Facile method for immobilization of genome editing protein based on dopamine polymerization

The CRISPR/Cas9 has attracted great attention due to the high simplicity and selectivity in editing the gene of interest. Immobilization of the CRISPR system on nonfunctionalized substrate surfaces with high efficiency would have various applications in biomedical research and diagnostic purposes. In this study, we introduce a facile method for surface immobilization of the CRISPR ribonucleoprotein mediated by polydopamine formation. A polypropylene surface was modified by dopamine polymerization, which can then readily react with the Cas9 protein by nucleophilic addition reactions. The efficiency of immobilization and bioactivity of Cas9 was characterized. The current approach can be used for immobilization of various proteins onto surfaces that are difficult to chemically functionalize while maintaining protein bioactivity.

고해상도 카메라 혹은 영상레이더 안테나를 장착하여 지구를 관측하는 인공위성의 구조체는 발사하중 및 우주환경의 궤도 상에서 탑재된 장비를 보호하게끔 설계되고 제작된다. 구조체는 발사체와의 공진을 피할 수 있는 강성을 가져야 하며, 주 구조물의 강도는 발사체로부터의 하중을 견딜 수 있도록 설계된다. 또한, 극한 우주환경 하에서 구조체의 변형이 최소화 되도록 설계된다. 상기 설계 내용이 완벽하게 구조체에 반영되기 위해서는 우주용 자재 및 공정의 적절한 선정이 이루어 져야 한다. 이 논문은 인공위성 구조체에 사용된 Metal, Non-metal 및 조립용 Hardware 자재 (규격 포함)와 측면패널/플랫폼 및 태양전지판 Substrate 등 주요 구조물의 제작공정에 대하여 기술한다. 그리고, 국산화가 이루어진 조립용 Hardware의 Dry Film Lubricant 공정에 대해서도 기술한다.
그래픽 노블의 문학적·예술적 위치와 정당성 -그래픽 노블『왓치맨』을 중심으로-

The purpose of this study is to prove the independence and legitimacy of graphic novel through the discussion on its definition and distinction from comics. Since the 1960s American graphic novels started as underground comics and made their positions firmly in the publication market with the pieces that were focused on their literature and artistic value for their fandom. Despite of a few fans, these books tried to pursue experimental and innovative design and this led to the competitions among the graphic novels with high qualities. The situation begun to impress the ordinary readers who are not familiar with graphic novels. They assured literacy critics the possibility of graphic novels as the texts dealing with the social issues such as personal experiences, history, religion, ideology and feminism. This research has chosen graphic novel, 『Watchmen』by Alan Moore as a case study and analyzed its literary, artistic and social values. As the result, the paper firstly concluded that the condition of book publication and its professionalism produced a number of creative and experimental graphic novels. Second, after the Cold War Era ended, the graphic novels focused with literary value started to appear in public, and this made the medium get a strong position. Third, graphic novels were basically born with the media hybridity such as graphic and novel, image and language, visual art and literature. Fourth, it is possible that the writers of graphic novel build a complicated narrative structure. They also have possibilities of segment, compression, attach and reiteration of time and space so that there can be various art direction. Fifth, the writers want their work to have a distinct design concept that corresponds perfectly to the world they create. Finally, the recent graphic novel pieces show that the media frees itself from the cliche that they had in the past and seeks for its own aesthetics. It also is qualified with mass appeal to be acknowledged as a independent cultural media.
슬링을 이용한 열린사슬운동과 닫힌 사슬 운동의 턱당기기 동작 시 목굽힘근 두께 비교

Purpose: The purpose of this study was to investigate the effect of open and closed kinetic chain exercises with a sling on neck flexor thickness during chin-in movement in office workers with forward head posture. Methods: Thirty-one selected subjects randomly performed chin-in movement in open kinetic chain and closed kinetic chain exercises using a sling. All subjects were measured for their neck flexor thickness using ultrasound measurement equipment during the performance of chin-in movement in open and closed kinetic chain exercised. A paired t-test was used to compare the thickness of the total neck flexor, superficial, and deep neck flexor, respectively, between open and closed kinetic chain exercises with chin-in movement. Results: The thickness of the superficial neck flexor diminished in size more greatly during the chin-in movement with the closed kinetic chain than the open kinetic chain, but this was not a statistically significant difference (p > 0.05). On the other hand, the thickness of the total neck flexor and deep neck flexor had more greatly increased during the chin-in movement with the closed kinetic chain than the open kinetic chain, and this was a statistically significant differences (p < 0.05). Conclusion: This study suggested that chin-in movement with a closed kinetic chain is more effective in activating the deep neck flexor than the open kinetic chain. Thus, we believe that the closed kinetic chain exercise using a sling is an effective intervention to correct the postural alignment of individuals with forward head posture.
Tumor-targeted delivery of CRISPR plasmid using HA-functionalized carbon dots

Gene therapy is a promising technology for cancer treatment due to the versatility and specificity of the drug. The CRISPR/Cas9 system has been attractive as a gene therapy since the target gene can be reprogrammed with high efficiency and selectivity. A non-viral method can effectively deliver the cargo to the target site without the problem from the viral method. Herein, polyethyleneimine carbon dots (PEI Cdots) were used to package efficiently the CRISPR plasmid and delivery them into cancer cells. A CRISPR plasmid which expressing Cas9 and sgRNA was prepared, complexed with PEI Cdots and functionalized with hyaluronic acid (HA) to both stabilize the complex and specifically target CD44 expressed on tumor cells. The HA-functionalized PEI Cdot/plasmid complex was characterized by dynamic light scattering and zeta potential measurements. Delivery of the complex into tumor cells was investigated and observing cellular uptake and knockout of the reporter gene by confocal microscopy.
Study on Sodium Chloride by X-ray Diffraction Method

Seventeen intensity peaks of Sodium Chloride were Collected by means of powder camera. The maximum 2θ yalue measured was 160° 42'. The plane identification of each peak showed that this crystal belongs to face-centered-cubic and a=5.647A. The theoretical and practical intensities of the reflecting planes are in good agreement with each other.
