        Colchicine과 oryzalin 처리에 의한 Cymbidium hybrids의 기내 배수화 효율 증진

        본 연구는 우리나라 주요 난과식물 중 하나인 심비디움의 배수체 육종을 위한 기초자료를 얻고자 실시되었으며, 기내심비디움의 PLBs에 colchicine과 oryzalin을 처리방법과 농도, 기간을 다양하게 하여 배수체 유도에 최적의 조건을 찾고자 수행하였다. Colchicine은 50, 100, 300, 500㎎·L<SUP>-1</SUP> 농도로 1, 2, 3주간 처리하였고 oryzalin은 3, 5, 10, 20㎎·L<SUP>-1</SUP> 농도로 1, 2, 3주 또는 3, 6, 9일간 처리하였는데 oryzalin에 비해 colchicine을 처리했을 때 생존율이 높게 나타났다. Flow cytometry를 이용하여 배수성을 검정해본 결과 Cymbidium Showgirl ‘Silky’의 경우, colchicine은 50㎎·L<SUP>-1</SUP> 농도로 1주간 처리 시 6배체 획득효율이 60%, oryzalin은 5㎎·L<SUP>-1</SUP> 농도로 2주간 처리 시 6배체 획득효율이 46.7%로 가장 높게 나타났다. Cymbidium Mystery Island ‘Silk Road’(Silk Road-4)의 경우, colchicine은 50㎎·L<SUP>-1</SUP> 농도로 1주간 처리 시 6배체 획득효율이 16.7%, oryzalin은 10㎎·L<SUP>-1</SUP> 농도로 3일간 처리시 6배체 획득효율이 6.7%로 가장 높게 나타났다. 즉, 두 hybrid 모두 oryzalin보다 colchicine을 처리했을 때 염색체 배가에 더 효율적이었다. 또한 처리 전 PLBs를 뾰족한 핀셋으로 10회 찌르는 방법을 통해 염색체 배가 효율을 향상시킬 수 있는 것으로 나타났다. This study aimed to investigate the efficiency of colchicine and oryzalin in inducing polyploidy in two Cymbidium hybrids [Showgirl ‘Silky’ and Mystery Island ‘Silk Road’ (Silk Road-4)]. Colchicine was used at concentrations ranging from 50 to 500 ㎎·L<SUP>-1</SUP>, with treatments lasting 1 to 3 weeks. Oryzalin was used at concentrations ranging from 3 to 20 ㎎·L<SUP>-1</SUP>, with treatments lasting 3 to 6 days or 1 to 3 weeks. The survival rate of PLBs was better in colchicine than in oryzalin solutions. The ploidylevels were screened using flow cytometry. In C. Showgirl ‘Silky’, the highest chromosome doubling efficiencies were obtained with the 1-week treatment in 50 ㎎·L<SUP>-1</SUP> colchicine (60%) and the 2-week treatment in 5 ㎎·L<SUP>-1</SUP> oryzalin (46.7%). In C. Mystery Island ‘Silk Road’ (Silk Road-4), the highest chromosome doubling efficiencies were obtained with the 1-week treatment in 50 ㎎·L<SUP>-1</SUP> colchicine (16.7%) and the 3-day treatment in 10 ㎎·L<SUP>-1</SUP> oryzalin (6.7%). Colchicine was more efficient than oryzalin in terms of polyploidy induction. Furthermore, pre-treatment, which entailed poking 10 times with forceps, improved the efficiency of chromosome doubling.

        심비디움 2배체, 4배체의 분화수명 조사 및 전자코를 이용한 향기패턴분석

        배수성에 따른 심비디움 꽃 향기의 변화를 알아보기 위해 전자코를 이용하여 C. Golden Elf ‘Sundust’의 2배체와 4배체 꽃을 PCA(주성분분석)와 DFA(판별함수분석)로 분석한 결과, 꽃 한 송이의 향기는 2배체에 비해 4배체에서 강하게 나타났고, 화기조직별 향기는 예주가 가장 강하게 나타났으며 꽃잎과 설판은 근접한 패턴을 보였다. 배수성에 따른 심비디움의 분화수명의 차이를 알아보기 위해 C. Golden Elf ‘Sundust’와 C. Elma ‘Orient Toyo’ 분화의 화경을 조사한 결과 개화기간뿐 만 아니라 화경당 화수, 화경장에서 모두 차이가 나타나지 않았다. 본 연구는 심비디움을 비롯한 다른 종류의 난과식물 배수체 육종에 있어 기초자료로 유용하게 활용 될 것이다. We investigated the intensity and pattern of the scent produced by diploid and tetraploid Cymbidium flowers, using an electronic nose with 6 metal oxide sensors (MOS). The MOS responses were evaluated by principal component analysis, discriminant function analysis, and sensor data. These analyses revealed that tetraploid flowers had a stronger scent than diploid flowers in Cymbidium Golden Elf ‘Sundust’. Furthermore, among the different flower partscolumn, lip, and petals-the column produced the strongest scent. There was no significant difference between the flowering periods of diploid and tetraploid potted Cymbidium Golden Elf ‘Sundust" and Cymbidium Elma ‘Orient Toyo’ grown in a greenhouse. Moreover, there were no significant differences between the number of flowers per flower stem and the length of flower stems on the diploid and tetraploid plants of these two Cymbidium cultivars. This study provides potentially useful information for the breeding of polyploidy Cymbidium in the floriculture industry.

        반응성 이온 식각법에 의해 제작된 탄소나노튜브 전극의 전기화학적 특성

        In this work, fabrication and electrochemical analysis of an individual multi-walled carbon nanotube (MWNT) electrode are carried out to confirm the applicability of electrochemical sensing. The reactive ion etching (RIE) process is performed to obtain sensitive MWNT electrodes. In order to characterize the electrochemical properties, an individual MWNT is cut by RIE under oxygen atmosphere into two segments with a small gap: one segment is applied to the working electrode and the other is used as a counter electrode. Electrical contacts are provided by nanolithography to the two MWNT electrodes. Dopamine is specially selected as an analytical molecule for electrochemical detection using the MWNT electrode. Using a quasi-Ag/AgCl reference electrode, which was fabricated by us, the nanoelectrodes are subjected to cyclic voltammetry inside a $2{\mu}L$ droplet of dopamine solution. In the experiment, RIE power is found to be a more effective parameter to cut an individual MWNT and to generate "broken" open state, which shows good electrochemical performance, at the end of the MWNT segments. It is found that the pico-molar level concentration of analytical molecules can be determined by an MWNT electrode. We believe that the MWNT electrode fabricated and treated by RIE has the potential for use in high-sensitivity electrochemical measurement and that the proposed scheme can contribute to device miniaturization.

        In this work, electrochemical characteristics and optical transmittance of carbon nanotubes (CNTs) counter electrodes which had different amount of CNTs in CNTs slurries were analyzed. Two-step heat treatment processes were applied to achieve well-fabricated CNTs electrode. Three sets of CNTs electrodes and dye-sensitized solar cells (DSSCs) with CNTs counter electrodes were prepared. As the amount of CNTs increased, sheet resistance of CNTs electrode decreased. CNTs electrode with low sheet resistance had low electrochemical impedance and fast redox reaction. On the other hand, in case of CNTs counter electrode with low density of CNTs, performance of the dye-sensitized solar cell was improved due to its high optical transmittance. We found that the transmittance of CNTs counter electrode influence the performance of dye-sensitized solar cells.

        실리콘 기판의 산화층이 다중벽 탄소나노튜브 성장에 미치는 영향

        Multi-walled carbon nanotubes (MWNTs) were synthesized on different substrates (bare Si and $SiO_2$/Si substrate) to investigate dye-sensitized solar cell (DSSC) applications as counter electrode materials. The synthesis of MWNTs samples used identical conditions of a Fe catalyst created by thermal chemical vapor deposition at $900^{\circ}C$. It was found that the diameter of the MWNTs on the Si substrate sample is approximately $5{\sim}10nm$ larger than that of a $SiO_2$/Si substrate sample. Moreover, MWNTs on a Si substrate sample were well-crystallized in terms of their Raman spectrum. In addition, the MWNTs on Si substrate sample show an enhanced redox reaction, as observed through a smaller interface resistance and faster reaction rates in the EIS spectrum. The results show that DSSCs with a MWNT counter electrode on a bare Si substrate sample demonstrate energy conversion efficiency in excess of 1.4 %.
