The distributions of G to A substitution ($G^{-75}{\rightarrow}A$) mutation in the human apolipoprotein A1 (APOAI) gene promoter region and glutathione S-tran-sferase Mu1 (GSTM1) gene deletion were examined in subjects with Korean population. The $G^{-75}{\rightarrow}A$ mutation of APOA1 was genotyped by the polymerase chain reaction (PCR) and subsequent digestion of the PCR product using either Mspl or Mval (n=206). The observed numbers of GG, GA and AA genotypes were 132, 63 and 11, respectively. The allele frequencies of G and A were 0.794 and 0.206, respectively. The GSTM1 gene deletion was simply examined by the PCR amplification (n=106). The observed numbers of null type ($GSTM1^*0/GSTM1^*0$) and positive type were 55 and 51, respectively. The allele frequency of $GSTM1^*0$ was 0.720.

Spindle cell carcinoma is an uncommon type of squamous cell carcinoma characterized by a mixture of malignant epithelial and mesenchymal cells. Most spindle cell tumors are polypoid and pedunculated. It is usually detected at an early stage, removed by laryngoscope guided polypectomy at the time of diagnosis, and seems to have good prognosis. The tools for diagnosing spindle cell carcinoma are histopathological analysis and immunohistochemical analysis. With reviews of literature, we report a 72-year-old patient complaining of hoarseness and dysphagia who was later diagnosed as spindle cell carcinoma.

To elucidate antidiabetic activity of Lycii Fructus. The extract of the Lycii Fructus was subjected to bioactivity-guided fractionation. The extract of the Lycii Fructus was partitioned suceesively with hexane, dichloromethane, ethylacetate and butanol. The butanol fraction, which is most abundant in the Lycii Fructus was subjected to silica gel column chromatography and resulted in seven fractions namely Fr. I - Fr. VII. Among tested several subfractions, Fr IV. Showed the highest antidiabetic activity in the streptozotocin-induced model rat.

Hypoglycemic effect of Tabebuia avellandae was investigated in the streptozotocin(STZ)-induced diabetic rats. Diabetes was induced in male Sprague-Dawley rats by injections of STZ (45 mg/kg, i.v.). Rats weighing 200-250 g were divided into 6 groups: normal, STZ-control, hexane fr., CHCl³, fr., BuOH fr. and H₂O fr. group. Normal and STZ-control rats received 3% Tween 80 only. Four groups of diabetic rats were administered orally at doses of 100, 400, 300 and 400 mg/kg/day of hexane, CHCl₃, BuOH and H₂O fr. respectively. Fractions were administered orally to the rats for 7 days after STZ injection. All rats were anesthetized with ether, blood samples were taken by cardiac puncture for clinical chemistry and the rats were killed by exsanguination. Liver, kidney, heart and spleen were removed, weighed and analyzed. We measured glucose, protein, cholesterol and triglyceride levels in the plasma and glycogen, cholesterol and triglyceride levels in liver. The extent of blood glucose decrement in rats administered HO₂ fraction was greater than that in the STZ-control rats. The serum cholesterol and triglyceride levels were significantly lowered by administration of HO₂ fraction compared with those of STZ-control group. Treatment of rats with Tabebuia avellandae fractions caused decreases in STZ-induced elevation of cholesterol and triglyceride. Liver triglyceride level was significantly lowered hexane and BuOH fraction group compared with STZ-control group. These results suggest that HO₂ fraction of Tabebuia avellandae has the hypoglycemic action against STZ-induced diabetic rats.

Male accessory glands and ejaculatory duct of Drosophila melanogaster are reproductive organs which synthesize secretory seminal proteins. Several products of these organs involved in egg laying, receptivity, and sperm stability or storage were isolated from their lumens. Despite their secretory process play an important role, exocytosis pathway in these organs is not well known. In the present study, we characterized secretory protein profiles and determined their secretory mechanisms. Eight accessory gland secretory proteins and two ejaculatory duct secretory proteins were detected in their lumens. All these proteins were constitutively synthesized in these organs and secreted to their lumens. Secretion of newly synthesized proteins initiated at about 1 h after synthesis, and reached the peak at 4 h after synthesis. It seems that secretion of the proteins may occur via constitutive exocytosis pathway.

The effects of Taheebo on the diabetic-pregnant rats and their fetus was investigated. It has been reported that diabetic condition of the pregnant rats can affect the process of liver formation and damage the respiratory function in the fetus. Therefore we investigated the effects of Taheebo on the prevention of liver damage and respiratory failure in the fetus and those results were compared with that of dexamethasone (DXM). In pregnant rats, streptozotocin(STZ, 45 mg/kg, 0.01 M citrate buffer) was injected into the pregnant rats on the third day of pregnancy. Methanol extracts of Taheebo(500 mg/kg p.o.) was administered once daily during pregnancy. DXM (10 ㎍/g i.p.) was injected into the pregnant rats in 16th and 18th days of pregnancy. Body weights were measured and fetal number and abortion rate in pregnancy rats were determined. Lecithin/sphingomyelin ratio in amniotic fluid and malondialdehyde, glycogen, triglyceride, protein and cholesterol levels in the liver homogenate were determined. Also blood glucose level was analyzed. Body weights of maternal rats were increased in the all groups except the DXM group. Fetal number of the Taheebo treated group was similar to the control group, and a significant increase in the body weights of fetus was observed in the STZ treated group and the Taheebo treated group compared with the control group. Blood glucose of fetus produced hypoglycemia in the control group and hyperglycemia in the diabetic-pregnant rats. The protein and cholesterol levels in fetus liver were significantly increased in the DXM treated group compared with the control group. Triglyceride content was significantly increased in all groups compared with the control group. Liver malondialdehyde level of fetus in the STZ treated group was similar to the control group. Glycogen level was significantly increased in the all groups compared with the control group. Methanol extract of Taheebo showed hypoglycemic effect on the pregnant rats. However, we could not observe any hypoglycemic effect on the fetus. There`s no difference between the control and Taheebo treated group in terms of the levels of triglyceride, cholesterol, protein and glycogen in the fetus liver. Further study to identify the effect of Taheebo on the fetus is under investigation.
모세관 젤 전기영동을 이용한 안드로젠 수용체 유전자 분석

연구에서는 모세관 젤 전기 영동에서 분자체 역할을 하는 poly(ethylene oxide)를 이용하여 남성불임(Androgen receptor) 유전자의 정확한 CAG 반복 배열의 길이를 측정하였다. 표준물질인 X174 RF/Hae III, 100bp step ladder를 이용하여 non-linear regression analysis를 통한 81명의 한국의 불임 남성과 89명의 정상적인 한국 남성을 대조군으로 하여, 남성 불임과 관련된 AR gene의 exon 1의 CAG 반복 수를 측정하였다. 불임 남성의 CAG 반복 길이는 24.97±2.6 range, 17-29)이고, 정상인 남성은 23.99±2.4(range, 18-29)이다. P값은 0.018로써(<0.05) 두 그룹의 결과는 통계학적으로 주목 할 수 있는 차이를 나타내었다. gel electrophoresis (CGE) with poly(ethylene oxide) has been applied to the measurement of CAG repeat number in Androgen receptor (AR) gene related to male infertility. Non-linear regression analysis was performed using the standard X174 RF/Hae III, 100bp step ladder DNA in order to investigate the exact number of CAG repeat. For 79 Korean infertile males and 89 controls, CAG repeats at exon 1 in AR gene was compared and analyzed by CGE. It turned out that CAG repeat numbers were 24.972.6 range, 17-29) for the infertile male, and 23.992.4 range, 18-29) for the control, respectively. P value (0.018) was less then 0.05, meaning that the result was statistically meaningful.

In the previous screening on antidiabetic effect of Lycii fructus by glucose transport method using N₂-STZ diabeted rat model, each extracts showed the potent antidiabetic activity. We obtained three compounds isolated from the water fraction, EtOAc fraction and n-BuOH fraction of Lycii fructus in the present work and their structures were identified as 1-carboxy-N,N,N-trimethylmethanaminium hydroxide inner salt, 2,4(1H,3H)-pyrimidinedione and 3,3`,4`,5,7-pentahydroxyflavone-3-rutinoside. Among the constituents separated from Lycii fructus, 2,4(1H,3H)-pyrimidinedione, 3,3`,4`,5,7-pentahydroxyflavone-3-rutinoside and ascorbic acid were shown a remarkable antidiabetic effect.

For the screening of antidiabetic effect, this study was carried out to some natural products by measurements of glucose level. MeOH and CH₂Cl₂ extracts of natural products were investigated for the effect on glucose level of N₂-STZ diabeted Rat. Among the natural products, Dioscorea Rhizoma, Trichosanthes Radix, Nelumbis Semem and Lycii Fructus were shown a remarkable antidiabetic effect.
