배추 종자 및 유묘에 대하여 GUS발현 또는 hepatitis B surface antigen (HBsAg)발현 벡터를 지니는 Agrobacterium tumefaciens LBA4404 세포를 이용하여 진공침윤(agroinfiltration)에 의한 형질전환을 시도하였다. 특히 ELISA를 이용한 HBsAg발현의 정량적 분석에서 agroinfiltration방법은 형질전환효율이 매우 저조하게 나타났다. 그러나 차아염소산나트륨 용액을 발아 전 또는 발아 중인 배추종자에 처리한 후 agroinfiltration을 실시한 경우 형질전환 효율이 $2\~5$배 증가하였다. 따라서 차아염소산나트륨 등의 화학연마제에 의한 종자의 상처발생이 Agrobacterium의 감염을 용이하게 함으로써 배추유묘에서의 일시유전자발현을 증대시키는 것으로 제안되고있다. Chinese cabbage (Brassica campestris ssp. napus var. pekinensis Makino) seeds/seedlings were transformed via vacuum-infiltration with recombinant Agrobacterium tumefaciens LBA4404 cells. The agroinfiltration method was determined to be unsuccessful for Chinese cabbage transformation during the analysis of hepatitis B surface antigen expression by ELISA. However, treatment of sodium hypochlorite solution, prior to agroinfiltration, to pregerminated or germinating 1 day- or 2 days-old seeds was proven effectively to enhance transformation efficiency, suggesting that chemical wounding caused by sodium hypochlorite reaction might facilitate Agrobacterium infection and, therefore, transient gene expression in Chinese cabbage sprouts.
다채 (Brassica campestris var. narinosa) 유묘의 형질전환 및 일시발현의 정량적 분석

Brassica campestris var. narinosa는 새싹채소로 애용되는 식물로서 간단한 용기에서 손쉽게 재배할 수 있다. 본 연구에서는 vacuum-infiltration을 통한 Agrobacterium-mediated transformation에 의해서 B. campestris var. narinosa 싹에서의 GUS일시발현을 성공적으로 확인하였다. 발아전의 종자가 발아를 이미 시작한 종자보다 그 형질전환 효율이 높게 나타났다. 한편, hydrogen peroxide를 2일 생장시킨 싹에 처리한 후 형질전환 하였을 때 GUS발현이 증가되는 것을 관찰하였다. 간염항원유전자의 형질전환과 ELISA에 의한 항원단백질의 발현량 분석 시 hydrogen peroxide 처리 싹이 비처리 형질전환 싹에서 보다 2배 이상의 발현량이 측정되었다. Tah tasai chinese cabbage (Brassica campestris var. narinosa), a vegetable plant popularly consumed as several-days-old seedlings in oriental countries, can be easily cultivated using a simple appliance. We demonstrated that Agrobacterium-mediated transformation via vacuum infiltration (agroinfiltration) resulted in a successful transient GUS gene expression in tah tasai chinese cabbage seedlings. Pre-germinated seeds were found to be more susceptible to Agrobacterium infection than one-day-old or two-days-old seedlings. We also demonstrated that hydrogen peroxide (HPO) treatment increased GUS expression especially for two-days-old seedlings. In ELISA using seedlings transformed with hepatitis B surface antigen (HBsAg) DNA by agroinfiltration, HBsAg protein synthesis increased more than two folds by HPO treatment to two-days-old seedlings in comparison to the mock-treated pre-germinated seeds.

The purpose of this study is to investigate the curricula of TESOL programs in Korea. Using program information on the Internet, this study also presents the results of empirical study of the TESOL courses. The sample consisted of 15 TESOL programs in Korea. It was found that the programs have not yet recognized and addressed the different needs of non-native speakers of English teachers. It is suggested that the curricula of TESOL programs in Korea needs to understand non-native English teachers as reflective professionals, agents of change in local contexts, and coaches for cross-culture communication.

N/A Pyrophosphoric modified polyesters (TATBs) were synthesized by polycondensation of adipic acid, trimethylopropane, 1.4-butanediol and tetramethyiene bis(orthophosphate). Two-component PU flame-retardant coatings (TATBCs) were prepared by blending TATBs with HDI-Biuret. Most of the physical properties of the flame-retardant coatings were comparable to those of non-flame-retardant coatings. Coatings containing 10 and 15wt% 1.4-butanediol. TATBC-10C and TATBC-15C were not flammable in the vertical flame-retardancy test.
단보 : NaOH처리에 의한 Agrobacterium이용 팽이균사체 형질전환

Agrobacterium harboring vector pCHBs with hygromycin phosphotransferase(hph) and hepatitis B virus surface antigen(HBsAg)gene was transformed into the mycelial culture of Flammulina velutipes. In particular, mild NaOH solution was treated to the mycelia before Agrobacterium infection step. This was purposed to generate putative surface wounds in the mycelial cell walls. The results showed that hygromycin-resistant(hygr) mycelia could be obtained only from NaOH-treated mycelia but not from intact mycelia. The integration of hygr gene in fungal genome was confirmed by PCR. In addition, a single transgene integration and heterologous protein expression in F. velutipes could be verified by Southern blot hybridization and western blot analysis, respectively. This study demonstrated an efficient tool for the Agrobacterium-mediated transformation of F. velutipes mycelia.
언어학적 전환, 비판적 언어학 전통, 그리고 비판적 담론연구의 출현

본 연구의 목적은 비판의식이 보태진 담론연구방법의 출현 배경과 지적 토대를 탐색하고 질적연구 분야에 어떻게 기여하고 어떤 한계점을 갖고 있는지 살펴보는 것이다. 비판적 담론연구를 위해 실증적 자료를 기획하고 수집하고 분석하는 연구방법 역시 학자군마다 다소 차이가 크기 때문에 본 연구는 알튀세(Althusser)의 이데올로기, 그람시(Gramsci)의 헤게모니. 푸코(Foucault)의 권력/담론/지식, 홀(Hall)의 문화연구로부터 구축된 구조주의와 언어학적 전환(linguistic turn)의 지식전통과 바흐친(Bakhtin), 폐슈(Pêcheux), 할러데이(Halliday)를 포함한 비판적 언어학(critical linguistics) 연구전통으로부터 페어클러프(Fairclough)의 담론연구 모형이 어떻게 출현할 수 있었는지 우선적으로 탐색하기로 한다. 페어클러프의 모형은 할러데이의 체계기능문법(systemic functional grammar)으로부터 차용된 텍스트 분석방법, 바흐친의 얽힌 텍스트의 해석, 사회구조 분석에 용이한 푸코의 고고학/계보학 이론, 그람시의 헤게모니적 투쟁논점을 모두 결합시키면서 미시적 분석과 거시적 분석의 상호 간 단점을 보완했다. 본 연구는 비판적 담론분석의 연구방법적 의미와 함께 한계점 역시 부연하고 추후 연구자들이 숙지해야 할 점을 정리했다. The purpose of this study is to provide theoretical and methodological resources to understand how CDS (Critical Discourse Studies) has been emerged as a research methodology. In this review of literature, the theoretical foundations will first be discussed on the tradition of linguistic turn and structuralistic movement, including Althusser’s ideology, Foucault’s power and discourse, Gramsci’s hegemony, Hall’s cultural study, as well as the tradition of critical linguistics including Halliday’s systematic functional grammar, and Bakhtin’s intertextuality. CDS clearly intends to incorporate social theories into discourse analysis and advocates social commitment and interventionism in researches. The main features and domains of CDS are discussed in this paper, with emphasis on the theory formation (how texts and discourses work in ideological interests with powerful political consequences) by one of CDS’s most prominent scholars, Norman Fairclough. Th e strengths as well as the limitations of the Fairclough’s model-based research are also discussed. In the conclusion, the article makes some suggestions for future directions of CDS methodological development.
