우울증 어머니에 의해 양육된 우울하면서 소극적인 아동을 위한 아동중심 놀이치료 사례 연구

본 연구는 어머니의 우울로 인하여 적절한 양육을 받지 못해서 우울하면서 소극적인 성향을 보이는 아동에게 아동중심 놀이치료를 실시한 놀이치료 사례이다. 아동에게는 총 45회의 놀이치료가 실시되었고, 어머니 교육 및 상담을 병행하였다. 놀이치료의 목표는 안정된 분위기를 제공하여 치료자와의 신뢰관계를 형성하고, 감정을 표현하고 부정적인 감정을 해소하며, 놀이상황에서 재양육경험을 통해 자율성을 회복 하고 자신감을 증진시키고자 하였다. 놀이치료를 통해 아동의 우울증상이 감소하고, 자아 존중감과 자신감이 증가하였으며, 가정 및 학교에서도 적응적인 모습을 보여 아동중심 놀이치료가 우울하고 소극적인 아동에게 효과적인 치료적 개입으로 밝혀졌다. The purpose of this study was to investigate the effect of child-centered play therapy on a child with depression and withdrawl, due to depression of mother. The subject was a 8-year-old girl who had received child-centered play therapy during 45 sessions. The purpose of this therapy was to express negative emotion and to enhance autonomy and self-confidence, Observed outcomes of therapy reported decreased depression, heighten self-esteem and self-confidence and improved adjustment in home and school. This study supports child-centered play therapy as effective intervention for children who have depression and withdrawl problems.

Plasma levels of cholesterol (TC) and triglyceride(TG) physiologically increase during pregnancy. The lipid increment is 23-53% above the pregravidic level for TC and 2- to 3-fold of the prepregnancy level for TG. If the TC and TG are higher than normal values in pregnancy, the patient must be carefully monitored. Acute pancreatitis is the main consequence of hyperlipidemia and occurs either during pregnancy, in the third trimester, or in the puerperium. Mortality is high both for the mother (21%) and the fetus (20%). We report a case of 28 year-old pregnant woman at 29 weeks gestation with hypercholesterolemia (TC = 357 ㎎/dl) and severe hypertriglyceridemia (TG = 1300 ㎎/dl). The patient was admitted to the hospital because of severe epigastric pain, nausea and vomiting. Total serum cholesterol was increased and trigyleride was markedly increased. The electroporesis pattern of serum lipoprotein showed increase in pre-beta lipoprotein fraction, suggesting IV hypertriglyceridemia pattern. According to a review of the literature, the incidence of pancreatitis during pregnancy is 1 in 1000 to 3000 pregnancies. Severe hypertriglyceridemia in pregnancy should be treated with a careful restriction of calories and fat: for preventing acute pancreatitis, hospitalization for intravenous fluid therapy and plasma exchange must be required.
Discrimination of Turkish Propolis from Different Geographical Origins by NMR Spectroscopy

In a study utilizing NMR spectroscopy and chemometrics, propolis samples from seven diverse geographic regions across Turkey were analyzed. To identify the optimal method for studying both the antimicrobial properties and compositional variations of propolis from different regions, we investigated metabolite extraction using three solvents: water only, ethanol only, and sequential water-ethanol extraction for residual components. Notably, water-soluble components exhibited significant variation among the samples, which is particularly interesting considering the potability of propolis in water-based solutions. Furthermore, the Muğla sample displayed a distinct water-soluble profile, likely due to its unique coastal location on the Aegean Sea. This specific climate may influence the propolis' chemical composition, resulting in a different mixture of components. Interestingly, the Muğla sample contained pharmaceutically active compounds like cinnamate, ferulate, and verapamil. This research establishes a valuable foundation for further exploration of propolis' antimicrobial potential.

The recombinant expression of proteins has been the method of choice to meet the demands from proteomics and structural genomics studies. Despite its successful production of many heterologous proteins, Escherichia coli failed to produce many other proteins in their native forms. This may be related to the fact that the stresses resulting from the overproduction interfere with cellular processes. To better understand the physiological change during the overproduction phase, we profiled the metabolites along the time course of the recombinant protein expression. We identified 32 metabolites collected from different time points in the protein production phase. The stress induced by protein production can be characterized by (A) the increased usage of aspartic acid, choline, glycerol, and N-acetyllysine; and (B) the accumulation of adenosine, alanine, oxidized glutathione, glycine, N-acetylputrescine, and uracil. We envision that this work can be used to create a strategy for the production of usable proteins in large quantities.
Recombinant Expression, Isotope Labeling and Purification of the Vitamin D Receptor Binding Peptide

The vitamin D receptor binding peptide, VDRBP, was overexpressed as a fused form with the ubiquitin molecule in Rosetta(DE3)pLysS, a protein production strain of Escherichia coli harboring an induction controller plasmid. The fusion protein was bound to the immobilized metal ions, and the denaturation and renaturation of the fusion protein were performed as a part of the purification procedure. After the elution of the fusion protein, the peptide hormone was released from its fusion partner by using yeast ubiquitin hydrolase (YUH), and subsequently purified by reverse phase chromatography. The purity of the resulting peptide fragment was checked by MALDI-TOF mass and NMR spectroscopy. The final yields of the target peptide were around 5 and 2 mg per liter of LB and minimal media, respectively. The recombinant expression and purification of this peptide will enable structural and functional studies using multidimensional NMR spectroscopy and X-ray crystallography.

The integrity of the resulting peptide fragment was checked by MALDI-TOF and NMR spectroscopy. The yield of the peptide was 3.0-1.5 mg per liter in LB or minimal medium, respectively. The recombinant expression and purification of this domain will enable us its structural and functional studies using multidimensional NMR spectroscopy.

Saccharomyces cerevisiae, which is a common species of yeast, is by far the most extensively studied model of a eukaryote because although it is one of the simplest eukaryotes, its basic cellular processes resemble those of higher organisms. In addition, yeast is a commercially valuable organism for ethanol production. Since the yeast data can be extrapolated to the important aspects of higher organisms, many researchers have studied yeast metabolism under various conditions. In this report, we analyzed and compared metabolites of Saccharomyces cerevisiae under salt and pH stresses of various strengths by using two-dimensional NMR spectroscopy. A total of 31 metabolites were identified for most of the samples. The levels of many identified metabolites showed gradual or drastic increases or decreases depending on the severity of the stresses involved. The statistical analysis produced a holistic outline: pH stresses were clustered together, but salt stresses were spread out depending on the severity. This work could provide a link between the metabolite profiles and mRNA or protein profiles under representative and well studied stress conditions.
