The rapid development of biotechnology has increased the importance of microorganisms or their genetic information. Thus, the Nagoya Protocol on access to genetic resources and the fair and equitable sharing of benefits arising from their utilization was established, and countries are working to secure industrially and academically useful bioresources to deal with the agreement. In the case of Korea, because 67% of bioresources are imported from abroad, we are required to secure domestic bioresources as well. The number of isolated foodborne illness-causing microorganisms is predicted to invrease based on the incresing number of outbreaks of foodborne illness each year. Consequently, appropriate long-term preservation methods are necessary to secure the isolated microorganisms for the purpose of research and resourcification. Therefore, the long-term preservation methods for bacteria, fungi, viruses, and protozoa were investigated in this study, from domestic and international bioresource banks, and the functions of the cryoprotectants were reviewed and discussed. This review should be informative in the preservation of microorganisms and contribute to the development of biotechnology.

Edema factor(EF) is a portion of anthrax toxin which produces edema when combined with protective antigen. This paper describes about technique for cloning, expression, purification and activity test of EF. Using the E. coli expression system, we could make recombinant EF protein although it's origin is Bacillus anthracis. And also we could culture massively and purify highly pure protein. Finally we confirm a enzyme activity of purified EF to increase intracellular cAMP level. Through establishing this technique, it can be possible to research about EF in depth and apply to expression and purification of many other protein in biology.
Quantitative Analysis on Cellular Uptake of Clustered Ferrite Magnetic Nanoparticles

Due to their ability to be internalized into cells by endocytosis through cell membranes, the application of nanoparticlesin therapeutic and diagnostic fields has received much interest. In particular, ferrite magnetic nanoparticles (MNPs) arewidely used as reagents for medical care, including in vitro magnetic separation, T2-weighted magnetic resonance imaging,magnetic hyperthermia therapy, and as drug delivery systems. However, little is known about the quantitative analysis ofthe cellular uptake of MNPs by the interaction of particle surfaces with biomolecules. Here, we quantitatively analyze theintracellular uptakes of 30 nm Fe- and Mn-ferrite MNPs. We confirm that the magnetic properties of MNPs change accordingto their microstructure and quantitatively analyze nanoparticle internalization in breast cell lines (MCF10A, MCF7, andMDA-MB-231) by measuring the magnetic moment using a vibrating sample magnetometer. Finally, we examine the effectof nanoparticle microstructure on cellular uptake in terms of the interaction between the nanoparticles and biomolecules.

Objective: Surface disinfection is important in the proper running of livestock farms. However, disinfection of farm equipment and facilities is difficult because they are made of different materials, besides having large surface areas and complex structures. 3-(trimethoxysilyl)-propyldimethyloctadecyl ammonium chloride (Si-QAC) is a quaternary ammonium salt-based disinfectant that attaches to various surfaces by forming covalent bonds and maintains its disinfecting capacity for a considerable time. Our aim was to evaluate the potential use of Si-QAC for disinfection of farm equipment and facilities.Methods: The short- and long-term antimicrobial and antiviral effects of Si-QAC were evaluated in both laboratory and farm settings using modified quantitative assessment method based on the standard operating procedures of the United States Environmental Protection Agency.Results: Si-QAC was highly effective in controlling the growth of the Newcastle disease virus and avian pathogenic Escherichia coli. Electron microscopy revealed that the mechanism underlying the disinfection activity of Si-QAC was associated with its ability to damage the outer membrane of the pathogen cells. In the field test, Si-QAC effectively reduced viral contamination of surfaces of equipment and space.Conclusion: Our results suggest that Si-QAC has great potential as an effective chemical for disinfecting farm equipment and facilities. This disinfectant could retain its disinfection ability longer than other commercial disinfectants and contribute to better farm biosecurity.
Pin-site scar revision with subcisions and acellular dermal matrix

External fixation is widely used in orthopedic and reconstructive surgery. However, it may leave retracted and depressed scars because it involves the percutaneous placement of transosseous pins or wires that are secured to an external scaffolding. These scars do not resolve or improve spontaneously, causing patients to experience dissatisfaction and possibly requiring a surgical intervention. Moreover, the management of scars that arise from external fixator pin sites is challenging because of their fibrotic and depressive features. However, by using subcisions and inserting acellular dermal matrix under the scars, the contour of the scars can be improved easily. Therefore, we present a case of pin-site scar revision using a technique that we developed.

IgA 신병증은 전 세계적으로 가장 흔한 원발성 사구체 질환으로 성인의 경우 30% 정도가 만성신부전으로 진행하며 소아의 경우 10% 정도가 만성신부전으로 진행한다. IgA 신병증은 전신증상이 없이 현미경적 혈뇨와 간헐적인 육안적 혈뇨를 특징으로 하며 심한 경우 단백뇨를 동반한다. IgA 신병증의 원인이나 발병기전은 정확히 알려져 있지 않으며. 국외에서는 IgA 신병증이 유전되는 가계에 대한 보고와 함께 관련 유전자 연구가 활발하지만 국내에서는 이에 대한 증례가 매우 드물다. 저자들은 국내에서 가족 내유전된 IgA 신병증에 대한 1례를 경험하였기에 문헌고찰과 함께 보고하는 바이다. IgA nephropathy is the most common form of primary glomerulonephritis and chronic glomerular disease worldwide including Korea. Familial gathering of IgA nephropathy suggests that genetic factors contribute to the development of this disease. Although there have been many reports on familial IgA nephropathy with genetic analysis and their pedigrees, there has been few reports in Korea. We reported a partial familial IgA nephropathy pedigree with a brief review of the literatures.
