      • HL-60 사람 백혈병 세포에서 irinotecan이 topoisomerase I과 DI의 활성과 발현에 미치는 영향

        Background DNA topois아nerases solve the topological problems associated with DNA replication, transcription, recombination, and chromatin remodeling by introducing temporary single- or double-strand breaks in the DNA. DNA topoisomerases have been shown to be the molecular targets of many antimicrobial and anticancer agents. Among topoisomerase- targeting drugs in clinical use at present, most act by trapping the covalent DNA-enzyme intermediates to convert a normal cellular enzyme to a DNA damaging agent. This studies were designed to elucidate whether pretreatment of HL-60 human leukemia cells with the topoisomerase I - directed drug irinotecan would increase the expression of topoisomerases and to investigate the activity of topoisomerase mediated by irinotecan in nuclear extract from HL-60 human leukemia cells. Methods We have conducted experiments on cell cytotoxicity in drug-treated HL-60 cells,topoisomerase purification, topoisomerase assay using gel electrophores and oligo chip microarray analysis,respectively. Results Irinotecan inhibited the relaxation activity of topoisomerase in pUC19 DNA at various concentrations (range 0.4-50 ᄍ M〉, Irinotecan IC(50) values was 1 uM for HL-60 cells. The levels of the topoisomerase Ktype IB) and IlKtype IA) mRNA remained no significant change from oligo chip microarray analysis. Conclusion Our results suggest that topoisomerase I protein is one of the cellular targets of irinotecan but this drug does not coordinate regulation with type I topoisomerase mRNA levels.

        유기폐자원을 이용한 고도하수처리 대체탄소원 개발 및 실플랜트 적용

        The purpose of this research was evaluated economical effect to apply alternative external carbon source. Conventional activated sludge process in municipal wastewater treatment plant was adapted and introduced to Biological nutrient removal processes to meet the newly enforced effluent quality standard for nutrient removal in Korea. Low COD/NH₄^(+)-N ratio and higher nutrient concentration of influent characteristics force to inject external carbon source for denitrifying recycled nitrate. In the most case, methanol was used as external carbon source. But Methanol is expensive and very dangerous in handling. So we could find cheaper and safer external carbon source substituted methanol in last study. This alternative external carbon source is named RCS(recoverd carbon source) and a by-product of fine chemical product at chemical plant. When RCS was applied real municipal wastewater treatment plant, average 55~65% of T-N removal efficiency, 8.8mg/l of effluent T-N concentration, 11.3mg/l of effleunt COD concentration were obtained without effluent COD increase as against used methanol. To apply RCS in municipal wastewater treatment plant obtain approximately 74.5% expenditure cost reduction in comparison with methanol dosage cost.

        수분함량, 상대습도 및 pH가 냉장 우육의 색도 및 색소에 미치는 영향

        본 연구는 냉장우육의 수분함량, 상대습도, pH, 색도 및 색소의 상관관계를 검토하였다. 등심과 사태의 표면 및 내부의 수분함량과 상대습도는 저장 중 일률적인 변화를 보이지 않았고, pH는 저장기간에 따라 높아지는 경향이었다. L값은 저장말기로 갈수록 높아졌고, b값은 표면이 내부보다 낮았다. 등심과 사태의 표면 및 내부에 공통적으로 myoglobin과 metmyoglobin이 음의 상관관계가 있었다. 그리고 등심의 표면과 내부는 oxymyoglobin과 metmyglobin이 음의 상관관계가 있고, 사태의 표면과 내부는 myoglobin과 L 값이 양의 상관관게가 있으며, myoglobin과 oxymyoglobin이 음의 상관관계가 있었다. 그러나 수분함량, 상대습도 및 pH는 색의 변화와 일률적인 상관관계는 없었다. This study was investigate correlation of moisture content, relative humidity, pH, color difference, myoglobin(Myo), oxymyoglobin(Oxy) and metmyoglobin(Met). The moisture content, relative humidity of loin and shank were not changed uniformly during storage, the pH was raised according to storage period. The L value was raised at the closing stage of the storage period, the surface of the b value was higher than interior. Between Myo and Met were positive correlation in common with loin and shank. The surface and interior of loin were negative correlation between Oxy and Met, the surface and interior shank were positive correlation between Myo and L value and were negative correlation between Myo and Oxy. The moisture content, relative humidity, pH and color were not correlation uniformly.

        고등학교 지리 수업에서의 GIS 활용 방안

        이 연구의 목적은 중등학교 지리과 교사들을 대상으로 GIS에 대한 개념과 필요성에 대한 인식 및 지리수업에서의 GIS 활용의 문제점 등을 설문 조사를 통해 파악하고, 이 결과에 기초하여, 교사들이 GIS를 쉽게 활용할 수 있는 수업지도안을 개발·제시하여 중등교육에서의 GIS 활용교육을 제고하는 것이다. 이를 위하여 첫째, 교사들의 GIS 인식에 대한 설문조사를 수행하였다. 둘째, GIS 활용수업을 위한 자료개발과 학습지도안을 설계하였다. 셋째, 5차시에 걸쳐 실험수업을 실시하였다. 넷째, 실험수업후의 설문조사와 소감문 분석을 통해 수업의 효과를 고찰하였다. 연구결과를 요약하면 다음과 같다. 첫째, GIS 활용수업에 대한 교사들의 인식수준은 높았으며, GIS 활용 수업모형에 대한 욕구가 높았다. 둘째, 지리교사들이 GIS 이용의 가장 큰 제약요인으로 지적한 것이 수업모형의 부족이었다. 셋째, 실험 수업 후 GIS 활용 수업에 대한 학생들의 반응은 매우 긍정적이었으며, 실습을 통하여 학생들은 GIS에 대해 보다 심도 있게 이해하게 되었다고 응답하였다. This study researches perception of the concept and necessity of GIS shown by geography teachers of secondary schools, and through the questionnaires it grasps the realities of applying rate of GIS and the problems in using GIS. This study develops and presents teaching plans for the teachers to access easily and apply to their classes directly. In the course of the study, experimental classes have had been conducted by using five teaching plans for the commercial high school students in the twelfth grade. The results are as follows; First, geography teachers accept the idea of education done by utilizing GIS in a positive and desirable way. second, geography teachers pointed out the lack of class models as a biggest limitation factor. Third, the attitude of students who used GIS were positive, and students? understanding on GIS were improved.

      • HL-60 사람 백혈병 세포로부터 topoisomerase type 1B의 동정

        Background : DNA topoisomerases fall into two categories -type 1 and type 2. For the type 1 enzymes, the DNA strands are transiently broken on at a time; for the type 2 enzymes, by contrast, a pair of strands in a DNA double helix are transiently broken in concert by a dimeric enzyme molecule. The two types can be further divided into four subfamilies: 1A, 1B, 2A and 2B. DNA topoisomerases solve the topological problems associated with DNA replication, transcription, recombination, and chromatin remodeling by introducing temporary single- or double-strand breaks in the DNA. Camptothecin is an antitumor alkaloid that has been isolated from the Chinese tree, Camptotheca acuminata. We have developed a procedure for the simultaneous purification of type 1 and 2 topoisomerase from HL-60 human leukemia cells, and identified type 1B topoisomerase. Methods : DNA topoisomerase was purified from HL-60 human leukemia cells by a simple and fast four-step procedure : seletive ammonium sulfate precipiation, chromatography on Ultrogel A6 and DNA- cellulose, followed by ultracentrifugation on a glycerol gradient. Enzyme activity, and molecular weight were assayed by measuring the relaxation of supercoiled pUC19 DNA using agarose gell elctrophoresis and sodium dodesyl sulfate denaturing gel electrophoresis, respectively. Results Type 1 and 2 topoisomerases were isolated from extracted nucleoprotein complexes by glycerol gradient centrifugation, respectively. Human type 1B topoisomerase was purified 8.6-fold as compared to the whole cell homogenate, with 12% yield. The purified type 1 topoisomerase has a molecular weight of 100 kDa as determined by SDS polyacrylamide gel electrophoresis. The enzyme relaxes supercoiled DNA in the absence of ATP or Mg2+. Camptothecin inhibited the relaxation activity of type 1 topoisomerase in pUC19 DNA by forming a cleavable complex at various concentrations (0.4-50 uM). Conclusion : The results showed that a type 1B topoisomerase has been purified to near homogeneity from HL-60 human leukemia cells, and partial purification of type 2A from small samples was achieved by isolation of cell nuclei. Thus, the potential antitumor activity of a type 1 and 2 topoisomerase-targeted drug might be easily screened by testing the drug's ability to cause DNA cleavage in the presence of enzymes.

        Forward Link Performance of Pre-Rake TDD-CDMA systems with Multi-Antenna

        저자는 참고문헌 [2]에서 복수의 송신안테나를 이용한 두 가지 Pre-Rake 송신다이버시티 방법 (system 1과 system 2)을 제안하였고 이론적인 해석과 컴퓨터 시뮬레이션을 통해 제안 시스템의 성능을 분석하였다. 그러나, 참고문헌 [2]에서는 전체 안테나를 이용하여 송신하는 system 2 에 대하여 다중사용자 환경을 고려한 성능 평가가 이루어지지 않았다. 따라서 본 논문에서는 다중사용자 환경에 대하여 system 2의 성능을 이론적으로 해석하고 한 개의 안테나를 선택하여 송신하는 system 1의 성능과 비교한다. 두 시스템의 성능평가 결과를 통해 사용자 수가 적을 때는 system 1이 좋은 성능을 보이며 사용자 수가 많아질수록 system 2가 system 1보다 좋은 성능을 나타내는 사실을 밝힌다. 이로써 사용자 수에 따라 Pre-Rake 송신다이버시티 타입을 기지국에서 선택하여 전송하는 것이 최상의 시스템 성능을 얻기 위해 필요함을 제시한다. The authors have proposed novel TDD-CDMA systems with Pre-Rake transmit diversity schemes (system 1 and system 2) using multiple transmit antennas in [2] and have also evaluated the system performance through the theoretical analysis and computer simulation. However, the performance of system 2 which transmit a signal using all antennas has not been evaluated for multi-user environment. Therefore in this paper, we analyze the performance of system 2 for multi-user environment and compare the performance with that of the already proposed system 1 which chooses only one antenna. From the numerical results, it is found that system 2 outperforms system 1 as the number of users increases while system 1 outperforms system 2 at a small number of users. Therefore in order to achieve the best system performance, the Pre-Rake transmit diversity type should be selected at the base station according to the number of users.
