        Bacteriophage Cocktail Comprising Fifi044 and Fifi318 for Biocontrol of Erwinia amylovora

        Erwinia amylovora is a plant pathogen that causes fire blight on apples and pears. Bacteriophages, which are viruses that selectively infect specific species of bacteria and are harmless to animal cells, have been considered as biological control agents for the prevention of bacterial pathogens. In this study, we aimed to use bacteriophages that infect E. amylovora as biocontrol agents against fire blight. We isolated bacteriophages Fifi044 and Fifi318 infecting E. amylovora, and characterized their morphology, plaque form, and genetic diversity to use as cocktails for disease control. The stabilities of the two phages were investigated at various temperatures and pH values and under sunlight, and long-term storage experiment was conducted for a year. To evaluate whether the two phages were suitable for use in cocktail form, growth curves of E. amylovora were prepared after treating the bacterial cells with single phages and a phage cocktail. In addition, a disease control test was conducted using immature apples and in vitro cultured apple plantlets to determine the biocontrol effects of the phage cocktail. The two phages were morphologically and genetically different, and highly stable up to 50°C and pH value from 4 to 10. The phages showed synergistic effect when used as a cocktail in the inhibition of host bacterial growth and the disease control. This study demonstrated that the potential of the phage cocktail as a biocontrol agent for commercial use.

      • Shape-Adaptable 2D Titanium Carbide (MXene) Heater

        <P>Prior to the advent of the next-generation heater for wearable/on-body electronic devices, various properties are required, including conductivity, transparency, mechanical reliability, and conformability. Expansion to two-dimensional (2D) structure of metallic nanowires based on network- and mesh-type geometries has been widely exploited for realizing these heaters. However, the routes led to many drawbacks such as the low-density cross-bar linking, self-aggregation of wire, and high junction resistance. Although 2D carbon nanomaterials such as graphene and reduced graphene oxide (rGO) have shown their potentials for the purpose, CVD-grown graphene with sufficiently high conductivity was limited due to its poor processability for large-area applications, while rGO fabricated with a complex reduction process involving the use of toxic chemicals suffered from a low electrical conductivity. In this study, we demonstrate a simple and robust process, utilizing electrostatic assembling of negatively charged MXene flakes on a positively treated surface of substrate, for fabricating a metal-like 2D MXene thin film heater (TFH). Our TFH showed a high optical property (>65%), low sheet resistance (215 ?/sq), fast electrothermal response (within dozens of seconds) with an intrinsically high electrical conductivity, and mechanical flexibility (up to 180? bending). Its capability for forming a firm and stable ionic-type interface with a counterpart surface allows us to develop a shape-adaptable and patchable thread heater (TH) that can be shaped on diverse substrates even under harsh conditions of conventional sewing or weaving processes. This work suggests that our shape-adaptable MXene heaters are potentially suitable not only for wearable devices for local heating and defrosting but also for a variety of emerging applications of soft actuators and wearable/flexible healthcare monitoring and thermotherapy.</P> [FIG OMISSION]</BR>

        접근성 개념을 적용한 문화서비스 평가 -남양주시를 대상으로-

        인간이 생태계로부터 취하는 비물질적인 혜택인 문화서비스에 대한 수요가 최근 국민총소득과 함께 증가하고 있다. 이에 대한 정량적 가치평가를 위해 국내외에서 다수의 연구가 이루어져 왔으나 개인의 사회, 문화적 이질성 등의 주관적 성향과 규모의 차이로 인해 국내 전 지역에 적용하기에는 어려움이 있다. 문화서비스가 발생하기 위해서는 자연생태계와 인간의 접촉이 이루어져야 하며 이러한 관점을 바탕으로 본 연구에서는 접근성이라는 개념을 활용하여 기존 문화서비스 가치평가방법이 가지는 사회, 문화적 주관성을 보완하고자 한다. 국내 전 지역 규모에 적용 가능하며 다양한 이해관계자의 공감대를 형성시킬 수 있는 포괄적인 문화서비스 평가 방법을 제시하는 것이 본 연구의 목적이다. 본 연구에서는 접근성을 근거로 개발된 유럽의 여가 서비스 평가방법 고찰을 통해 국내에 구축된 도로 네트워크와 인구분포현황과 함께 국립생태원에서 수행한 ‘생태자산 간이평가‘ 결과를 활용하여 ’문화서비스 접근성 평가 (CSOS)‘를 남양주시에 시범 적용하였다. 그 결과 남양주시에 산재한 특정 생태자산 및 주거지를 중심으로 우수한 문화서비스 접근성이 존재하는 특정 공간을 도출할 수 있었다. 또한 이를 바탕으로 오늘날 보편적으로 이용되고 있는 SNS 데이터를 활용한 InVEST의 Recreation 평가 모형과 조망점을 활용한 Scenic quality평가 모형간의 차별성과 시범연구로서 가지는 가중치 설정에 대한 한계점을 고찰하였다. A cultural ecosystem service(CES), which is non-material benefit that human gains from ecosystem, has been recently further recognized as gross national income increases. Previous researches proposed to quantify the value of CES, which still remains as a challenging issue today due to its social and cultural subjectivity. This study proposes new way of assessing CES which is called Cultural Service Opportunity Spectrum(CSOS). CSOS is accessibility based CES assessment methodology for regional scale and it is designed to be applicable for any regions in Korea for supporting decision making process. CSOS employed public spatial data which are road network and population density map. In addition, the results of ‘Rapid Assessment of Natural Assets’ implemented by National Institute of Ecology, Korea were used as a complementary data. CSOS was applied to Namyangju-si and the methodology resulted in revealing specific areas with great accessibility to ‘Natural Assets’ in the region. Based on the results, the advantages and limitations of the methodology were discussed with regard to weighting three main factors and in contrast to Scenic Quality model and Recreation model of InVEST which have been commonly used for assessing CES today due to its convenience today.

        키워드 네트워크 분석을 활용한 생태자산 연구 경향 분석

        이 연구의 목적은 국내 외 생태자산 관련 연구 경향을 파악하는 것이다. 생태자산과 의미가 유사한 12개 키워드와 연관 있는 분야 및 영역들을 확인하면서 연구흐름을 이해하고자 하였다. 우리는 키워드 네트워크 분석을 이용하여 방대하게 이루어지고 있는 국내외 연구들을 분석하였다. 키워드 네트워크 분석은 정보의 구조적 흐름을 이해하고 방향성을 파악하는데 유용하다. 국외 연구들은 지속가능성을 중심으로 보전, 경제적 평가, 관리, 정책 분야로 연결되는 것을 보았다. 국내연구 분석은 문헌자료가 적어 다양한 흐름들을 확인하기 어려웠으나 지역의 활성화를 위해 경제적인 가치를 확인하고자 하는 연구들이 있었다. 이 연구는 향후 국내 생태자산 관련 연구 방향들에 설정하는데 활용할 수 있다. The purpose of this study was to determine domestic and foreign ecological asset research trends. We aimed to understand ecological assets research directions and trends by comprehensively analyzing 12 keywords, including those similar to keywords for comparable assets, to identify related fields and regions. Extensive analysis of domestic and foreign studies was conducted through keyword network analysis of textural information. This approach is helpful for understanding the flow of information and identifying research directions. Foreign studies based on sustainability were connected with 'Economic assessment', 'Management' and 'Policy' areas. It was difficult to determine domestic research trends because there are fewer domestic studies than foreign. There were studies that sought to identify economic value of developing regions. This research can be used to guide the research direction for future ecosystem asset analysis in Korea.

        Isolation of Nine Bacteriophages Shown Effective against Erwinia amylovora in Korea

        Erwinia amylovora is a devastating bacterial plant pathogen that infects Rosaceae including apple and pear and causes fire blight. Bacteriophages have been considered as a biological control agent for preventing bacterial infections of plants. In this study, nine bacteriophages (ΦFifi011, ΦFifi044, ΦFifi051, ΦFifi067, ΦFifi106, ΦFifi287, ΦFifi318, ΦFifi450, and ΦFifi451) were isolated from soil and water samples in seven orchards with fire blight in Korea. The genetic diversity of bacteriophage isolates was confirmed through restriction fragment length polymorphism pattern analysis. Host range of the nine phages was tested against 45 E. amylovora strains and 14 E. pyrifoliae strains and nine other bacterial strains. Among the nine phages, ΦFifi044 and ΦFifi451 infected and lysed E. amylovora only. And the remaining seven phages infected both E. amylovora and E. pyrifoliae. The results suggest that the isolated phages were different from each other and effective to control E. amylovora, providing a basis to develop biological agents and utilizing phage cocktails.

        국내 하천에 분포하는 ampicillin 내성균의 다양성

        임상과 자연환경에서 발견되는 항생제내성균의 문제는 보건학적 관심의 대상이 되고 있으며 환경 중의 미생물은 항생제 내성유전자 확산의 한 요인으로 판단되고 있다. 본 연구에서는 국내 하천 7지점으로부터 분리한 배양성 ampicillin 내성균의 다양성을 조사하였다. 분리된 498종의 세균들 중에서 Aeromonas와 Acidovorax 속의 내성균 분포가 가장 높았으며, 66%의 분리균들이 조사된 14종의 항생제 중 6종 이상의 항생제에 내성을 나타내었다. 그리고 7종의 항생제에 내성을 보이는 비율이 19.7%로 가장 높았다. 또한 항생제감수성 조사 결과를 바탕으로, 분포율이 높았던 Aeromonas, Acidovorax, Pseudomonas, Elizabethkingia 속의 내성균에 대한 자연내성 특징을 파악하였다. The widespread emergence of antibiotic resistant microorganisms in clinics and natural environments has attracted public concern. Especially, microorganisms inhabiting natural environment is considered as a source responsible for increasing the abundance of antibiotic resistant genes in ecosystem. In this study, the diversity of culturable bacteria resistant to ampicillin was investigated with water samples collected from seven locations in Korea. The genera belonging to Aeromonas and Acidovorax were dominant among the isolated 498 strains. The 66% of isolates showed multi-drug resistance against more than six antibiotics among tested fourteen ones and isolates resistant to seven antibiotics were the most prevalent with 19.7% abundance. Using the antibiotics susceptibility results, the intrinsic resistance profile was suggested for the most dominant genera, Aeromonas, Acidovorax, Pseudomonas, and Elizabethkingia.
