To satisfy the increasing needs for blood transfusion it is necessary not only to find new blood donors but also to repeat the collection of blood from the same donors. It was recognized that too frequent blood donations might become detrimental to the health of the donor, above all in female donors in foreign countries where the blood program relies on voluntary donation. Unfortunately, the sources of much blood depends solely on professional donors except in a few instances in Korea. Thus, anemia of professional donors after repeated blood donation must be controlled. For this purpose, we studied hemoglobin levels and serum iron levels of professional donors, coming to Korean Red Cross Blood Service and Seoul Medical College Hospital. The data obtained in this study were as follows: (1) 6. 0% of male professional donors and 17.98% of female professional donors were disqualified owing to a low hemoglobin levels. On the other heand, only 0.8% of male voluntary donors and 10.6% of female voluntary donors were rejected. (2) 7.3% of male donors and 33.3% of female donors were rejected their donations because of low hemoglobin levels on their 5-6th visits. (3) The serum iron levels appeared to be decreased after repeated blood donations. Specially, serum iron levels of female donors decreased to 73. 6 mcg. /d. after 2-4 blood donations yearly. So, it might be recommended to limit the frequency of blood donations from female.

Malignant glioma is the most frequent type in brain tumors. The prognosis of this tumor has not been significantly improved for the past decades and the average survival of patients is less than one year. Thus, an effective novel therapy is urgently needed. TNF-related apoptosis inducing ligand (TRAIL), known to have tumor cell-specific killing activity, has been investigated as a novel therapeutic for cancers. We have developed Ad-stTRAIL, an adenovirus delivering secretable trimeric TRAIL for gene therapy and demonstrated the potential to treat malignant gliomas. Currently, this Ad-stTRAIL gene therapy is under phase I clinical trial for malignant gliomas. Here, we report preclinical studies for Ad-stTRAIL carried out using rats. We delivered Ad-stTRAIL intracranially and determined its pharmacokinetics and biodistribution. Most Ad-stTRAIL remained in the delivered site and the relatively low number of viral genomes was detected in the opposite site of brain and cerebrospinal fluid. Similarly, only small portion of the viral particles injected was found in the blood plasma and major organs and tissues, probably due to the brain-blood barrier. Multiple administrations did not lead to accumulation of Ad-stTRAIL at the injection site and organs. Repeated delivery of Ad-stTRAIL did not show any serious side effects. Our data indicate that intracranially delivered Ad-stTRAIL is a safe approach, demonstrating the potential as a novel therapy for treating gliomas.

To find anititumor metabolites in Korean basidiomycetes, the shake-cultured mycelia of eight of the higher fungi were extracted with hot water and the extracts, after being partially purified, were subjected to in vivo antitumor test. When administered i. p. at the dose of 30mg/kg/day for ten consecutive days into the female ICR mice, which had been implanted with $1{\times}10^{6}$ / cells of sarcoma 180 twenty four hours before the first injection, the extracts of Agaricus campestris, Lyophyllum decastes, Lyophyllum ulmarium, Armillaria Tabescence and Calvatia exipuliformis respectively showed inhibition ratios of 64.1%, 65.45, 60.-%, 53.0 and 49.3%. These five species were selected for further study, whereas the extracts of Phallus impudicus, Coprinus comatus and Pholiota squarrosa whih showed the inhibition ratios of 31.2%, 33.5% and 19.0% were discontinued.

To investigate antitumor constituents of Hydnum repandum, which belongs to edible basidiomycetes, the mycelia separated from the carpophore were shake-cultured and subjected to hot water extraction. The extract was concentrated under vacuum and mixed with a three-fold volume of 95% ethanol to yield precipitates. The water soluble fraction of the precipitates was dialyzed and then lyophilized to yield a water soluble protein-polysaccharide fraction (=WPPF). This exerted antitumor activity against sarcoma 180 implanted in ICR mice. When administered i.p. at the dose level of 20 ㎎/㎏ once daily for ten consecutive days, it showed an inhibition ratio of 54.3%. WPPF was found to be composed of a polysaccharide moiety (42% of WPPF) and a protein moiety (28% of WPPF) when determined by colorimetric method using anthrone reagent and Folin's phenol reagent. The polysaccharide moiety of WPPF was found to contain glucose (57.4%), mannose (19.3%), galactose (10.8%), xylose (6.8%), and fucose (5.7%), when the methanolysate of WPPF was analysed by GLC method.
