis on the development of bronchopulmonary dysplasia triggered by postnatal systemic inflammation in neonatal rats

        <P><B>Background:</B></P><P>Prenatal or postnatal systemic inflammation can contribute to the development of bronchopulmonary dysplasia (BPD). We investigated whether prenatal intra-amniotic (i.a.) inflammation or early postnatal systemic inflammation can induce BPD in a rat model.</P><P><B>Methods:</B></P><P>One microgram of lipopolysaccharide (LPS) or vehicle was injected into the amniotic sacs 2 d before delivery (E20). After birth, 0.25 mg/kg of LPS or vehicle was injected into the peritoneum of pups on postnatal day (P)1, P3, and P5. On P7 and P14, peripheral blood (PB), bronchoalveolar lavage fluid (BALF), and lung tissue were obtained and analyzed.</P><P><B>Results:</B></P><P>Postnatal i.p. injections of LPS significantly increased neutrophil counts in PB and BALF on P7 and P14. Similarly, proinflammatory cytokine and angiogenic factor transcript levels were increased in the lung by i.p. LPS on P7. Alveolar and pulmonary vascular development was markedly disrupted by i.p. LPS on P14. However, pretreatment with i.a. LPS significantly negated the detrimental effects of postnatal i.p. LPS on PB and BALF neutrophil counts and on lung proinflammatory cytokine expression and histopathological changes.</P><P><B>Conclusion:</B></P><P>Exposure to early postnatal systemic LPS induces BPD, an arrest in alveolarization, in neonatal rats. Preceding exposure to i.a. LPS protects the lungs against BPD triggered by postnatal systemic inflammation.</P>

        신장 ; 혈액투석에서 투석액 칼슘농도와 칼시트리올이 골대사에 미치는 영향

        목적: 현재 혈액투석에 사용 중인 투석액 칼슘 농도는 1.25mmol/L, 1.5 mmol/L, 1.75 mmol/L로 구분된다. 고칼슘 투석액은 부갑상선호르몬의 분비를 억제시키는 효과가 있으나 고칼슘혈증 위험을 증가시키고, 저칼슘 투석액은 adynamic bone disease을 개선시키는 효과가 있으나 칼슘 평형을 악화시킬 우려가 있으므로 투석액 칼슘 농도의 적절한 선택은 쉽지 않다. 저자들은 표준칼슘 농도인 1.5 mmol/L 투석액을 사용하였을 때 칼시트리올 용량 변화와 아울러 요독성 골질환에 미치는 효과를 평가하고자 하였다. 방법: 투석액 칼슘 농도 1.75 mmol/L로 1년 이상 혈액투석 중인 환자 36명에서 투석액 칼슘 농도를 1.5 mmol/L로 전환 후 1년간 경과를 추적하였다. 투석액 칼슘 농도를 낮추기3개월 전에 측정한 혈청 iPTH 농도에 따라 환자를 1군(iPTH< 150 pg/mL, n=21), 2군(iPTH 150-300 pg/mL, n=7), 3군(iPTH>300 pg/mL, n=8)으로 구분하여 혈청 칼슘, 인, 알칼리포스파타제, iPTH 농도를 3개월 간격으로 측정하였고, 인결합제 및 칼시트리올 용량 변화를 조사하였다. 결과: 투석액 칼슘 농도 1.75 mmol/L 사용 중 1군, 2군 및 3군의 iPTH 농도는 각각 57±48 pg/mL, 191±46 pg/mL, 589±200 pg/mL로 유의한 차이가 있었다(p<0.001). 투석액칼슘 농도를 1.5 mmol/L로 전환 후 1군에서 혈청 iPTH가 12개월에 걸쳐 유의하게 증가하였다(p=0.01). 3개월째부터 증가가 현저하여(57±48 vs. 287±266 pg/mL, p<0.01) 이후증가세를 유지하였다. 그러나 2군에서는 유의한 변화가 없었고, 3군의 혈청 iPTH는 12개월에 걸쳐 유의하게 감소하였는데(p=0.02) 특히 9개월째 감소가 기저치에 비해 낮았다(589±200 vs. 242±246 pg/mL, p<0.01). 혈청 알칼리포스파타제도 혈청 iPTH와 유사한 변화 양상을 보였다. 투석액칼슘 농도를 1.5 mmol/L로 전환한 후 1군과 2군에서 인결합제와 칼시트리올 투여 용량의 유의한 변화는 없었으나, 3군에서 칼시트리올 용량이 6개월째부터 유의하게 증가하였고 (4.4±5.1 vs. 9.6±2.9 μg/month, p<0.05) 나머지 기간 동안증가 추세를 유지하였다. 결론: 혈액투석액 칼슘 농도를 1.75 mmol/L에서 1.5 mmol/L으로 전환한 후 적절한 인결합제와 칼시트리올 투여 용량을 조정하면서 adynamic bone disease와 이차성 부갑상선항진증환자에서 혈청 iPTH 측정치가 호전되는 경향을 보였다. 신성골형성장애가 있는 혈액투석 환자에서 투석액 칼슘 농도 1.5 mmol/L를 선택하는 것이 효과적일 수 있다. Background/Aims: Whereas higher dialysate calcium (Ca) levels may pose a risk of hypercalcemia, lower levels may induce a negative Ca balance. We evaluated the effect of lowering dialysate Ca levels from 1.75 to 1.5 mmol/L and explored the appropriate use of calcitriol to regulate bone metabolism in hemodialysis patients. Methods: The dialysate Ca levels of 36 patients were reduced from 1.75 to 1.5 mmol/L. They were divided into three groups according to basal intact parathyroid hormone (iPTH) level (group 1, iPTH<150 pg/mL, n=21; group 2, iPTH 150-300 pg/mL, n=7; group 3, iPTH>300 pg/mL, n=8). Data were collected at 3-month intervals for 1 year. Results: Throughout the study period, no significant difference in phosphate binders, serum Ca, phosphorus (P), or Ca × P products was observed among groups. However, iPTH, alkaline phosphatase (AP), and calcitriol dosage patterns differed among groups. In group 1, iPTH and AP increased significantly over 12 months (p=0.01). In group 2, iPTH and AP showed no significant changes. In group 3, iPTH and AP declined significantly over 12 months (p=0.02). Calcitriol dosage did not change in groups 1 and 2, but increased significantly in group 3 (p=0.001). Conclusions: After converting hemodialysate Ca levels from 1.75 to 1.5 mmol/L, the initially different iPTH concentrations converged to a modestly elevated level. The use of 1.5 mmol/L hemodialysate Ca may thus be appropriate for both high- and low-turnover bone disease if phosphate binders and calcitriol are combined appropriately.         Expression of cellular retinoic acid-binding protein-I (CRABP-I) in the cerebrospinal fluid of adult onset moyamoya disease and its association with clinical presentation and postoperative haemodynamic change

        <P><B>Objective</B></P><P>The elevation of cellular retinoic acid-binding protein-I (CRABP-I) has been suggested as a candidate in the pathogenesis of paediatric moyamoya disease (MMD). However, few studies have addressed CRABP-I in adult onset MMD. The aim of this study was to examine the expression of CRABP-I in the cerebrospinal fluid (CSF) of adult onset MMD, and to evaluate its association with clinical presentation and postoperative haemodynamic change.</P><P><B>Methods</B></P><P>This study examined the CSF from 103 patients: bilateral MMD, n=58 (56.3%); unilateral MMD, n=19 (18.4%); atherosclerotic cerebrovascular disease (ACVD), n=21 (20.4%); and control group, n=5 (4.9%). The intensity of CRABP-I was confirmed by western blotting and expressed as the median (25th–75th percentile). The differences in CRABP-I expression according to disease entity (unilateral MMD vs bilateral MMD vs ACVD), initial presenting symptoms (haemorrhage vs ischaemia) and postoperative haemodynamic change (vascular reserve in single photon emission CT and basal collateral vessels in digital subtraction angiography) were analysed.</P><P><B>Results</B></P><P>CRABP-I intensities in bilateral MMD (1.45(0.86–2.52)) were significantly higher than in unilateral MMD (0.91(0.78–1.20)) (p=0.044) or ACVD (0.85(0.66–1.11)) (p=0.004). No significant differences were noted based on the initial presenting symptoms (p=0.687). CRABP-I was not associated with improvement in vascular reserve (p=0.327), but with decrease in basal collateral vessels (p=0.023) postoperatively.</P><P><B>Conclusions</B></P><P>Higher CRABP-I in the CSF can be associated with typical bilateral MMD pathogenesis in adults. Additionally, postoperative basal collateral change may be related to the degree of CRABP-I expression.</P>

        The gonadosomatic index (GSI), gonadal development and changes in hormones in plasma level of the indoor cultured grunt (Hapalogenys nitens) were investigated by histological study from August 2011 to October 2012. The GSI showed similar trends with gonad developmental stages during the culture periods. Changes in plasma level of estradiol-$17{\beta}$ of female H. nitens reached the highest value before the spawning period, and seasonal changes in plasma level of estradiol-$17{\beta}$ were similar in trends of oocyte developments and GSI changes. Testosterone levels of male H. nitens reached the highest value before and after the spent stage. Ovarian developmental stages of H. nitens could be classified into early growing stage, late growing stage, mature stage, ripe and spawning stage, recovery and resting stage. The testicular developmental stages could be divided into growing stage, mature stage, ripe and spent stage, and recovery and resting stage.

        N/A Objectives : Oxidized LDL is thought to play a key role in atherogenesis. Among their wide variety of biological properties, oxidized LDL injures the endothelium as an early event of atherogenesis. However, the mechanisms by which oxidized LDL injures endothelial cells are not definitely known. In order to evaluate the involvement of GTP-binding protein on the mechanism of apoptosis, we studied the effects of pertussis and cholera toxin on oxidized LDL-induced apoptosis in bovine aortic endothelial cells(BAECs). Methods : Oxidized LDL elicited apoptosis in cultured BAECs as shown by characteristic morphological and biochemical changes. Chromatin condensation and nucleus fragmentation were visualized by using fluorescence microscopy of intact cells staining by acridine orange/ ethidium bromide. DNA fragmentation was quantified by an ELISA with specific antibody for bromodeoxyuridine- labelled DNA fragments and confirmed with DNA ladder formation. Results : Studies using a combination of bacterial toxins which change the function of GTP-binding protein suggest that oxidized LDL-induced apoptosis was regulated by GTP-binding protein. Oxidized LDL-induced apoptosis was not changed by pretreatment of BAECs with pertussis toxin. In contrast, pretreatment with cholera toxin completely prevented the oxidized LDL- induced apoptosis. Conclusion : These results show that oxidized LDL induces apoptosis of BAECs and suggest that cholera toxin-sensitive G-proteins are involved in signal transduction of oxidized LDL-induced apoptosis of BAEC.

        The tendency of glioblastoma to metastasize to the cerebrospinal fluid(CSF) is well documented, reaching approximately 20% in autopsy series. Nevertheless, clinical symptoms from spinal seeding seem to be infrequent or are at least rarely diagnosed. The authors report a case of 41-year old female patient with a cerebral glioblastoma multiform who developed paraplegia due to a pathologically confirmed spinal matastasis.

        재발한 희돌기 교세포종에 대한 BCNU-CDDP 연속정주 화학요법

        Eight patients with recurrent oligodendroglioma were treated with 1.3-bis(2-chloroethyl) nitrosourea(BCNU) and CDDP continuous infusion chemotherapy. They were 5 with benign oligodendrogliomas and 3 with anaplastic oligodendrogliomas. All the recurrent tumors had been treated with surgery and radiotherapy. Four patients had already received chemotherapy with ACNU. Seven of them showed response to continuous infusion chemotherapy. The time from the response to progression was 15 to 67 weeks. No severe complication of the chemotherapy was found. In conclusion, BCNU-CDDP continuous infusion chemotherapy is an effective treatment modality in recurrent oligodendrogliomas.

        The liquid anion exchanger, Amberite LA-2, has been used to separate the trace amount of uranium from synthetic graphite in sulfuric acid solution As reducing the impurity metals with NaHSO_(3) the complete extraction of uranium has been made successfully in organic phase. The mixed solution, NaOH-HClO_(4)-H_(2)O_(2), was found to be good solution for stripping and developing of the uranium complex.
