        벼 재조합집단을 이용한 Low Glutel in 계통의 품질분석 및 유망계통 육성

        고령화 사회로 접어들면서 만성 신부전증 환자는 증가되고 있으며, 저단백질 식품에 대한 수요도 증가되고 있다. 중국173/주남벼 재조합 집단을 이용하여 저Glutelin 고품질 벼 품종개발의 육종효율 증진을 위해 DNA마커의 이용성을 검토하였으며, 밥맛향상을 위해 미질관련 형질들과 상관분석을 수행하였다. 중국173호/주남벼 재조합 집단에서 SDS PAGE결과 얻어진 저Glutelin 표현형과 functional DNA marker의 유전자형이 일치하여 DNA marker를 이용한 선발이 가능함을 확인하였다. 밥맛향상을 위해 미질관련 형질들과 상관분석을 수행한 결과, 윤기값과 Amylose 함량은 유의한 부의 상관을 나타내었으며, 단백질 함량은 윤기값에 많은 영향을 미치지 않았다. 반면, 단백질 조성에 따른 저Glutelin 계통군과 정상계통군의 비교에서 저Glutelin 계통군은 정상군에 비해 유의하게 윤기값이 낮았으며, 저Glutelin-저Amylose 계통군과 정상계통군은 대등한 윤기값을 유지하였다. 따라서 고품질 저Glutelin 품종개발을 위해 저Amylose 유전자의 활용이 바람직한 것으로 판단되었다. 이들 분석결과와 주요 농업적 특성을 고려하여 저Glutelin-저Amylose인 유망계통 밀양262호를 육성하였으며, 금후 고품질 저Glutelin 품종개발에 활용될 것으로 기대된다. In chronic renal failure, to reduce the burden on the kidneys, it is necessary to provide patients with therapy based on a low-protein diet. This study was conducted to improve eating quality for developing low easy to-digest protein rice variety with Lgc1 gene. Two hundred twenty six recombinant inbred lines (RILs) were used, which were derived from the cross between Chugoku173, a low glutelin rice variety developed in Japan, and Junambyeo, a high quality rice variety in Korea. We investigated glutelin phenotype using SDS PAGE and its genotype by functional DNA marker. As results of them were consistent, this DNA marker is expected to use of MAS for the Lgc1- mediated low glutelin variety. The eating quality of rice measured by Toyo meter (GMT) showed significantly negative correlation with amylose content, but there was no significance with protein content in this population. Dividing the RILs into normal amylose group (app. 19%) and low amylose group (app. 13%), the eating quality estimated by GMT was generally lower in low glutelin lines, but that of low amylose group was higher than that of normal amylose group. On the basis of several results, it is supposed that low amlyose gene can be used to breed the low glutelin rice variety with high eating quality. Through the analysis of agronomic traits related to eating quality, Milyang262, low glutelin and amylose rice variety, were developed.

        손상된 흰쥐 척수에 태아 척수 조직 이식이 미치는 영향

        Objective:Previous studies have demonstrated that axon regeneration or remyelination after spinal cord injury occurs when provided with a suitable substratum such as fetal spinal cord (FSC). We carry out this study to determine whether FSC transplants can reduce the glial scar at the interface between host and graft. Methods : Hemisectioned spinal cord injury was made by aspiration at T3 or T4 spinal cord level in rat. Cell suspension of E-14 FSC was introduced into the injured cavity contaning glial scar tissue. To indentify the transplanted cells from host tissue, FSC cells were labeled with DiI. Rats were sacrificed at 1,2,3, and 8 weeks after transplanation and spinal cord was undergone serial sections for immunocytochemistry and histological observation. The observation by electron microscope was carried out too. Results : We could observe that the FSC transplants survived in host spinal cord and generally occupied most of the neuron-depleted area. Examination of serial sections through the graft-host interface which had been immunoreacted for glial fibrillary acidic protein demonstrated that the glial scar was no longer a continuous wall separating the graft and host tissues at eight weeks after injury. We could observe oligodendrocyte and the reformed myelin at the interface by electron microscope. Conclusion : The fetal spinal cord transplant can reduce an established glial scar or restrict the reformation of a scar following surgical manipulation, and that the FSC transplant can promote remyelination.         Mild Water Deficit Increases the Contents of Bioactive Compounds in Dropwort

        The objective of this study was to determine the effects of mild water deficit on the contents of bioactivecompounds in dropwort (Oenanthe stolonifera). Dropwort plantlets with 2-3 offshoots were transplanted into plastic potscontaining horticultural growing medium. The plantlets were grown at 22°C under a relative humidity of 60%, photosyntheticphoton flux density (PPFD) of 285 μmol·m-2·s-1, 12-hour light period, and CO2 concentration of 600 μmol·mol-1andsubirrigated with nutrient solution for 3 weeks after transplanting. To induce mild water deficit, we placed wick culturesystems (4- and 6-cm long wicks) between the bottom of each pot and the water surface at 3 or 4 weeks after transplanting. The control plants were subirrigated at the pot bottom without wick treatment. The soil water content of plants underwater-deficit averaged 45% lower than that of the control at 5 weeks after transplanting. Moreover, mild water-deficittreatment reduced the leaf water potential by 7-25% compared to the control. However, mild water-deficit treatmentdid not significantly affect the fresh or dry weights of dropwort shoots at 6 weeks after transplanting. All treatmentssignificantly inhibited the photosynthetic rate at 5 weeks after transplanting. In particular, water-deficit treatment usinga 6-cm long wick for 2 weeks resulted in a 16% lower photosynthetic rate than that of the control. The totalphenolic content and antioxidant capacity of dropwort stems did not significantly differ between treatments. However,in the leaves, total phenolic content and antioxidant capacity of all plants subjected to water-deficit treatments averaged43 and 64% higher, respectively, than those of the control. The total anthocyanin content in all treatment groups,except for treatment with a 4-cm wick at 4 weeks, was significantly higher than that of the control. These resultssuggest that dropwort plants subjected to mild water deficit effectively accumulate antioxidant phenolic compounds,including anthocyanins, which is more reminiscent of reddish, small dropwort plants that grow under limited waterconditions than of water dropwort.

        KVN과 동아시아 VLBI 관측시설을 이용한 SiO 메이저 모델이미지 모의실험

        We report results of image simulations of the KVN and VLBI experiments of the KVN with several other East Asia VLBI facilities. To investigate their imaging capability a model-generated image of 7 mm SiO maser emission in Mira variables is used. The resulting simulations show that the joint VLBI experiments of the KVN with East Asia VLBI facilities can produce reasonably good images at 7 mm spectral line experiments. However, there are no apparent differences in peak flux densities and images themselves in the simulations among different combinations of these facilities. In addition, the simulated images of observations which include bigger antennas do not show any expected improvement to the image sensitivity. The small variations in the peak flux density and similar image sensitivity, irrespective of different antenna sizes or numbers of baselines used in the simulations, turn out due to a specific characteristic of the adopted model image. Test simulations using another SiO maser image from R Cas observations prove that the participation of bigger antennas in the VLBI experiments does improve image sensitivity. We confirm the need of additional longer baselines in the experiments of the East Asia VLBI facilities to study very compact maser clumps on sub-milliarcsecond scales.

        통일형 벼에서 메소트리온계 제초제 저항성 연관 DNA marker 탐색

        This study was conducted to identify DNA markers related to resistance to herbicide containing mesotrione in Tongil type rice. Two Tongil type elite lines; Milyang154 and Suweon382, showed resistance to mesotrione, whereas the others were susceptible at 20 days after mesotrione application, and severe growth inhibition was observed in the remaining 13 lines. As a result of analysis of mesotrione resistance using 190 F2 populations derived from a cross of Hanareum2 (susceptible) and Milyang154 (resistant), the mesotrione resistance locus was shown to be a single dominant gene with a 3:1 segregation ratio (X2=1.19, P=0.31). To identify a DNA marker closely linked to the mesotrione resistance gene, bulked segregant analysis (BSA) was adopted. The DNA marker RM3501 was identified on chromosome 2 with a recombinant value of 0.53 to the mesotrione resistance gene. Mst1(t) was located between SSR (simple sequence repeat) markers RM3501 and RM324 with a physical map distance of 10.2 Mb–11.4 Mb on chromosome 2. The band pattern of agarose gel electrophoresis of the SSR marker RM3501 showed the same segregation pattern with respect to mesotrione treatment in 20 Tongil type varieties and a BC2F2 segregation population derived from a cross between Unkwang (resistant) and Hanareum2 (susceptible). Thus, the RM3501 DNA marker could be used in breeding programs for Marker Assisted Selection in mesotrione resistant rice breeding.

        Ethanol Extract of Bupleurum falcatum Improves Functional Recovery by Inhibiting Matrix Metalloproteinases-2 and -9 Activation and Inflammation after Spinal Cord Injury

        Matrix metalloproteinases (MMPs) are zinc-dependent endopeptidases that degrade the extracellular matrix and other extracellular proteins. Upregulation of MMPs activity is known to be required for the inflammatory cell infiltration after spinal cord injury (SCI) and most likely contributes to early blood spinal barrier disruption and inflammation, thereby leading to the impairment of functional recovery. Here, we examined the effect of ethanol extract of Bupleurum falcatum (BF) on functional recovery by inhibiting MMP-2 and -9 activation and inflammation after SCI. Rats received a moderate, weight-drop contusion injury to spinal cord were administered orally with BF at a dose of 100 mg/kg for 14 d and functional recovery was measured by Basso- Beattie-Bresnahan locomotor open field behavioral rating test, inclined plane test and foot print analysis. To examine the neuroprotective effect of BF, TUNEL staining and counting were also performed. In addition, the expression and/or activation of MMP-2, MMP-9 and inflammatory mediators such as TNF-Ձ, IL-1Ղ, COX-2, and iNOS were examined by RT-PCR and gelatin zymography using spinal cord tissue from 1 d after injury. Our data showed that BF significantly inhibited the expression and activation of both MMP-2 and MMP-9 after SCI. The mRNA expressions of TNF-Ձ, IL-1Ղ, COX-2, and iNOS were also significantly attenuated by BF. Furthermore, BF reduced apoptotic cell death at 1 d after injury, thereby significantly reduced lesion volume and improved functional recovery. Taken together, these results suggest that BF can be used as a potential therapeutic agent for treating acute spinal injury.
