        Kinetic Studies of Aspartase from Hafnia alvei byTemperature Dependence Activity Changes

        The temperature dependence of the kinetic parameters of the aspartase-catalyzed reaction has been examined in the direction of deamination. The pK1values at 37$^{\circ}C$, 25$^{\circ}C$, 16$^{\circ}C$ and 7$^{\circ}C$ were 6.2 $\pm$ 0.1, 6.3 $\pm$ 0.1, $6.7{\pm}0.3$ and 6.9 $\pm$ 0.3, respectively. On the other hand, the pK2 values at 37$^{\circ}C$,25$^{\circ}C$, 16$^{\circ}C$ and 7$^{\circ}C$ were 8.1 $\pm0.2$, 8.3 $\pm$ 0.2, 8.2 $\pm$ 0.3 and 8.0 $\pm$ 0.2,respectively. The enthalpy of ionization, DHion, calculated from the slope of pK1, are 6.0 $\pm$ 0.3 kcal/mol. These results validate the prediction that aspartase requires a histidine residue for a general base, and a cysteine (or having a carboxyl functional group) for a general acid.

        항공기 진동 범위 충족을 위한 레이다센서 장착구조물의 구조적 안정성 검증

        In this paper, the structural stability of a radar sensor mounting structure satisfying aircraft vibration range is described. Vibration measurement of mounting structure and radar sensor for aircraft vibration was performed for the mounting structure for flight test of radar sensor. In order to design the mounting structure satisfying the structural stability, it was designed by referring to the propeller aircraft vibration standard of MIL-STD-810G. To verify the structural stability of the constructed structure, a dummy radar sensor was mounted on the aircraft using the designed and constructed mounting structure, and the amount of vibration applied to the mounting structure was measured using an acceleration sensor. Structural stability of the mounting structure was confirmed by using the vibration data measured in the dynamic operating environment such as takeoff, cruising, and landing. 본 논문에서는 항공기 진동 범위 충족을 위한 레이다센서 장착구조물의 구조적 안정성을 검증하는 내용을 기술한다. 레이다센서의항공기 탑재 비행시험을 위하여 설계 및 제작된 장착구조물을 대상으로 항공기 진동에 대한 장착구조물 및 레이다센서의 진동계측을실시하였다. 구조적 안정성을 만족하는 장착구조물을 설계하기 위해서 MIL-STD-810G의 프로펠러 항공기 진동규격을 반영한구조해석을 통해 설계하였다. 제작된 장착구조물의 구조적 안정성을 확인하기 위하여 항공기에 더미 레이다센서를 설계 제작된장착구조물로 장착하고 가속도센서를 이용하여 장착구조물에 인가되는 진동량을 계측하였다. 이륙, 순항, 착륙 등 동적 운용환경에서레이다센서의 진동데이터를 계측하고 분석함으로써 비행운용환경에서 장착구조물의 구조적 안정성이 확보되었음을 확인하였다.

      • NBS에 의한 아스파타제 효소의 화학 변형 연구

        아세토니트릴 용매 내에서 p-니트로염화벤조일과 o-클로로아닐린과의 반응을 온도 (15, 20, 25℃)와 압력변화(1, 200, 500, 1000bar)에 따라 반응속도론적으로 연구하였다. 반응속도상수로부터 활성화파라미터 (ΔV≠, Δβ≠, ΔH≠, ΔS≠, ΔG≠, Ea)를 구하였다. 온도가 증가함에 따라 속도상수는 증가하고, 이때 ΔV≠, Δβ≠및 ΔS≠는 모두 음의값을 나타내었으며, 압력변화에 따른 속도론적 연구는 전반적인 반응을 associative SN2반응으로 진행됨을 알 수 있다. Aspartase from Hafnia alvei was inactivated by N-bromosuccinimide(NBS) following pseudo-first order kinetics. The apparent reaction orders were 2.9 ×10-3min-1㎛-1, indicating that inactivation was due to a indole group in the active site of aspartase and participation of the indole group in the essential step in the catalytic reaction. Spectrophoto-metric studies indicated that about one oxyindole group per molecule of enzyme was formed following complete loss of enzymatic activity.

        Effect of Millimeter Wave for Differentiation of Bone Marrow Stromal Cells to Osteoblasts-like cell

        In this study, we have evaluated the effects of millimeter wave by assessing the osteogenic differentiationproperties of bone marrow mesenchymal stem cells (BM-MSCs). To measure the effect of millimeter wave, milli-meter wave irradiation was applied to these cells continuously for 7 days and Dulbecco's Modified Eagle's Medium(DMEM)/F12 was used with a supplement. The morphology did not show cytotoxicity, while the viability of themillimeter wave treatment group cells was similar to that of the control group. Also, the level of collagen type I,vimentin, osteoprotegerin, osteopontin, bone morphogenetic protein (BMP)-2, and osteocalcin mRNA wereincreased and the level of phospho extracellular signal-regulated kinase (pERK), osteonectin, and BMP-2 expres-sion were increased in the millimeter wave treatment group. Furthermore, fluorescence activated cell sorter (FACS)analysis showed that CD 90 and CD 105 expression decreased in the millimeter wave group and immunocytochem-ical staining analysis showed that BMP-2 and osteopontin expression had increased in the millimeter wave group. In conclusion, our results suggest that an appropriate millimeter wave exposure can be promoted as a potent positivemodulator of bone marrow stromal cells into osteoblasts-like cells.

      • 산소전달계수 및 기질공급방법이 Baker's Yeast의 유가식 배양에 미치는 영향

        Baker's yeast의 생산공정에서 가장 중요한 문제가 되는 것이 산소를 충분히 공급하여 주는 것과 최적의 기질공급방법, 기질공급량을 선정하는 것이라 할 수 있겠다. 본 연구에서는 기질공급방법을 변화시켜 가면서 유가배양실험을 실시하여 생산성과 최종 세포수율을 최적으로 나타내는 기질공급방법과 기질공급량을 검토하여 보았고, 교반속도를 변화시켜 가면서 유가식 배양 실험과 산소전달계수 측정실험을 통하여 유가식 배양에서 산소전달계수가 baker's yeast의 생산에 미치는 영향을 분석하여 보았다. 기질공급방법은 sigmoidal한 것이 가장 최적으로 나타났고, 최적의 기질공급량은 산소전달계수가 변함에 따라 상응하여 변하였으며, baker's yeast의 생산은 산소전달계수가 변함에 따라 크게 영향을 받았다. In Baker's yeast production process the most important problems are that oxygen must be sufficiently supplied and that substrate feeding policy and the determination of total fed-sugar must be optimized. In this study substrate feeding policy and optimal total fed-sugar demonstrating the maximum productivity and yield were investigated in fed-batch fermentation at various substrate feeding modes. The effects of oxygen transfer coefficient on baker's yeast production were investigated by varying agitation speed and carrying out the separate oxygen transfer experiments. The sigmoidal substrate feeding policy appeared the optimum method to reach the highest cell yield. Optimum total fed-sugar have varied according to oxygen transfer coefficeent. In general, the production of baker's yeast was observed to be largely dependent on oxygen transfer coefficient in the fermenter.

        Cloning and Characterization of UDP-glucose Dehydrogenase from Sphingomonas chungbukensis DJ77

        Sphingomonas chungbukensis DJ77 has the ability to produce large quantities of an extracellular polysaccharide that can be used as a gelling agent in the food and pharmaceutical industries. We identified, cloned and expressed the UDP-glucose dehydrogenase gene of S. chungbukensis DJ77, and characterized the resulting protein. The purified UDP-glucose dehydrogenase (UGDH), which catalyzes the reversible conversion of UDP-glucose to UDPglucuronic acid, formed a homodimer and the mass of the monomer was estimated to be 46 kDa. Kinetic analysis at the optimal pH of 8.5 indicated that the Km and Vmax for UDP-glucose were 0.18 mM and 1.59 mM/min/mg,respectively. Inhibition assays showed that UDP-glucuronic acid strongly inhibits UGDH. Site-directed mutagenesis was performed on Gly9, Gly12 Thr127, Cys264, and Lys267. Substitutions of Cys264 with Ala and of Lys267 with Asp resulted in complete loss of enzymatic activity, suggesting that Cys264 and Lys267 are essential for the catalytic activity of UGDH.
