폐암의 c-Ha-ras 암유전자 발현에 관한 in situ Hybridization 검색

In order to elucidate the c-Ha-ras oncogene expression in human lung cancers, hybridization in situ was employed witha biotinylated oncogene probe, of which signal was detected immunohistochemically by the streptvidin-biotin peroxidase complex. Formalin-fixed, paraffin-embedded tissue sections from six squamous cell carcinoma of the lung yield that the expressions of c-Ha-ras were localized exclusively in cytoplasm as fine grains. Grade of c-Ha-ras expression was strong or moderately increased in 77.7% of the 18 microscopic fields from six cases, and weak in the remaining 22.3%. For specificity testing, no signal was detectable by employing hybridization in situ with exclusion of c-Ha-ras probe during the whole procedure. In addition, pretreatment with RNAse and DNAse completely abolished labeling with the biotinylated c-Ha-ras cDNA to the target tissue sections. In conclusion, these findings strongly support that quantitative increase in c-Ha-ras expression plays its role in the development and progression of the human lung cancers.
