수종의 식물수출물의 항산화 및 Melanin 합성 억제효과

본 연구에서는 식물 추출물을 이용하여 항산화 활성 및 tyrosinase 활성 억제 효과를 측정하였다. 식물 추출물의 폴리페놀물질의 총 함량은 Acer psedo-siebolianum의 추출물이 16.4 mg/g로 가장 높은 추출량을 나타내었다. 항산화활성 측정에서는 Acer ginnala 에서 IC50값으로 21.3 μg/mL으로 가장 좋은 활성을 나타내었다. 반면에 L-DOPA를 기질로 하여 mushroom tyrosinase의 활성 억제측정에서는 Distylum racemosum 1,000 mg에서 49.1%로 다른 추출물에 비하여 상대적으로 높은 활성을 나타내었다. Tyrosinase 활성 억제력이 가장 높은 D. racemosum의 추출물을 이용하여 ethanol 분획과 ethyl acetate 분획으로 분리하여, 이중 D. racemosum의 ethanol 분획에서 항산화 활성 IC50 값은 0.9 μg/mL, tyrosinase 활성억제는 IC50값이 118.1 μg/mL로 ethyl actate 분획보다 높은 활성을 나타내었다. 또한 ethanol 분획을 이용하여 B16/F1 melanoma cell에서는 60 μg/mL까지는 세포독성을 나타내지 않았으며 80 μg/mL의 농도에서 약간의 세포독성을 나타내었다. 에탄올 분획을 이용한 세포내 melanin 색소의 생산억제 IC50값은 75.4μg/mL로 분석되었다. 이러한 결과로 D. racemosum의 에탄올 추출물이 B16/F1 melanoma cell세포의 melanin색소합성대사에 관여하여 색소합성을 저해하는 것으로 보인다. Plants extracts are good resources to find functional compounds for human health. The following eight plants were collected and total phenolic contents were determined. Acer psedo-siebolianum showed the highest phenolic contents, 16.4 mg/g, whereas Cercidiphyllum japonica showed the lowest contents, 1.9 mg/g. The DPPH free radical scavenging capacities of the plant extracts showed high activity in following order : Acer ginnala (21.3 μg/mL) > Cornus walteri (23.9 μg/mL) > Distylum racemosum (29.2 μg/mL) > Castanopsis cuspidata var. Thunbergii (31.7 μg/mL) > Acer psedo-siebolianum (34.6 μg/mL) > Thuijopsis dolabrata cv. Aurea (53.1 μg/mL) > Cercidiphyllum Japonica (115.2 μg/mL). Also the mushroom tyrosinase inhibitory activities of total extracts were determined at different concentration. D. racemosum extract showed highest (49.1% at 1,000 mg) in inhibitory activity than other seven extracts. The ethanol fraction (IC50 value: 118.1 μg/mL) from D. racemosum showed more inhibitory activity than ethyl acetate fraction (IC50 value: 203 μg/mL). The ethanol fraction on showed no significant cytotoxicity in B16/F1 cells line up to 60 μg/mL. Over 80 μg/mL of ethanol fraction showed cytotoxicity in B16/F1 cells. The melanin contents of cells were significantly attenuated by ethanol fraction in a dose-dependent manner. The IC50 value of ethanol fraction was 75.4 μg/mL.

본 연구는 은행 열매 오일의 멜라닌 생성 억제 효과를 확인한 것이다. 은행나무 열매 오일은 DPPH assay와 FRAP assay를 사용하여 라디컬 소거능을 시험하였다. 결과적으로 은행나무 열매 오일은 DMSO를 용매로 0.06% 녹였을 때, DPPH assay에서 9.96% 소거활성을 나타내었고 FRAP는 1.33 mM의 ferric sulfate (FeSO<sub>4</sub>)를 생성하였다. 은행 열매 오일은 tyrosinase inhibition assay에서 37.72%의 억제력을 가졌고 B16/F10 세포에 멜라닌 생합성 실험을 통해 확인하였다. 은행 오일 0.06%에서 α-MSH 처리 구에 비해 48.02%의 멜라닌 생성을 억제하였다. Tyrosinase, tyrosinase related protein-1 (TRP-1), tyrosinase related protein-2 (TRP-2), microphthalmia-associated transcription factor (MITF)의 유전자 발현 수준은 control군에 비해 0.04%와 0.06% 농도 군이 크게 감소하였다. 결과적으로 은행 열매 오일 추출물이 멜라닌 생성을 억제하는 효과가 있는 것으로 확인되었다. In this study, we investigated the inhibitory effect on melanin synthesis of Ginkgo biloba seed oil. The results showed 9.96% inhibitory effect scavenging activity on DPPH and showed a value of 1.33 mM of FeSO<sub>4</sub> at a concentration of 0.06% in DMSO by using FRAP assay. G. biloba seed oil inhibited tyrosinase activity up tp 37.72% and suppressed the biosynthesis melanin up to 48.02% at 0.06% in B16/F10 mouse melanoma cell. In G. biloba seed oil treated group tyrosinase, TRP-1, TRP-2 and MITF gen expression levels significantly decreased compared to the contral group at a concentration of 0.04% and 0.06%. In conclusion, these results indicated that G. biloba seed oil extract have a good antimelanogenetic effects.

A nuclear fuel test loop (after below, FTL) is installed in the IR1 of an irradiation hole in NANARO far testing the neutron irradiation characteristics and thermo hydraulic characteristics of a fuel loaded in a light water power reactor or a heavy water power reactor. There is an in-pile section (IPS) and an out-pile section (OPS) in this test loop. When HANARO is operated normally, the fuel loaded into the IPS has a nuclear reaction heat generated by a neutron irradiation, To remove the generated heat and to maintain the operation conditions of the test fuel, a main cooling water system (MCWS) is installed in the OPS of the FTL. The MCWS is composed of a main cooler, a pressurizer, two circulation pumps. a main heater, an interconnection pipe line and instruments. The interconnection pipeline is a closed loop which is connected to an inlet and an outlet of the IPS respectively. The MCWS is under a cold function test during a start-up period. This paper describes the system flow network analysis results of the flow control of a main cooling water system in the HANARO fuel test loop. It was confirmed through the results that the flow was met the system design requirements.

HANARO, an open-tank-in-pool type research reactor of 30 MWth power in Korea, has been operating normally since its initial criticality in February, 1995. For the last ten years, HANARO has verified the mechanical and structural integrities of the pressure retaining components of the safety related systems and the integral attachments of the supports and restraints of the components within a specified boundary through ten years periodic in-service inspections in accordance with Article IWD in ASME SEC. XI and Notice 2004-13 (Jul. 2nd, 2004) of the minister of Korea Ministry of Science and Technology. From the results, it was confirmed through the ISI that the pressure retaining components and parts were stable to within the specified boundaries for their mechanical and structural integrities.

When HANARO is normally operated, the fuel has a nuclear reaction heat generated by a neutron irradiation. To remove the generated heat and to maintain an operation condition of the test fuel, a main cooling water system (MCWS) is installed in the FTL. This paper describes the performance characteristics of the main coolant pump. It was confirmed through the pump performance test that the performance was met the design requirement. But the pump operation point is about 96% of the pump shutoff head and the pump efficiency is very low. For an improvement of the pump efficiency, the pump impeller is selected to be met the pump specific velocity.

HANARO, an open-tank-in-pool type research reactor of 30 ㎿th power in Korea, which is different from a power plant reactor, exhausts a fission heat generated from the reactor core into the atmosphere through a secondary cooling tower instead of an electric power production from the heat. During every one month inspection of the cooling tower, a No. 4 cooling fan gear reducer of the cooling tower recoded a high unstable vibration above the limit. To find the reason, a frequency analysis was conducted. It was conformed through the results of the frequency analysis that the frequency of the high vibration was 354 ㎐ which was calculated two times rotation of the pinion gear of the gear reducer. There was a broken of the gear surface of the pinion gear transferred the rotation power. After the repair of the pinion gear, the reducer was operated normally.
