Optimization of Matrigel-based culture for expansion of neural stem cells

Neural stem cells (NSCs) are capable of self-renewal and can differentiate into neurons and two types of glial cells. NSCs have great potential for basic studies of neurogenesis and neurodegenerative diseases, and for therapeutic transplantation applications. NSCs can be derived from the fetal or adult brain. In vitro expansion of NSCs may help to elucidate their properties involved in neurogenesis and is crucial to obtain on-demand the large numbers of these cells needed for clinical transplantation trials. Laminin is an extracellular matrix molecule commonly used to culture NSCs as an attached monolayer. However, laminin is costly if NSCs need to be cultured in large quantities. Matrigel is a soluble basement membrane biomaterial, which has been widely used for feeder-free cultures of human embryonic stem cells. In the present study, we evaluated the ability of Matrigel to support the attachment and long-term proliferation of NSCs. We investigated the growth and adherence of NSCs derived from the fetal mouse brain on Matrigel at various concentrations (0.01–1%). We found that 0.02% Matrigel is sufficient for NSCs to be expanded in vitro in long-term culture. The NSCs cultured on Matrigel-coated plates showed typical cellular and molecular characteristics and had multipotency to be differentiated into neurons and glial cells. The Matrigel-based monolayer culture system established in this study provides a cost-effective approach to maintain NSCs in vitro in long-term cultures.

Solubility of nitrogen in liquid iron and iron-Ni-Cr alloys has been measured by the sampling method under 1 atm N₂pressure. Results obtained are as follows ; 1. Nitrogen solubility and free energy change of nitrogen solution in liquid iron melts respectively are, log (wt % N)= -357/T -1.152 ΔG˚= 6830+22.06T J/g-atom N 2. Enthalpy change in liquid Fe-8%Ni-Cr alloys is, ΔH = 11060-2.33(wt % Cr) J/g-atom N 3. Interaction parameter in Fe-8%Ni-Cr alloy is e = -128/T+0.031
일측성 성대마비 환자에서 Cross-Linked Hyaluronic Acid Gel을이용한 성대주입술의 효과 및 안전성

Background and Objectives:Hyaluronic acid (HA) is a natural polysaccharide, which resides in the extracellular space of all mammalian connective tissue and creates viscoelasticity, hydrating, lubricating and stabilizing matrix. The authors aimed to determine the clinical efficacy and safety of using HA derivative as an injection material to correct the glottal insufficiency of patients with unilateral vocal cold palsy (VCP). Subjects and Method:Twenty-eight consecutive patients with recent unilateral VCP received percutaneous HA derivative (Rofilan) injections under local anesthesia. Acoustic, aerodynamic and stroboscopic analyses were prospectively provided for the 14 subjects prior to the injection, and 1 week and 3 months after the injection. Perceptual GRBAS grading by speech language pathologists and subjective ratings of the hoarseness and aspiration by the patients were also obtained. Results:Acoustic (jitter and shimmer) and aerodynamic (maximal phonation time) parameters were significantly improved after the injection (p<0.05). Perceptual grading and subjective scaling also showed significant improvement. Those beneficial effects could be maintained for longer than 3 months but with a tendency of losing their effects with time. Early or delayed significant side effects were not observed. Conclusion:Vocal fold injection with HA derivative is a convenient, safe and useful method of correcting glottal insufficiency for more than 3 months. The patients with recently developed unilateral VCP with unknown possibility of recovery would be suitable candidates for HA injection. Further long-term follow-up studieswill answer the durability of the HA injection effect.

시속 300km로 주행하는 고속철도 열차의 주행안정성 및 승차감을 확보하기 위해서는 열차하중을 고려한 정교한 정적, 동적해석 및 설계가 이루어져야 한다. 따라서 차량과 궤도의 동적상호작용 및 차량과 교량의 동적상호작용, 터널의 미기압 및 공기압, 대단면 터널굴착의 안정성평가, 열차주행에 의한 지반진동의 예측 등에 전산수치해석기법의 활용 및 개발이 현재 활발히 이루어지고 있다. 그러나 고속철도 보유국을 포함한 선진국들에 비하면은 이러한 전산수치해석분야에 있어서 아직도 더 많은 연구 및 개발이 본 공단을 포함하여 학계 및 연구소에서 이루어져야 하겠으며, 본 경부고속철도 건설사업으로 인하여 차량, 전기시설분야의 각종 첨단기술개발 및 발전과 더불어 하부 토목구조물의 건설 및 설계 해석분야에 많은 발전이 기대된다 하겠다.
