한우육의 육질에 중요 영향을 미치는 근내지방축적 최적기에 발현되는 단백질 동정

생후 24개월까지의 한우에서 채취한 등심조직 10점으로부터 단백질을 추출한 후 이차원 전기영동법으로 이들 단백질을 분리하였다. 근내지방축적이 활발한 12에서 24개월령의 시료 단백질과 근내지방축적이 미약한 0에서 6개월령의 단백질들을 이차원 전기영동 gel에 전개하여 상호 비교함으로써 지방축적이 활발한 시기에 특이적으로 발현되는 단백질들을 선발하여 성질을 규명하고자 하였다. 참고로 지방축적능력이 떨어지는 비거세우와의 비교를 병행하여 근내지방축적이 활발한 시기에 발현이 되는 8종의 단백질을 선발하였다. 이차원 전기영동 gel에서 단백질을 분리하였기 때문에 각 단백질의 정확한 등전점과 분자량을 알 수 있었고 이들 정보를 이용하여 SWISS-PROT의 단백질 database를 ExPASy server를 통해 검색하여 동일 단백질일 가능성이 높은 여러 종류의 단백질 이름을 얻게 되었다. 이들 단백질을 ammonium sulfate로 분획하고 이차원 전기영동하여 순수 분리시킨뒤 PVDF 막에 옮겨주어서 아미노산의 microsequencing을 하였다. 성공적으로 얻은 세 개의 아미노산 서열은 모두 예상한 단백질이 선발한 단백질과 동일 단백질임을 확인시켜 주었고 아직 아미노산 서열을 모르는 단백질들도 동일 단백질일 가능성이 높음을 시사했다. 검색결과 알려진 단백질들은 고기의 색깔, 지방운반, 향미성분 축척에 연관될 수 있는 기능을 가진 단백질들임이 알려졌다. Proteins from the loin tissues age-ranged from 0 to 24 months of ten Korean cattle were extracted, separated and compared on two-dimensional(2-D) gels to identify the proteins whose expression is highly correlated to marbling. We also compared the difference of loin proteins between castrated and non-castrated bull cows on two-dimensional gels. As the marbling in the loin of the cattle is optimized at 18 to 24 months, eight proteins expressed significantly higher level in 24-month than in 0- or 6-month were selected in terms of isoelectric points(pIs) and molecular weights. Using these values, we searched the Swiss-Prot database via the ExPASy molecular biology server with TagIdent program. The proteins with the nearest molecular weights and isoelectric points were selected from the lists. These possible candidates were confirmed by N-terminal microsequencing of the eight selected proteins. Three proteins, myoglobin, hemoglobin and ATPase, whose N-termini were not blocked could be microsequenced and found to be exactly matched to the selected candidates. It is suggested that the proteins increasingly expressed in marbling periods can be involved in meat color, lipid transport and flavor improvement.

In this study, study about fly ash and blast-furnace slag used as substitutive materials for cement, and the influence on the neutralization of high flow concrete durability with these substitutive materials was performed and analyzed. The results are as follows 1) Aggregate segregation was partially improved with the progress of the admixture input at the mix proportion above slump flow 65 cm 2) Compressive strength with the progress of the increasement of fly ash input was decreased in early age, but decrease range was improved in long term age. Also, in case of blast-furnace, similar or improved compressive strength was appeared. 3) The neutralization depth with fly ash input was noticeably increased. But blast-furnace slag was effective to prevent. 4) In this experiment, high powder content was advantageously affected on preventive effect of the neutralization, and the relationship between the compressive strength and the neutralization depth was inversely proportional.
신생아 편측 신우요관이행부 폐색의 자연경과 : 중증 수신증 5례

목 적 : 신우요관이행부 폐색에 의한 신생아 편측수신증의 치료는 수술요법과 대기요법 사이에서 여전히 논란의 여지를 제공하고 있다. 이는 자연경과에 대한 불확실성과 정확한 진단방법의 부재에 기인하고 있다. 이에 저자들은 수신증의 정도가 심하고 폐색이 의심되나 신기능을 잘 유지하고 있는 편측 수신증의 신장을 전향적으로 관찰하여 이의 자연경과를 이해하고자 하였다. 방 법 :지난 3년간 산전초음파로 진단된 편측수신증 환아 중, 수신증의 정도가 3등급 이상이고 이뇨성 DTPA신주사상 요로폐색을 나타내지만, 분리신기능이 $40\%$이상인 환아 5례를 대상으로 3개월마다 뇨검사, 초음파검사와 이뇨성신주사를 시행하며 추적 관찰하였다. 결 과 :평균 추적관찰 기간 15개월 동안 5례 모두에서 분리신기능의 저하 없이 추적이 가능하였으며, 수신증의 정도는 약간의 호전을 보였으며, 배설지연의 정도 또한 호전 내지는 그대로 유지가 되었다. 결 론 :중증 수신증의 신우요관이행부 폐색 환아에서 분리신기능이 좋을 경우 수신증의 정도나 배설지연의 정도에 의존한 성급한 수술적 치료는 지양되어야 하며, 분리신기능이 유지되는 한 지속적인 추적관찰을 할 수 있으리라 생각된다. Purpose: Unilateral ureteropelvic junction obstruction with high grade of hydronephrosis an obstructed diuretic renogram but good differential renal function were studied to evaluate the natural history of untreated obstruction. Materials and Methods: During the last three years, we encountered 18 kidneys with hydronephrosis detected on prenatal ultrasound. In all these patients other urologic abnormalities were excluded by voiding cystourethrography and other radiological studies. Diuretic renography was initially performed at an age ranging from 2 weeks to 4 weeks. Five had high degree of hydronephrosis and obstructed diuretic renogram despite good relative renal function. Periodic urine examination, ultrasonography and diuretic renogram were done at regular intervals. Results: We have followed 5 neonates with unilateral hydronephrosis and suspected ureteropelvic junction obstruction with noroperative treatment for 15 months. During followup, percentage and relative renal function were maintained in all patients and hydronephrosis improved. Obstructive patterns on diuretic renogram were variable. Conclusions: These findings help to define the natural history of untreated good functioning hydronephrotic kidneys. Many newborn kidneys with severe hydronephrosis are not obstructed despite even high grade of hydronephrosis and obstructed diuretic renogram.

Protein profiles of beef loin were constructed by comparing two-dimensional gel electrophoresis patterns of the proteins from different growth stages of Hanwoo, Korean cattle. Proteins from the lean muscle of 0, 6, 12 and 24 months old Hanwoo were separated by isoelectric focusing (IEF) on 16 cm tube gels, and further processed second dimensionally by 12% SDS-polyacrylamide gel electrophoresis (PAGE) using 18 × 20 ㎝ gel. Proteins with pI values ranging 3.0 to 9.0 and molecular weight of 15 to 100 kDa could be clearly detected on gel by silver staining. Interestingly, many of the proteins significantly increased and decreased during growth happened to be low molecular ones. To isolate the increased proteins, the soluble proteins were obtained from the tissue by 1% Triton X-100 extraction, then, fractionated by 30% and 50% ammonium sulfate. The isolation condition of each particular protein was determined. According to the conditions, two of the increased proteins were isolated, and transferred to PVDF membrane and microsequenced.
