HP1492 is a NifU-like protein of Helicobacter pylori (H. pylori) and plays a role as a scaffold which transfer Fe-S cluster to Fe-S proteins like Ferredoxin. To understand how to bind to iron ion or iron-sulfur cluster, HP1492 was expressed and purified in Escherichia coli (E. coli). From the NMR measurement, we could carry out the sequence specific backbone resonance assignment of HP1492. Approximately 91% of all resonances could be assigned unambiguously. By analyzing results of CSI and TALOS from NMR data, we could predict the secondary structure of HP1492, which consists of three a-helices and three b-sheets. This study is an essential step towards the structural characterization of HP1492.
pH Effect on the Structure of Reduced NifU–like Protein from Helicobacter pylori

Helicobacter pylori (H. pylori) survives in acidic and fluctuating pH conditions of the stomach. The pH effect on H. pylori proteins is important for the advanced understanding of its evolution and viability, although this bacterium has the molecular machinery that neutralizes the acidic condition. HP1492 is known as a conserved NifU–like protein from H. pylori. NifU is a nitrogen fixation protein that mediates the transfer of iron–sulfur (Fe–S) cluster to iron–sulfur proteins like ferredoxin. Commonly, the monomeric reduced state of NifU can be converted to the dimeric oxidized state by intermolecular disulfide bond formation. Because it remains unclear that HP1492 actually behaves as known NifU protein, we first found that this protein can adopt both oxidized and reduced forms using size exclusion chromatography. Circular dichroism experiment showed that HP1492 is relatively well–structured at pH 6.5, compared to other pH conditions. On the basis of the backbone resonance assignment of HP1492, we further characterized the residues that are sensitive to pH using NMR spectroscopy. These residues showing large chemical shift changes could be mapped onto the secondary structure of the protein. Our results could provide the foundation for structural and biophysical studies on a wide spectrum of NifU proteins.

Purpose: The purpose of this study was to compare universal goniometry (UG), which is commonly used in clinical practice to measure the range of motion (ROM) of finger joints with a wearable soft sensor glove, and to analyze the reliability to determine its usefulness. Methods: Ten healthy adults (6 males, 4 females) participated in this study. The metacarpophalangeal joint (MCP), interphalangeal joint (IP), and proximal interphalangeal joint (PIP) of both hands were measured using UG and Mollisen HAND soft sensor gloves during active flexion, according to the American Society for Hand Therapists’ measurement criteria. Measurements were taken in triplicate and averaged. The mean and standard deviation of the two methods were calculated, and the 95% limits of agreement (LOA) of the measurements were calculated using the intraclass correlation coefficient (ICC) and Bland–Altman plot to examine the reliability and discrepancies between the measurements. Results: The results of the mean values of the flexion angles for the active range of motion (AROM) of the finger joints showed large angular differences in the finger joints, except for the MCP of the thumb. In the inter-rater reliability analysis according to the measurement method, the ICC (2, 1) value showed a low level close to 0, and the mean difference by the Bland–Altman plot showed a value greater than 0, showing a pattern of discrepancy. The 95% LOA had a wide range of differences. Conclusion: This study is a preliminary study investigating the usefulness of the soft sensor glove, and the reliability analysis showed a low level of reliability and inconsistency. However, if future studies can overcome the limitations of this study and the technical problems of the soft sensor glove in the development stage, it is suggested that the measurement instrument can show more accurate measurement and higher reliability when measuring ROM with UG.
Acrinathrin 외 농약 65종에 대한 독성시험의 분석 연구

This study analyzed the ADI(Acceptable Daily Intake) of the toxicity assessment report on 65 pesticides including Acrinathrin, including 31 herbicides, 15 insecticides, 15 bactericides, 4 plant growth regulator and 1 herbicide safener registered in domestic country. NOAEL and ADI of pesticide toxicity study evaluated at domestic and foreign institute, toxicity study type based ADI instillation, grade of ADI level, major toxicity and target organ were analyzed. As a result, ADI analysis among agencies, the agency with the detail pesticide toxicity data is the Food Safety Commission in Japan(FSC, 53%, 44/66). Other agency is EPA, EFSA, JMPR, INCHEM, in that order. Major contents of the ADI difference among institution was the type of the toxicity test, modification of significant figures and safety factor. As for major toxicity study affecting ADI instillation, the combined chronic/carcinogenicity test was the highest 35%(23/66). Other toxicities were chronic toxicity, reproductive and developmental toxicity, carcinogenicity, repeated dose toxicity test in that order. As for the classification of ADI grade, a number of ADI range was usually confirmed in the 0.01 mg/kg bw/day<x≤0.1 mg/kg bw/day(38%, 25/66). The major target organ affecting ADI instillation was the liver(hypertrophy, vacuolar degeneration etc). After that, the brain(cholinesterase related lesions) and thyroid gland were listed in that order, and other organs were selected as target organs. Therefore, it will be contributed to evaluate the toxicological characteristics of pesticides by ADI classification and analysis of 66 pesticides.
