The developed final shattering machine for labor-saving mechanization of shattering of sesame consisted of input part, shattering part, re-shattering part for unshattered pod and pneumatic sorter. The bundle of sesame was held as upside down and fed into the machine continuously. Then, the fed bundle of sesame was shattered by side shock and agitation. The performance of shattering for the sun dried bundle of sesame of conventional manual work and final shattering machine was compared. Since the shattering ratio measured by the final machine was 97.2% at the first operation, in case of fully dried sesame by drying stand, the harvest of sesame can be completed by only one time shattering operation. The work hour per area of 10 a for the mechanical work and the manual work were 0.3 hour and 13.9 hour, respectively. The total shattering ratio of the final machine with vertical feedings of bundle of sesames was 97.2%.

위치지정 변이기법을 이용한 Bacillus thuringiensis var. kurstaki NRD 6-Stu 1의 육성

위치지정 변이기법을 이용하여 Bacillus thuringiensis var. kurstaki NRD 6의 내독소 유전자의 N말단 독성부위를 변이시켜 Bacillus thuringiensis var. kurstaki NRD 6-Stu 1을 육종하였다. 내독소에 대한 염기서열 조사결과, 변이주의 내독소 유전자부위에 Stu 1 인식부위가 생성되었고 특히, 이부위의 177번 염기 A가 C로 치환된 silent mutation이 일어났음을 확인하였다. 변이주의 내독소 유전자의 항원성질은 친주와 차이가 없었고 Choristoneura femiferana-1에 대한 독성도는 0.015~0.030ng으로 친주의 0.01~0.024ng과 유사하였으나 다른 친주계열인 B.t.kurstaki NRD 5의 500~1000ng과 B.t.kurstaki NRD 4(무독성)보다 강하였다. A cloned delta-endotoxin gene from Bacillus thuringiensis var. kurstaki NRD 6 was mutated at N-terminal toxic portion of the protoxin by site-directed mutagenesis. A Stu 1 restriction site was created in protoxin region of the mutant B.t.kurstaki NRD 6-Stu 1 which changed A to C in 177 region of the protoxin(silent mutation). Antigenic property of crystalline inclusion (delta-endotoxin) of the mutant was no difference to intact strain by the western blot analysis and its toxidity threshold value was also showed 0.015~0.030ng against Choristoneura femiferana-1 insect, similar with 0.01~0.024ng of parent strain, but stronger than that of B.t.kurstaki NRD 5 (500~1000ng) and B.t.kurstaki NRD 4(none toxic).
소목 추출물의 구강암 및 골육종 세포주에 대한 항암작용에 관한 연구

Anticancer effect of methanol extract of Caesalpinia sappan L. on oral carcinoma (KB) and osteosarcoma (HOS) cells were investigated in this study. In order to elucidate the anticancer mechanism of Caesalpinia sappan L, we analyzed telomerase inhibitory effect of the methanol extract of Caesalpinia sappan L. In addition we prepared 5 fraction samples according to its polarity differences and analyzed anticancer effects on oral carcinoma and osteosarcoma cells. Following results are obtained in this study. 1. 50% cell proliferation inhibitory value ($IC_{50}$) of the methanol extract of Caesalpinia sappan L. against oral carcinoma (KB) cells and osteosarcoma (HOS) cells were $9.0{\mu}g/ml$ and $10.9{\mu}g/ml$, respectively. 2. The methanol extract of Caesalpinia sappan L. showed inhibitory effect of telomerase which is required for cancer cell immortality. Therefore, it seems that the anticancer effect of methanol extract of Caesalpinia sappan is at least partially due to telomerase inhibitory effect. 3. Five fraction samples were prepared according to its polarity and 88.7% of ingredient of total methanol extract was transferred to ethylacetate fraction. Thin layer chromatography analysis showed that dichloromethane fraction contained ingredient with relatively high polarity and ethylacetate fraction contained similar ingredient found in total methanol extract. 4. Anticancer effect was observed in n-hexane, dichloromethane, and ethylacetate fractions. The highest anticancer effect was found in dichloromethane fraction which had $IC_{50}$ value of 4.4 and $>4.0{\mu}g/ml$ against oral carcinoma (KB) cells and osteosarcoma (HOS) cells, respectively.
보강된 복합재 패널의 최적설계를 위한 유전알고리듬의 연구

The paper describes the use of genetic algorithms (GA's) to the minimum weight design of stiffened composite panels for buckling constraints. The proposed design problem is characterized by mixture of continuous and discrete design variables corresponding to panel elements and stacking sequence of laminates, respectively. Design space is multimodal and non-convex, thereby introducing the need for global search strategies. Advanced strategies in GA's such as directed crossover, multistage search and separated crossover are adopted to improve search ability and to save computational resource requirements. The paper explores the effectiveness of genetic algorithms and their advanced strategies in designing stiffened composite panels under various uniaxial compressive load conditions and the linrlit on stacking sequence of laminates.
호알칼리성, 고온성 Bacillus sp. TA-11의 분리와 $\beta$-galactosidase의 생산

$\beta$-galactosidase를 강력하게 생산하는 호알칼리성, 고온성 세균 TA-11를 부엽토에서 분리하여 Bacillus sp. TA-11로 동정 하였다. Bacillus sp. TA-11에 의한 $\beta$-galactosidase 생산은 lactose 1.5%, peptone과 yeast ext. 각각 0.4%, $MgSO_4$ 0.2%, $NH_4$CL 0.05% 및 NaCl 0.2% 등을 함유한 배지의 pH를 10.0으로 하여 시험균을 접종한후 $50^{\circ}C$에서 2일간 진탕배양 하였을 때 가장 좋았다. A alkalophilic, thermophilic bacterium TA-11 which produce $\beta$-galactosidase highly was isolated from compost and identified to the genus Bacillus. $\beta$-galactosidase production was maximized when it was incubated in synthetic medium containing 1.5% lactose. 0.4% peptone, 0.4% teast ext., 0.2% $MgSO_4$ 0.05% $NH_4$Cl and 0.2% NacL(initial pH; 10.0) at $50^{\circ}C$ for 2 days in reciprocal shaker.
