Recently, for the purpose of choosing reasonable and economical construction, the decision making analyses, such as VE (Value Engineering) and LCC (Life Cycle Cost), are generally performed. But, these methods have some limitations for the application to subway tunnel construction method. So, the AHP (Analytical Hierarchy Process) analysis is applied to determine relative importance for obtaining objectivity in quantitative analysis. And, the LCSC (Life Cycle Social Cost) method is developed to account for the social loss and risk by tunnel construction. From the case of the subway lot, those methods are very useful to decision making analysis. And, in that case, both quantitative and quantitative analysis appraisement, the drilling and blasting method is evaluated useful alternative proposal in comparing with the cut and cover method. 지하철 터널의 합리적인 건설공법 채택을 위해 최근 건설공법의 선정에 적용되고 있는 VE 및 LCC기법의 문제점을 분석하여 개선안을 도출하고자 정성적인 분석에 있어서 AHP기법을 통한 가능한 여러 대안들간의 상대적 중요도를 결정하는 방식으로 객관성을 확보하고자 하였다. 또한, 정량평가에 있어서는 경제적인 터널구조물 건설을 목적으로 설계단계에서 기존 LCC 기법에서 고려하지 못하는 건설공사에 따른 사회적 손실을 비용으로 환산하는 LCSC 평가기법을 제안하였다. 실제 지하철 ○○공구에 적용사례를 통해 그 효용성을 검증하였으며, 검토결과 해당 공구에서는 정성평가 및 정량평가 항목 모두에서 개착공법 보다는 터널공법의 채택이 합리적인 대안으로 평가되었다.

Diverse bacteria are known to colonize plants. However,only a small fraction of that diversity has been evaluated for their biopesticide potential. To date, the criteria for sampling and selection in such bioprospecting endeavors have not been systematically evaluated in terms of the relative amount of diversity they provide for analysis. The present study aimed to enhance the success of bioprospecting efforts by increasing the diversity while removing the genotypic redundancy often present in large collections of bacteria. We developed a multivariate sampling and marker-based selection strategy that significantly increase the diversity of bacteria recovered from plants. In doing so, we quantified the effects of varying sampling intensity, media composition, incubation conditions,plant species, and soil source on the diversity of recovered isolates. Subsequent sequencing and highthroughput phenotypic analyses of a small fraction of the collected isolates revealed that this approach led to the recovery of over a dozen rare and, to date, poorly characterized genera of plant-associated bacteria with significant biopesticide activities. Overall, the sampling and selection approach described led to an approximately 5-fold improvement in efficiency and the recovery of several novel strains of bacteria with significant biopesticide potential.
개선된 DRASTIC 기법을 이용한 터널굴착 예정지역의 지하수 오염 가능성 평가기법 개발에 관한 연구

지하수 오염에 대한 광역적인 취약성 평가를 위한 DRASTIC 시스템은 미국 EPA에서 개발된 것으로서 지하수위, 충진률, 대수층매체, 토양매체, 지형경사, 비포화대매체, 수리전도도등 수리지질학적 요소들을 이용하여 지하수 오염 취약성을 상대적으로 평가하기 위한 표준화된 시스템으로 현재 널리 사용되고 있다. 본 연구에서는 GIS 기법을 활용하여 기본 DRASTIC 모델에 구조선 밀도, 토지이용, 터널굴착으로 인한 지하수위 저하의 영향을 추가한 변형된 DRASTIC 모델을 개발하고 ${\bigcirc}{\bigcirc}$지역의 터널굴착으로 인한 지하수 오염가능성을 평가하고자 하였다. 통계적인 분석을 위하여 각각의 인자별 벡터 레이어를 격자레이어로 전환하고 각 인자별 상관계수, 3방향의 주성분분석에 의한 공분산, 고유치, 고유벡터 등을 구하여 인자들의 상관관계를 비교 분석하였다. 또한, 일반적인 DRASTIC 레이어와 구조선 밀도 레이어, 토지이용 레이어, 터널굴착영향 레이어간의 상관계수를 각각 구하고, 이를 각각의 가중치로 적용하여 최종적인 변형된 DRASTIC 모델을 구축하고자 하였다. 변형된 DRASTIC 모델을 사용한 결과 일반적인 DRASTIC 모델에 비해 오염 가능지역이 많이 세분화되었고, 그 결과를 활용하여 오염가능성이 높은 취약지역을 구체적으로 구분할 수 있었다. The DRASTIC system is widely used for assessing regional groundwater pollution susceptibility by using hydrogeological factors such as depth to water, net recharge, aquifer media, soil media, topography, vadose zone media, hydraulic conductivity. This study is providing Modified Drastic Model to which lineament density, land use, influence of groundwater drawdown caused by tunnel excavation are added as additional factors using geographic information system, and then to evaluate groundwater contamination potential of ${\bigcirc}{\bigcirc}$ area. For statistical analysis, vector coverage per each factor is converted to grid layer and after each correlation coefficient between factors, covariance, variance, eigenvalue and eigenvector by principal component analysis of 3 direction, are calculated, correlation between factors is analyzed. Also after correlation coefficients between general DRASTIC layer and rated lineament density layer, between general DRASTIC layer and rated land use layer, between general DRASTIC layer and rated tunnel excavation influence layer are calculated, final modified DRASTIC model is constructed by using them with each weighting. When modified DRASTIC model was compared with general DRASTIC model, contamination potential in modified DRASTIC model is fairly detailed and consequently, vulnerable area which has high contamination potential could be presented concretly.
복합기능성 Bacillus velezensis GH1-13 균주의 대량배양 최적화 및 특성

Bacillus genus are found abundantly in various sites and their secondary metabolites were used as potential agents in agriculture, notably plant growth promoting and bio-control. The objective of this study was to develop the culture conditions of GH1-13 strain including higher cell growth, stable endospore-forming and enhancement of potential agents which are related with plant growth promoting and phytopathogen suppression. The optimal carbon and nitrogen sources were determined by glucose and soy bean flour, respectively, then resulted in 7.5×109 cells/mL, 6.8×109 endospore cells/mL and sporulation yield of 90% after 30 h cultivation in 500 L submerged fermenter at 37℃, pH 7.0. Cells and cell-free supernatant of GH1-13 strains showed the potent antifungal activity against phytopathogenic fungi of Colletotrichum gloeosporioides. It was also confirmed that indole-3-acetic acid (IAA) production of GH1-13 strain was greatly increased by addition of 0.3% tryptophan.
