Wheatgrass is being studied because it is considered a better source of various flavonoids, phytochemicals. The Korean origin cultivar of Keumgang and Arihuk have been known for containing various functional ingredients at relatively higher level compared to imported wheat. However, the anti-inflammatory activity has not yet been clearly elucidated. Therefore, the focus of this study was to determine the anti-inflammatory effects of ethanol extracts of whole wheat and wheat sprouts of Keumgang and Ariheuk. Treatment with ethanol extracts of whole wheat grain and wheat sprouts showed low cytotoxicity and inhibition of nitric oxide (NO) production. In particular, NO production was inhibited at a concentration-dependent manner when ethanol extracts of the whole wheat grain extracts were treated. As well, it was confirmed that both the whole wheat grain and wheat sprout extracts showed the inhibitory effects on the expression of inflammatory cytokines (iNOS, CoX2, IL-6). Therefore, taken the above results together, it was believed that the ethanol extracts of whole wheat and wheat sprouts of Keumgang and Ariheuk can be developed as functional ingredients with anti-inflammatory effects.

Canavalia gladiata (Jacq.) DC., known as sword bean, is a tropical crop belonging to the legume family and has been used for food and medicinal purposes in East Asia. However, studies on the efficacy of sword bean has been limited to mature bean. Several studies have recently been focused on immature sword bean pod (ISBP), but there are still insufficient research data on their functionality. Therefore, this study was aimed to demonstrate the anti-inflammatory effects of ISBP in lipopolysaccharide (LPS)-induced RAW264.7 cells with ISBP extracted with 30% ethanol. The radical scavenging activity was significant in both ABTS assay and DPPH assay. The nitric oxide production was reduced in a dose-dependent manner, and mRNA expression of iNOS, COX-2 and IL-1β were significantly decreased by ISBP treatment in LPS-induced RAW264.7 cell line. Protein expression of p-ERK was down regulated by ISBP, suggesting that the expression of inflammatory cytokines may be regulated through the MAPK pathway. In conclusion, the ISBP could alleviate inflammation by regulating the expression of MAPK pathway and related cytokines.

Sword bean (Canavalia gladiata (Jacq.) DC.) has long been used as food ingredient and herbal medicine in East Asia. However, there has been no study on the analysis of active compounds and physiological activity according to the extraction conditions of edible part. Sword bean was extracted with hot water and different concentration of ethanol for comparative analysis. A major indicatory active component was found to be rutin which was highest at 30% ethanol extract. Total polyphenol of sword bean was highest with 70% ethanol extract. Antioxidant activity (DPPH and ABTS) and total flavonoid contents of 30% ethanol extract was the highest among the extracts. In the MTT assay, both the hot water extract and the ethanol extract did not show apparent cytotoxicity up to 2mg/ml in RAW 264.7. NO production was inhibited dose-dependently in all extracts in RAW 264.7 cells. In conclusion, the optimal extraction conditions were established with the highest amount of the active compound, highest antioxidant activity and effectiveness in reduced NO production. This study would provide useful information on the utilization of sword bean as a functional food ingredient.

Immature sword bean pod (ISBP) has long been known to be effective against bronchial diseases and inflammation. Although research on the mechanism of its efficacy is ongoing, research on plasma metabolites has not been conducted. In this study, the efficacy and metabolite analysis of hot water extract of ISBP were determined in OVA-induced mouse model. The levels of histamine and IgE increased by OVA induction were decreased in a dose-dependent manner by ISBP treatment. Histological analysis confirmed by H&E staining showed that the lung tissue damaged by OVA induction was recovered by ISBP treatment. Compared to the control group, the level of creatine was decreased and that of citrate was increased in the OVA group which indicated creatine may cause dysregulation in blood glucose and inflammation. Moreover, acidification by citrate had progressed and the increased lactate level observed in the dexamethasone and low ISBP groups was assumed to be related to reduced inflammation. Moreover, total lipid level was decreased with ISBP groups. In conclusion, this study confirmed hot water extract of ISBP treatment reduced inflammation and changed responsible metabolites.
