        영지버섯균 발효 꾸지뽕나무 가지 톱밥 추출물의 항염증 활성

        염증은 외부 자극으로부터 보호하는 중요한 면역반응이다(Jeong et al., 2012). 그러나 NO 및 inflammatory cytokine과 같은 염증 매개 인자의 과도한 생성은 비정상적인 염증 반응을 초래할 수 있다(Paradise et al., 2010). 염증 매개 인자의 생성과 염증 신호 전달을 조절하는 중요한 역할을 하는 대식세포는 LPS 자극에 의해 NF-κB가 활성화 되어 inflammatory cytokine 등의 염증 매개 인자들이 분비가 증가되며 염증 반응을 심화시킨다(Lee et al., 2012). 본 연구에서는 영지버섯균 발효 꾸지뽕나무 가지 톱밥 추출물을 이용하여 LPS로 염증을 유도한 Raw264.7세포에서 염증 관련 인자들의 생성 및 발현에 미치는 영향을 분석하여 항염증 효과를 확인하였다. NO는 염증에서 중요한 역할을 하며 대식세포의 염증 반응 조절 평가시 대표적인 지표로 사용되며 iNOS에 의해 생성이 유도되며 PGE<sub>2</sub>는 염증 매개 인자로 inflammatory cytokine 생성에 관여하며 COX-2에 의해 생성이 유도된다(Paradise et al., 2010; Posadas et al., 2000). 발효추출물이 RAW264.7세포에서 세포독성 없이 NO의 생성과 PGE<sub>2</sub>의 생성을 감소시켰으며 이 결과는 발효 추출물 처리에 의해 iNOS와 COX-2의 발현이 감소한 것과 일치하였다. 또한 염증 반응을 조절하는 대표적인 inflammatory cytokine으로 알려진 IL-1β, TNF-α의 생성이 감소되었다. 따라서 발효추출물은 NO, PGE<sub>2</sub>, inflammatory cytokines 생성을 감소시켜 항염증 효과를 나타낸다고 볼 수 있다. NF-κB는 iNOS, COX-2 및 inflammatory cytokine의 발현을 조절하는 전사인자로 IκB와의 결합에 의해 NF-κB의 핵 내 이동이 억제되며 불활성화상태로 존재한다. LPS 자극에 의해 IκB가 인산화 및 분해되면 NF-κB가 활성화되어 핵 내로 이동하여 염증성 매개인자들의 발현을 유도하고, 염증성 질환 뿐만 아니라 다양한 질환을 유발시키는 것으로 알려져 있다 (Kawai and Akira, 2006; Ghosh and Ksarin, 2002). 본 연구에서는 LPS에 의해 자극된 RAW264.7 세포에서 IκB의 분해 및 NF-κB의 전이가 발효추출물에 의해 감소됨을 확인하였으며 발효추출물에 의한 iNOS, COX-2 및 inflammatory cytokine 감소는 NFκB 활성화 감소에 의해 조절되는 것으로 사료된다. 이러한 결과들을 통해서 발효추출물이 NF-kB 활성화 억제를 통해 염증 매개 인자들의 생성 및 발현을 감소시킴으로서 염증 반응 억제 효과를 나타냄을 확인하여, 염증 관련 질환에 대해 예방 및 개선을 위한 항염증 기능성 소재로 사용될 수 있음을 시사한다. In this study, we evaluated the anti-inflammatory effect of extract from Cudrania tricuspidata twig sawdust fermented with Ganoderma lucidum mycelium. Fermented Cudrania tricuspidata twig sawdust extracted with 70% ethanol and elucidated the potential signaling pathway in lipopolysaccharide (LPS)-induced RAW264.7 cells. Fermented Cudrania tricuspidata twig sawdust inhibits LPS-stimulated nitric oxide (NO) production without affecting cell viability in a dose-dependent manner and production of LPS-induced pro-inflammatory cytokines such as interleukin (IL)-1β, tumor necrosis factor (TNF)-α and prostaglandin2 (PGE<sub>2</sub>). Fermented Cudrania tricuspidata twig sawdust also suppressed the expression of the pro-inflammatory mediators such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in LPS-stimulated RAW264.7 cells. Moreover, Fermented Cudrania tricuspidata twig sawdust significantly attenuated LPS-induced IkappaB (IκB) degradation and suppressed nuclear factor kappa B (NF-κB) nuclear translocation. These results suggest that fermented Cudrania tricuspidata twig sawdust may have great potential for the development of anti-inflammatory agent.

        Effects of a hybrid online and offline program for facilitating father-infant interactions in South Korea: a quasi-experimental study

        Purpose: This study examined the effects of an educational program for fathers on improving father-infant interactions, child-rearing knowledge, and attachment. Methods: In this quasi-experimental study, the participants were recruited by convenience sampling among fathers with infants (2-6 months of age) residing in three districts of Seoul. Fifteen participants in the experimental group and 17 participants in the control group completed the follow-up investigation. A 5-week online and offline intervention program with five sessions was provided to the experimental group. The data were analyzed using generalized estimating equations (GEEs). Results: There was a significant difference in the change in father-infant interaction scores of the experimental group, especially in the caregiver aspect with a significance level of .100 in time and group-to-group interactions (B=6.46, p=.051, 95% confidence interval [CI]=-0.02-12.94). The changes between the groups and times were not statistically significant when it came to infant development knowledge and father-infant attachment. Conclusion: We conclude that hybrid online and offline education should be implemented as an effective method to improve fathers’ interactions with their children based on accurate knowledge about infant development.

        금화규 꽃과 천년초 열매 복합 추출물의 미백 및 주름 개선 효과 분석

        This study was conducted to investigate the whitening and anti-wrinkle effects of complex extract of Abelmoschus manihot flower and Opuntia humifusa fruit (AOCE) and their potential applications in cosmetic products. AOCE led to a concentration-dependent reduction in intracellular tyrosinase activity and melanin content. Furthermore, the expression of tyrosinase, TRP-1, and TRP-2, which are involved in melanin synthesis, was downregulated by AOCE. AOCE inhibited decreases in hyaluronic acid and pro-collagen, reduced MMP-1 activity, and lowered the expression of MMP-3 and MMP-9, which are involved in wrinkle formation by degrading collagen. The complex extract of AOCE showed enhanced whitening and anti-wrinkle effects compared with the effects of single extract treatment, suggesting a synergistic effect among individual extracts. AOCE shows whitening and anti-wrinkle effects and may be used as a safe natural cosmetic material.

        야간 라이딩을 위한 스마트 애슬레져 패션디자인 개발 연구

        The purpose of this study is to develop the fabric electronics-based safety protection smart athleisure fashion for night riding. Based on the characteristic analysis of products being used during night riding, the fabric LED Display and fabric type capacitive touch sensor to emphasize human friendliness were designed in detachable form through an intuitive interface to develop the smart athleisure fashion of prototype. This is meaningful in that it proposed smart Athleisure fashion products differentiated from existing products, considering the functional aspects based on fabric electronics that emphasized human friendliness and the design aspects of the customized system that can diversify design through exchange and compatibility with other Athleisure products due to detachable form. Follow-up study will focus on the improvement of textile sensor fit for the physical properties of a textile with reinforced wearability and flexibility by using Fabric Electronics and proposed customized smart fashion based on it. 본 연구는 야간 라이딩을 위한 패브릭 일렉트로닉스 기반의 안전 보호용 스마트 애슬레져 패션 개발 연구를 목적으로 하며 인간 친화성이 강화된 Fabric LED Display와 Fabric Capacitive touch sensor를 전도성 원사를 개발 및 활용하여 탈·부착 형태로 설계하여 구현하였다. 이러한 모듈의 탈·부착성은 다른 애슬레져 제품과 기능적·심미적인 요소를 교환 및 호환을 할 수 있도록 설계하여 기능뿐만 아니라 디자인의 다양화가 가능한 커스터마이즈 시스템으로 기존 제품들과 차별화된 스마트 애슬레져 패션 제품을 제안하였다는 점에서 연구의 의의가 있다. 후속 연구에서는 Fabric Electronics를 활용하여 착용성과 유연성이 강화된 직물의 물리적 속성에 적합한 직물센서를 개발하고 이를 활용한 커스터마이즈 스마트 패션을 제시할 예정이다.

        세탁과 건조에 따른 양모 위편성물의 편성조직별 형태 변화

        The purpose of this study is to analyze the structural properties of 100% wool fabrics knitted with various stitch types and to evaluate dimensional stability from shrinkage in wet cleaning and drying. Materials were weft-knitted from twenty-four different stitches with 7 gauge using a computerized flatbed knitting machine. Weight, thickness, density, and length were measured. A domestic washing machine and a tumble dryer were used for the shrinkage test. The results are as follows: Knitted fabrics were divided into 3 groups based on weight per unit area. Porous knits show light weight whilst milano, pintuck, rib stitches belong to the heaviest group. A positive correlation between weight and thickness was found and the same result was obtained for wale density and weight. Dimensional shrinkage of knitted fabrics was increased during repetitive wet cleaning and drying regardless of knit stitches. Especially, fabrics knitted with float, tuck, cable, and links & links stitches samples were contracted more than 15% in the first treatment whereas 2x1 rib stitch showed 1% shrinkage rate. Fisherman and milano stitches contracted in both course and wale direction with similar shrinkage rates. However, porous knits with float and tuck stitches shrank in course direction by 20% as well as cable samples contracted from 5% to 20% after repeated washing and drying. On the other hand, 30% and 15% contraction of wale direction occurred in orderly float and links & links stitches, respectively. Machine dried knits have a higher shrinkage rate than air-dried knits, but the drying method did not affect to the direction of contraction. In conclusion, variations of knit, tuck, and float stitches affect knit construction and dimensional stability from shrinkage in wet cleaning and drying of wool knitted fabrics.

        Dose-Dependent Effect of Smoking on Risk of Diabetes Remains after Smoking Cessation: A Nationwide Population-Based Cohort Study in Korea

        Background This study aimed to evaluate the dose-dependent effects of smoking on risk of diabetes among those quitting smoking. Methods We analyzed clinical data from a total of 5,198,792 individuals age 20 years or older who received health care check-up arranged by the national insurance program of Korea between 2009 and 2016 using the Korean National Health Insurance Service database. Cumulative smoking was estimated by pack-years. Smokers were classified into four categories according to the amount of smoking: light smokers (0.025 to 5 smoking pack-years), medium smokers (5 to 14 smoking pack-years), heavy smokers (14 to 26 smoking pack-years), and extreme smokers (more than 26 smoking pack-years). Results During the study period, 164,335 individuals (3.2% of the total population) developed diabetes. Compared to sustained smokers, the risk of diabetes was significantly reduced in both quitters (hazard ratio [HR], 0.858; 95% confidence interval [CI], 0.838 to 0.878) and nonsmokers (HR, 0.616; 95% CI, 0.606 to 0.625) after adjustment for multiple risk factors. The risk of diabetes gradually increased with amount of smoking in both quitters and current smokers. The risk of diabetes in heavy (HR, 1.119; 95% CI, 1.057 to 1.185) and extreme smokers (HR, 1.348; 95% CI, 1.275 to 1.425) among quitters was much higher compared to light smokers among current smokers. Conclusion Smoking cessation was effective in reducing the risk of diabetes regardless of weight change. However, there was a potential dose-dependent association between smoking amount and the development of diabetes. Diabetes risk still remained in heavy and extreme smokers even after smoking cessation.

        아이스플랜트 추출물의 3T3-L1 지방세포 내 TNF-α 유도 염증 및 인슐린 저항성 개선 효과

        Obesity is classified as a low-grade chronic inflammatory disease. Obesity-induced inflammation is associated with the development of insulin resistance (IR). In this study, we investigated the effects of Mesembryanthemum crystallinum extract (MCE) on tumor necrosis factor-alpha (TNF-α)-induced inflammation and IR in 3T3-L1 adipocytes, as well as underlying mechanisms. Lipolysis was monitored by oil red O staining. Glucose uptake was measured by 2-deoxyglucose (2-DG). The production of cytokines and adiponectin was evaluated using enzyme-linked immunosorbent assay (ELISA). Protein expression was measured using western blotting. IR was induced in differentiated 3T3-L1 adipocytes by treatment with TNF-α. MCE treatment decreased lipolysis while increasing adiponectin production. MCE inhibited the production of pro- inflammatory cytokines such as interleukn (IL)-6 and monocyte chemoattractant protein (MCP)-1, as well as the Nuclear factor-κB (NF-κB) translocation. Additionally, MCE increased glucose uptake, prevented the protein expression of phosphorylated insulin receptor supstrate (IRS) and increased the protein expression of Glucose Transporter Type 4 (GLUT4) in a dose-dependent manner. These results demonstrate the potential of MCE in improving chronic inflammation and insulin sensitivity in obesity. Therefore, MCE can be used as a novel candidate for developing functional foods.

        코끼리마늘 추출물이 TM3 Leydig 세포의 테스토스테론 합성에 미치는 영향

        This work investigated the effects of Allium ampeloprasum extract (AME) on testosterone synthesis in TM3 Leydig cells under hydrogen peroxide (H2O2)- induced oxidative stress. AME showed no increment of cell proliferation in the oxidatively stressed cells. Testosterone level lowered when the cells were treated with H2O2, but increased in the presence of AME. To determine the effect of AME on the testosterone biosynthetic pathway, we examined the expression level of the proteins involved in the synthesis or degradation of testosterone. The protein expression levels of CYP11A1, 3β-HSD2 and 17β-HSD3 (which are involved in testosterone synthesis) were decreased by H2O2 but increased by the AME treatment. Furthermore, the protein expression levels of 5α-reductase2 and aromatase (which are involved in testosterone degradation) were increased by H2O2 but decreased by the AME treatment. In conclusion, AME increased the testosterone level by increasing the expression of testosterone synthetase and decreasing the expression of the testosterone converting enzyme in TM3 Leydig cells under H2O2-induced oxidative stress. These results indicated that AME can alleviate andropause syndrome by restoring testosterone.
