        Inflammasome-Dependent Peroxiredoxin 2 Secretion Induces the Classical Complement Pathway Activation

        Peroxiredoxins (Prxs) are ubiquitously expressed peroxidases that reduce hydrogen peroxide or alkyl peroxide production in cells. Prxs are released from cells in response to various stress conditions, and they function as damage-associated molecular pattern molecules. However, the secretory mechanism of Prxs and their roles have not been elucidated. Thus, we aimed to determine whether inflammasome activation is a secretory mechanism of Prxs and subsequently identify the effect of the secreted Prxs on activation of the classical complement pathway. Using J774A.1, a murine macrophage cell line, we demonstrated that NLRP3 inflammasome activation induces Prx1, Prx2, Prx5, and Prx6 secretion in a caspase-1 dependent manner. Using HEK293T cells with a transfection system, we revealed that the release of Prx1 and Prx2 relies on gasdermin-D (GSDMD)-mediated secretion. Next, we confirmed the binding of both Prx1 and Prx2 to C1q; however, only Prx2 could induce the C1q-mediated classical complement pathway activation. Collectively, our results suggest that inflammasome activation is a secretory mechanism of Prxs and that GSDMD is a mediator of their secretion. Moreover, secreted Prx1 and Prx2 bind with C1q, but only Prx2 mediates the classical complement pathway activation.

        가끔 우리는 생각지도 못한 일이 생겨 돼지가격이 곤두박질치는 것을 본다. 지난 2002년 5월 경기 충북 일대에서 발생한 구제역이 바로 그런 경우인데, 양돈기업들은 그때 전 재산이나 다름없는 수만 마리의 돼지들을 땅속에 묻어야 했다. 돼지 가격을 흔드는 것은 구제역, 돼지콜레라 뿐만이 아니다. 돼지 가격이 좋을 때 갑자기 사육 두수가 늘어나면 언제 또 돼지 값이 폭락할 지 알 수가 없다. 지난 10년간의 돼지 가격은 거의 매년 폭락과 폭등을 반복하고 있다. 가격이 좋을 때에는 괜찮겠지만 문제는 가격이 폭락할 때이다. 가격이 좋아서 큰돈을 버는 것도 좋지만 거의 해마다 반복되는 가격 폭락을 피하고 돼지를 제값에 팔 수 있다면 얼마나 좋을까? 여기에 '선물거래'가 해답이 될 수 있다.

        Higher Extraction of Phytochemical Compounds from Tartary Buckwheat Seeds by the Application of Surfactant Formulation

        The aim of this study was to determine the suitability of surfactant to extract higher phenolic compound, flavonoid and antioxidant activity from Tartary buckwheat and evaluate the potentiality of surfactant as a screening agent for breeding purpose. Primarily, we employed two types of surfactant (Hydrophilic: Tween 20 and Lipophilic: Span 80) to select the suitable surfactant agent for the extraction of optimum bioactive compounds. Between two surfactants, Tween 20 showed highest efficiency at 4 mM concentration to extract total phenolic content (TP), total flavonoid (TF) and antioxidant activity (AA). Tween 20 at 4 mM concentration was fixed for further analysis along with hot water (90℃) treatment as a control. In our findings, highest TP (118 ㎎/g), TF (38 ㎎/g) and AA (76%) was achieved in KW21 and KW22 among the fifteen accessions of Tartary buckwheat. In other way, TP, TF and AA was 200%, 120% and 110% higher in surfactant formulation compared with control treatment, respectively.

        피브린 글루/알지네이트 상호 침투 구조체에 의한 지혈 및 기계적 물성 개선

        Fibrin glue (FG) has been worldwidely used as a potent surgical adhesive for microvascular blood vessel anastomoses. It needs to improve some problems in present clinical uses by allowing a quicker clotting time and higher mechanical properties. The purpose of this study aims to engineer the novel fibrin glue/alginate (ALG) interpenetrating network (FG-ALG IPN) adhesive to achieve both requirements. The alginate networks in FG shorten the clotting time and enhance the mechanical properties. However, the incorporation of ampicillin (AMP) as antibiotics does not affect both clotting time and mechanical properties. As a consequence, this novel FG-ALG IPN is expected to be used for hemostasis, tissue adhesive, suture support, wound care, sealing and filling of body defect, drug carriers, and tissue-engineered scaffolds.

        시츄견에서 질세포 검사를 이용한 발정 유기 효과의 확인

        To investigate the change of vaginal epithelial cell in estrus-induced Shih-tzu bitch, estrus was induced by PMSG (50 IU/kg, for 10 days) and hCG (1,000 IU, on Day 10) in 12 anestrus Shih-tzu bitches. Day-changes of vaginal epithelial cells during the whole period of hormone treatment were investigated in each experimental bitches. The first day of vulvar bleeding and the first day of male acceptant was on days (), and days after the first PMSG treatment. The duration of proestrus and estrus was days, and days. Characteristic features of vaginal cytology during the estrous cycle were the high proportion of large intermediate cell, superficial cell, anuclear cell and erythrocyte in proestrus, superficial cell and anuclear cell in estrus, and parabasal cell, small and large intermediate cell, and leukocyte in diestrus, respectively. When it was timed from the first day of PMSG administration (Day 0), the cornification index (CI) was the high proportion in proestrus and estrus. The CI peak was maintained above 80% between Day 11~14 (4 days) and CI showed a peak at Day 12. These results indicated that the 12 Shih-tzu bitches showed positive estrus induction by vaginal smear test and observation of clinical estrus sign.

        Peptide-Grafted Lactide-Based Poly(ethylene glycol) Porous Scaffolds for Specific Cell Adhesion

        While biodegradable polyesters or their copolymers have been used widely as scaffolding materials in tissue engineering, these polymers intrinsically lack the cell-binding motifs on the surface. In this study, a GL-PEG,which is a poly(ethylene glycol) monoacrylate combined with glycerol (G)-lactide (L) triols, was synthesized and fabricated into a porous three dimensional (3D) structure. In addition, peptide ligands, Arg-Gly-Asp (RGD) or Arg-Glu-Asp-Val (REDV) were grafted directly onto the GL-PEG, producing GL-PEG-GRGD and GL-PEG-GREDV scaffolds to improve the specific cell adhesion. Both GL-PEG and GL-PEG-GRDG scaffolds were also prepared as a control. The ESCA spectra showed that peptide-grafted GL-PEG carried a nitrogen peak, which is indicative of the amine group in the peptide sequence. The nitrogen content in the GL-PEG-peptides was significantly higher than that on the the GL-PEG. When the peptide-grafted GL-PEG scaffolds were seeded with fibroblasts and endothelial cells (ECs) and cultured for up to 4 days, specific cell interactions were identified by scanning electron microscopy (SEM). More fibroblasts were found on the GL-PEG-GRGD scaffold but more ECs were found attached to GL-PEGGREDV. The total protein assay also supported the same trend that the fibroblasts/GL-PEG-GRGD or EC/GL-PEGGREDV constructs had the highest protein content with the specific cells, compared with the other groups. This data suggests that cell adhesion could be specific and dependent on the type of peptide ligands. Such 3D porous and crosslinked peptide-grafted GL-PEG scaffolds can be very useful for harboring specific cell populations for tissue engineering and vascular stents.

        정상인과 난청인에서 다화자 잡음을 이용한 소음환경하에서의 50% 인지 신호대잡음비의 비교

        The most common complaint of individuals with hearing impairment is the difficulty in understanding speech against background noise. In our previous study, we carried out a speech-in-noise test using multi-talker babble noise and spondee words. In the present study, we compared 50%-signal to noise ratios (SNRs) of hearing-impairment group with that of normal-hearing group, and analyze the effects of age and noise intensity affecting the results. Subjects and Method: Thirty listeners (of which each 10 listeners are in their forties, fifties, and sixties) with normal hearing and thirty listeners (each ten listeners in their forties, fifties, and sixties) with hearing impairment participated. Spondee words and multi-talker babble noise made from recording the voices of 10 males and 10 females were presented by a loudspeaker at a distance of one meter in front of the subject. Fifty percent-SNRs were measured at 50 dB HL, 60 dB HL, 70 dB HL, and 80 dB HL of noise intensity. Results: The mean 50% SNR for normal-hearing listeners in the forties, fifties and sixties of age were -13.4±2.1 dB, -11.5±2.3 dB, and -12.6±2.0 dB, respectively. The mean 50% SNR for hearing-impairment listeners in the forties, fifties and sixties of age were -11.2±3.4 dB, -9.5±3.0 dB, -8.7±2.7 dB, respectively. Hearing-impairment group showed significantly worse results than the normal- hearing group in all ages. Conclusion:These results support that our word-in-noise test using multi-talker babble noise is useful in measuring audibility against background noise for individuals with hearing-impairment.         소형 애완견에서 분만일 예측에 대한 질세포 검사의 활용

        The aim of this study was to estimate of prediction of whelping day and to confirm the accuracy of prediction of whelping day in small pet bitches. The gestation length from the each based on days was 66.64 ± 2.32 days (Mean ± S.D) when the Cornification index (CI) was over 90% after the first vaginal discharge, 64.65 ± 2.87 days from the day of CI peak, 63.46 ± 1.63 days from the day of ovulation by progesterone concentrations, and 57.67 ± 2.43 days from the first day of cytologic diestrus, respectively. The whelping day was estimated 66 days when the CI was over 90% after the first vaginal discharge, 64 days from the day of CI peak, 63 days from the day of ovulation by progesterone concentrations, and 57 days from the first day of cytologic diestrus, respectively. The accuracy of the prediction of whelping day was 90.0% with a precision of ± 2 days when the CI was over 90% after the first vaginal discharge, 77.5% from the day of CI peak, 86.2% from the day of ovulation by progesterone concentrations, and 81.2% from the first day of cytologic diestrus, respectively. These results indicated that the prediction of parturition day by vaginal cytology was useful method for management of reproduction and parturition in small pet bitches.

        Thin film composite membrane prepared by interfacial polymerization as an ion exchange membrane for salinity gradient power

        For extracting the salinity gradient energy, membrane is a key material with a high power density. For this purpose, thin film composite (TFC) membranes were synthesized by interfacial polymerization using various acyl chloride and amine monomers. TFC membranes were characterized by FT-IR, SEM, XPS, and AFM. Among the range of TFC, the best candidate was prepared by combining 3,5-diaminebenzonic acid (0.5 wt% with distilled water) and 1,3,5-benzenetricarbonyl trichloride (0.2 wt% with n-hexane). The TFC membrane (thickness = 0.02 mm) generated a power density of 7.2 W/m2 at salinity gradient between 0.005 and 0.5 M KCl solutions.
