Anti-allergic Effect of Low Molecular Weight β-Glucan Prepared by γ-Irradiation

This study evaluated the anti-allergic effect of low molecular weight β-glucan (25 kDa) degraded by γ-irradiation. Allergy was induced in mouse models administrated with γ-irradiated low molecular weight and non-irradiated high molecular weight β-glucan, respectively,for 7 days. However, only the mice treated with low molecular weight β-glucan had a significantly decreased the total serum IgE and ovalbumin specific-IgE levels, and significantly increased Th 1 cytokine expression compared to only allergy induced mice. The allergic responses could be reduced by low molecular weight β-glucan treated γ-irradiation through the increased expression of Th 1response and decreased expression of total IgE and antigen specific IgE.
Scutellarein Reduces Inflammatory Responses by Inhibiting Src Kinase Activity

Flavonoids are plant pigments that have been demonstrated to exert various pharmacological effects including anti-cancer, anti-diabetic, anti-atherosclerotic, anti-bacterial, and anti-inflammatory activities. However, the molecular mechanisms in terms of exact target proteins of flavonoids are not fully elucidated yet. In this study, we aimed to evaluate the anti-inflammatory mechanism of scutellarein (SCT), a flavonoid isolated from Erigeron breviscapus, Clerodendrum phlomidis and Oroxylum indicum Vent that have been traditionally used to treat various inflammatory diseases in China and Brazil. For this purpose, a nitric oxide (NO) assay, polymerase chain reaction (PCR), nuclear fractionation, immunoblot analysis, a kinase assay, and an overexpression strategy were employed. Scutellarein significantly inhibited NO production in a dose-dependent manner and reduced the mRNA expression levels of inducible NO synthase (iNOS) and tumor necrosis factor (TNF)-α in lipopolysaccharide (LPS)-activated RAW264.7 cells. In addition, SCT also dampened nuclear factor (NF)-κB-driven expression of a luciferase reporter gene upon transfection of a TIR-domain-containing adapter-inducing interferon-β (TRIF) construct into Human embryonic kidney 293 (HEK 293) cells; similarly, NF-κ B nuclear translocation was inhibited by SCT. Moreover, the phosphorylation levels of various upstream signaling enzymes involved in NF-κB activation were decreased by SCT treatment in LPS-treated RAW264.7 cells. Finally, SCT strongly inhibited Src kinase activity and also inhibited the autophosphorylation of overexpressed Src. Therefore, our data suggest that SCT can block the inflammatory response by directly inhibiting Src kinase activity linked to NF-κB activation.

This study was investigated the effect of tumor cell cytotoxicity of gamma irradiatedChaga mushroom extract (CME). CME was prepared by hot water extraction at 70 C for 4 hoursand lyophilized. Ten mg ml-1 of lyophilized CME powder was dissolved with deionized water andthen irradiated at the doses of 10, 50, 100, and 150 kGy by cobalt 60 gamma irradiator. The gammairradiatedand non-irradiated CME were treated into the cancer cell, including human stomachcancer and human colon cancer. Cytotoxicity against the cancer cell was increased in gammairradiatedCME and antioxidant activity was also increased in gamma-irradiated CME, as irradiationdose increased. Therefore, it was considered that gamma irradiation was effective method forimprovement of the cancer cell cytotoxicity and antioxidant activity of Chaga mushroom extract.
Low Molecular Weight β-Glucan Stimulates Doxorubicin-induced Suppression of Immune Functions in Mice

The aim of this study was to evaluate the protective effect of low molecular weight β-glucan (LMG)against doxorubicin (DOX)-induced immune suppression of tumor-bearing mice. The tumor size and spleen cell functions such as spleen cell proliferation, cytokine production (interferon-γ and interleukin-2), and the population of CD4+ and CD8+ T cells were estimated. In the tumorbearing mice, the tumor size was significantly (p<0.05)decreased by DOX treatment. However, there was no significant difference between mice treated with high molecular weight β-glucan (HMG) and mice treated with LMG. Spleen cell proliferation and cytokine production were significantly (p<0.05) decreased in only DOX treated group, but increased in all β-glucan treated groups with DOX. Moreover, the populations of CD4+ and CD8+ T cells were also increased in the LMG-treated group. It appears that LMG effectively reduces the DOX-induced immune toxicity through activation of immune cells such as splenocytes.

This study investigated the effect of gamma irradiation on immune enhancing activityof Chaga mushroom extract (CME). CME was prepared by hot water extraction at 70 C for 4 hoursand lyophilized. Lyophilized CME powder was dissolved with deionized water at 10 mg ml-1 andthen irradiated at the doses of 10, 30 and 50 kGy by cobalt 60 gamma irradiator. The gammairradiatedand non-irradiated CME were treated into the splenocyte separated from mouse. Cellproliferation and cytokine production of the immune cells were increased by gamma-irradiatedCME and these increases were more prominent when CME was irradiated at higher doses. Therefore,it is considered that gamma irradiation can be an effective method for improvement of theimmunomodulating activity Chaga mushroom extract.
