불완전 진균 Microsporum nigricans Yeeh sp.nov의 특성

토양으로부터 dermatophyte로 인정되는 불완전 곰팡이를 분리한 후 그 형태학적 및 생리학적 특성등을 검토하여 이미 알려진 종과 비교한 결과 분리주는 Microsporum속에 속하였으나 colony의 색깔, macroconidium의 형태 또는 세포수 등과 관련하여 관련종인 Microsporum gypseum 및 Microsporum nanum과 비교해서 육안적.현미경적인 다른 특징을 나타내었다. 이 분리주는 Microsporum 속의 아직 보고된 바 없는 새로운 종으로 사료되었으며 육안적으로 진한 흑색을 나타내었으므로 Microsporum nigricans Yeeh sp. nov.로 이름붙였다. An imperfect fungus recognized as a dermatophyte was isolated from soil samples and the macroscopic, microscopic and physiological characteristics were examined. From the comparisons of this isolate with the known species, the strain belonged to the genus Microsporum. She showed mant different characteristics from the related species, including colony color and shape, or number in a cell of macrocondium. This isolate was considered to be a novel species in the genus Microsporum. Therefore the name was given to her as Microsporum nigricans Yeeh sp. nov. because of dark blackish color from the macroscopic observation.

Cadimium ion-resistant microorganism was isolated from the sludge of wastewater. The physiological, morphological and other cultural data showed that this strain belonged to Citrobacter freudii. A clearcut distinction of growth among nutrient broth, typtic soy broth and synthetic medium was demonstrated. The resistant cells showed only slight mutagenic action. During the growth of bacterial population in resting state, the organisms reduced the initial level of resistance to cadmium ions when they were not kept in contact with cadmium ions in bacteral multiplication. And cadmium ion-resistant and cadmium ion-sensitive strain were found to show equal, lower or higher sensitivity to other heave metals.
Characterization of cytosine deaminase with substrate specificity to 5-fluorocytosine

A cytosine deaminase from the cell-free extract of an isolate was examined after ethyl alcohol reactionation. The enzyme catalyzed the conversion of 5-fluorocytosine to 5-fluorouracil by the possession of specificity to the substrate. The optimum temperature and storage time on the stability of the enzyme were at below $50^{\circ}C$ and near 2 days in tris-HCl buffer. The maximum activity was also presented ar 9.0 in pH and $45^{\circ}C$ in temperature. The pHs and temperatures for the enzyme activity ranged from 8.5-9.5 and from 40-$50^{\circ}C$, respectively. the presence of $Ag^{+}, Hg^{2+}, Zn^{2+}$ in the reaction mixture resulted in the marked inhibition in the activity, but 1mM of $Fe^{3+}, K^{+}$, or $Na^{+}$ increased the enzyme activity. The enzyme preparation was vot affected by inhibitors used except N-ethylmaleimide of 1 and 10mM, and considerably activated by 1mM of pyrophosphate and 10mM of phosphate.

Some properties of an extracellular cytosine deaminase produced from Arthrobacter sp.JH-13 were examined after 20-80% of ammonium sulfate fractionation. Among some substrates, this enzyme utilized cytosine and 5-fluorocytosine as a substrate. The optimum pH and temperature for the activity of this enzyme were found to be near 8.0 and $40^{\circ}C$, respectively. The ensyme was more stable in 0.2M of Tris-HCl buffer than 0.2M of potassium phosphate buffer. The enzyme was generally stable below $50^{\circ}C$, but inactivated completely at $70^{\circ}C$. 1mM of $Fe^{3+},\;K^+\;and\;Na^+$ increased the enzyme activity, but 0.01mM of $Co^{2+},\;Cu^{2+},\;Ni^{2+},\;Hg^{2+},\;Ag^{2+},\;Zn^{2+},\;Ba^{2+},\;and\;Mg^{2+}$ markedly inactivated the enzyme activity. 0.1mM of p-chloromercuribenzoate, trichloroacetic acid, and N-ethylmaleimide compleyely inhibited the enzyme activity, but 0.1mM of 2-mercaptoethanol slightly increased the enzyme activity. 20-80%로 황산암모늄 분획한 효소액을 사용하여 Arthrobacter sp. JH-13균주가 생산하는 세포외 cytosme deaminase의 성질을 검토하였다. 검토한 기질 중, 본 효소는 cytosine과 5- f1uorocytosine을 기질로 이용하였으며, 효소의 활성에 대한 최적 pH는 8.0부근이었고, 최적 온도는 $40^{\circ}C$부근으로 나타났다. 본 효소는 0.2M의 p potassium phosphate 완충액 (pH 8.0) 보다 0.2M의 tris-HCl 완충액 (pH 8. 0) 에서 더욱 안정하였다. 온도에 대한 안정성을 검토한 결과. $50^{\circ}C$ 부근까지는 대체로 안정하였으나, $70^{\circ}C$에서는 완전히 활성을 잃었다. 또한 ImM의 $Fe^{3+},\;K^+\;Na^+$ 이온은 효소의 활성을 증가시켰으나 0.01mM의 $Co^{2+},\;Cu^{2+},\;Ni^{2+},\;Hg^{2+},\;Ag^{2+},\;Zn^{2+},\;Ba^{2+},\;Mg^{2+}$ 이온들은 효소의 활성올 강력하게 저해하였다. O.lmM의 p-mercuribenzoate, trichloroacetic acid, N-ethylmaleim mide등은 효소의 활성을 완전히 저해하였으며. O.lmM의 2-mercaptoethanol은 효소의 활성올 약간 증가시키는 것으로 나타났다.
