당뇨 백서에서 p38 Mitogen-Activated Protein Kinase 억제제 (FR167653)가 세극막 관련 단백의 발현에 미치는 영향

목적: 본 연구에서는 실험적 당뇨 백서에서 p38 MAPK 억제재가 단백뇨 발생 및 세극막 관련 단백의 발현 변화에 미치는 영향을 알아보고자 하였다. 방법: Sprague-Dawley 백서 32마리를 대상으로, 16마리는 streptozotocin (65 mg/kg)으로 당뇨를 유발시켰으며 (DM군), 16마리는 위약을 투여하였다 (C군). 각 군에서 8마리는 p38 MAPK 억제제인 FR167653을 5 mg/kg의 용량으로 6주간 매일 근주하였다 (C+FR,DM+FR). 당뇨 유발 6주 후 24시간 소변을 수집한 다음 희생시켜 신장을 적출한 후 사구체를 분리하였다. 24시간 뇨알부민 배설량은 ELISA로 측정하였으며, 사구체 내 nephrin, P-cadherin, 그리고 ZO-1의 mRNA와 단백 발현은 각각 real-time PCR과 Western blot으로 분석하였다. 결과: 당뇨 유발 6주 후 24시간 뇨알부민 배설량은 C군 (0.29±0.04 mg/day)파 C+FR군 (0.32±0.04 mg/day)에 비하여 DM군 (1.09±0.17 mg/day, p<0.05)에서 유의하게 증가되었으며, 이러한 증가는 FR167653 투여로 의미있게 억제되었다 (0.38±0.05 mg/day, p<0.05). 세극막 관련 단백 중 nephrin mRNA 및 단백 발현은 DM군에서 C군에 비하여 각각 1.9배, 1.6배 증가되었으며, 이러한 증가는 FR167653 투여로 각각 74%, 89% 억제되었다 (p<0.05). 반면에, FR167653은 DM군에서 발현이 감소된 P-cadherin과 발현이 증가된 ZO-1에 의의있는 영향을 미치지 않았다. 결론: 이상의 결과로, FR167653은 당뇨 백서에서 p38 MAPK 활성화를 효과적으로 억제하였고, 뇨알부민 배설을 유의하게 감소시켰으며, 이러한 효과는 nephrin의 발현 변화의 호전과 관련이 있을 것으로 생각된다. Purpose: This study was undertaken to investigate the effect of a p38 mitogen-activated protein kinase (p38 MAPK) inhibitor, FR167653, on urinary albumin excretion and on the expression of slit diaphragm-associated proteins in diabetic rats. Methods: Thirty-two Sprague-Dawley rats were injected with diluent [control (C), N=16] or streptozotocin intraperitoneally (DM, N=16). Eight rats from each group were treated with 5 mg/kg/day FR167653 (C+FR, DM+FR) for 6 weeks. At the time of sacrifice, 24-hour urinary albumin excretion was determined by ELISA. Glomerular nephrin, P-cadherin, and ZO-1 mRNA and protein expression were determined by real-time PCR and Western blot, respectively, with sieved glomeruli. Results: Urinary albumin excretion was significantly higher in DM compared to C rats, and this increase in albuminuria was significantly inhibited by the administration of FR167653 in DM rats. Glomerular phospho-p38 MAPK protein expression was significantly increased in DM rats compared to C rats, and FR167653 treatment significantly attenuated the increase in phospho-p38 MAPK expression in DM glomeruli. Nephrin mRNA and protein expression were higher in 6-week DM compared to C glomeruli, and these increases were significantly abrogated with FR167653 treatment in DM rats. In contrast, FR167653 had no effects on the decrease in P-cadherin expression and the increase in ZO-1 expression observed in DM glomeruli.

Purpose: This study was undertaken to elucidate whether CC chemokine receptor 2 (CCR2) exists in human peritoneal mesothelial cells (HPMCs) and whether monocyte chemoattractant protein-1 (MCP-1) has direct effects on epithelial to mesenchymal transition (EMT) and fibronectin expression in HPMCs. Methods: HPMCs were isolated from a piece of human omentum and were incubated with M199 media containing 5.6 mM glucose (LG), 5.6 mM glucose+94.4 mM mannitol (LG+M), LG+10 ng/mL recombinant human MCP-1 (LG+MCP-1), or 100 mM glucose (HG) with or without a specific inhibitor of CCR2, 1.0 μM RS102895, for 4 days. Levels of secreted MCP-1 in culture media were determined by ELISA. Western blot was performed to determine fibronectin, E-cadherin, α-smooth muscle actin (α-SMA) and CCR2 protein expression. Results: MCP-1 protein levels were significantly increased in HG-conditioned media compared to LG media (p<0.05). CCR2 protein was expressed in HPMCs, but there was no difference between LG-and HG-stimulated cells. α-SMA protein expressions in HG and LG+MCP-1 groups were significantly higher relative to LG cells, while E-cadherin protein expressions were decreased in HG and LG+MCP-1 groups compared to LG cells (p<0.05). In addition, there were significant increases in fibronectin mRNA and protein expression in HG and LG+MCP-1 groups (p<0.05). These HG-induced changes were significantly abrogated upon pre-treatment with RS102895. Conclusion: HG and MCP-1 directly induce EMT and enhance fibronectin expression in HPMCs, and these HG-induced changes were attenuated by the inhibition of MCP-1/CCR2 system, suggesting that increased MCP-1 levels by HG may contribute to the development of peritoneal fibrosis.

The purpose of this study was to observe the effect of eating a raw egg by professional or nonprofessional voice users on their voice quality and the duration of the effect. 20 professional voice users and 20 nonprofessional voice users participated in the experiment and they had gone through stroboscopy to have no vocal or laryngeal diseases. The voice exam was performed three times: before eating a raw egg (1st period), right after eating it (2nd period), and 10 minutes later (3rd period). By using Multi-dimensional Voice Program which is a software of Computerized Speech Lab 4500 as a voice analysis instrument, the authors checked the F0, Jitter, Shimmer, Noise to harmonic ratio (NHR), and Voice Range Profile (VRP). Results showed as follows: Firstly, vocal hygiene was good in 57.5% of the total subjects and was poor in 42.5%. 40% of professional voice users and 75% of nonprofessional voice users hand good quality. 77.5% of the total subjects had the vocal fatigue while 22.5% of the subjects did not. 95% of the professional voice users and 60% of nonprofessional voice users complained the vocal fatigue. 60% of the total subjects reported a subjective vocal symptom. 65.0% professional voice users and 70.0% of nonprofessional voice users reported a voice symptom. From the results above, we suggest that eating a raw egg may lead to imporve voice quality of the professional voice users.
손실 비용 및 ㎿+㎹Ar-miles 배전 요금제를 고려한 분산전원의 위치 결정

이 논문은 ㎿-㎹Ar mile 법에 의한 요금제를 배전계통에 대하여 적용하며, 배전손실에 의한 비용과의 비교를 목표로 하였다. 특히 분산전원이 배전계통에 투입되는 경우에 있어서 투입되는 부하점에 따라 배전 손실 내지 배전요금이 변할 수 있음을 확인하였고, 배전손실이 최소가 되는 지점과 배전요금이 최저가 되는 지점이 반드시 일치하지는 않음을 알 수 있었다. 8모선의 배전계통에서 시뮬레이션이 이루어졌으며, 각 배전선로 별로 임피던스, 길이, 용량, 연간 필요 수입액등을 정하였다. 논의의 간소화를 위하여 분산전원은 상시 정격 출력하는 것으로 가정하였다.
