Solid Phase Microextraction 및 Purge & Trap을 이용한 생물시료 중 휘발성 유기화합물의 GC/MS 분석비교

생물시료에 존재하는 휘발성 유기화합물 중 n-butylbenzene과 1,2-dibromo-3-chloropropane (DBCP)를 기체크로마토그래피/질량분석기-선택이온검색법에 의해 수행하였다. 시료 중 휘발성 유기화합물은 $100{\mu}m$ polydimethyl siloxane (PDMS) fiber를 사용하여 headspace solid phase microextractio (SPME) 및 purge & trap 방법에 의해 추출 및 비교하였다. SPME에 의한 회수율은 n-butylbenzene의 경우 85.8%, DBCP의 경우 92.4%로 나타났고 검출한계는 각각 $0.15{\mu}g/kg$, $0.05{\mu}g/kg$로 나타났다. 반면, purge & trap의 경우 회수율은 n-butylbenzene의 경우 115.2%, DBCP의 경우는 80.9%로 나타났고 검출한계는 각각 $0.04{\mu}g/kg$ 및 $0.70{\mu}g/kg$로 나타나 두 방법에 있어 국내에서 규제하는 검출한계측면에서 큰 차이는 없었다. The analysis of n-butylbenzene and 1,2-dibromo-3-chloropropane (DBCP) as volatile organic compounds in biota samples was performed by gas chromatography/mass spectrometry-selected ion monitoring mode. The target compounds, n-butylbenzene and DBCP, in biota samples were extracted by headspace solid phase microextraction (SPME) with $100{\mu}m$ polydimethyl siloxane (PDMS) fiber and purge & trap method. The extraction recoveries of these compounds obtained by SPME was 85.8% for n-butylbenzene and 92.4% for DBCP, respectively. Each value of method detection limit were $0.15{\mu}g/kg$ and $0.05{\mu}g/kg$, respectively. While in the case of purge & trap method, the extraction recovery was 115.2% for n-butylbenzene, 80.9% for DBCP and method detection limit were $0.04{\mu}g/kg$ and $0.70{\mu}g/kg$, respectively. The extraction yields and detection limits of these compounds obtained by purge & trap were equivalent to those by SPME.

To develope a microbial pesticide for the control of mulbeny longicorn beetle, Apriona germari, Beauveria spp. Were isolated from the infected Apriona germari larvae. The morphology of Berauveria spp. Was observed by phase contrast and scanning electron microscope. In addition, the Beauveria spp. Isolated from Apriona germari were identified by the rando amplification of polymorphic DNA using polymerase chain reaction. The results showed that the Beauveria spp., SFB-1A and SFB-3A, issolted from Apriona germari were identified with B. bassiana and B. brongniartii, respectively, suggesting that the random amplification of polymorphic DNA is effective for the identification of Beauveria spp.
흰굳음병균(Beauveria bassiana) 단균사의 대량배양생산을 위한 액체배지 개발

To develope a liquid culture media for the mass culture of Beauveria bassiana blastospore, wheat bran and soybean cake were selected as a carbon and nitrogen source, respectively. The composition and final concentration of wheat bran nd soybean cake in liquid culture media were determined by the growth of B. bassiana blastospore. The results showed that the growth of B. bassiana blastospore was effective in the rate of 3 wheat bran : soybean cake and in 5% of final concentration of the liquid culture media, suggesting that the liquid culture media using wheat bran and soybean cake are available.
흰굳음병균(Beauveria bassiana) 포자의 대량배양을 위한 고체배지의 개발

To develope a solid culture media for the mass culture of Beauveria bassiana conidia, wheat bran was seleted as C source and N source. The pellet media were prepared without(P-I) and with (P-II) solidified ingredient. The growth of Beauveria bassiana on he media type was more effective in media of pellet type that powder type. In addition, production of Beauveria bassiana conidia on the media size was more effective in media of S type(Ø 7 mm). And the yield of Beauveria bassiana conidia at 20°C cultivation on 3 weeks post inoculation was similar to that of 25°C. Above-mentioned results showed that pellet medium(P-I) was effective to growth of Beauveria bassiana, suggesting that the pellet media may be useful in the mass production of Beauveria bassiana conidia.
