      • 한국 재래종 연초(Nicotiana tabacum L.)의 화학적 특성

        Field experiment was conducted to find out the chemical characteristics of 12 Korean local varities, Local varities were compared with oriental, flue-cured and burley tobacco. Among tested varities, Hyangcho and Matskawa were desirable in respect of flavor, leaf color and texture. Hyangcho and Matskawa were higher in the content of nicotine and petroleum ether extract. Gajacho, Byeolcho and Kukjuo were higher in the content of reducing sugar, but lower nicotine, total nitrogen and petroleum ether extract. The nicotine and total nitrogen content of Kwangcho, Daruma and Catterton were lower than that of Br. 21. In the content of volatile organic acid which were major characteristic component of oriental tobacco, the Korean local varities were lower in the content of 3-methyl butanoic and 3-methyl pentanoic acid, but hirities, Hyangcho, Matskawa and Daruma were higher in the content of volatile organic acid. Korean local varities, especially small leaf group, were higher in the content of benzyl alcohol and furfural than any other type tobacco.

      • 근적외 분광분석법을 이용한 버어리종 잎담배 화학성분 분석

        This study was carried out to analyze chemical components in burley tobacco using near infrared spectroscopy(NIRS). Samples were collected in '96 and '97 crop year. Calibration equations were developed by modified partial least square. The standard error performance(SEP) of '96 crop year samples between NIRS and standard laboratory analysis were 0.25% for nicotine, 0.18% for total nitrogen, 0.59% for crude ash, 0.32% for ether extracts, and 0.14% for chlorine, respectively. The analytical results of '97 crop year samples were similar to those of '96 crop year samples. The analytical result of '97 crop year samples analyzed by '96 calibration equation was more inaccurate than that of '96 crop year samples. The SEP of '96 or '97 crop year samples applying calibration equation derived from '96 plus '97 crop year samples was similar to that of '96 or '97 crop year samples analyzed by '96 or '97 calibration equation, respectively. The SEP of '97 crop year samples analyzed by calibration equation derived from '96 plus '97 crop year samples was more accurate than that of '97 crop year samples analyzed by '96 calibration equation. To improve the analytical inaccuracy caused by the difference of crop year between calibration and prediction samples, we need to include the prediction sample spectra which were different from calibration sample spectra in recalibration sample spectra, and then develop recalibration equation. The NIRS can apply to analyze burley leaf tobacco, leaf process or tobacco manufacturing process which were required the rapid analytical result.

        Cisplatin과 Paclitaxel (Taxol)의 투여순서가 NIHOVCAR-3 세포주의 세포주기와 세포사에 미치는 영향

        목적 : 난소암 세포주에 cisplatin과 taxol을 병합 투여하는 경우에 투여 순서에 따른 세포주기 교란과 apoptosis의 발생 및 세포독성의 차이를 관찰함으로써 항암 효과를 향상시킬 수 있는 투여 순서를 알아보고자 한다. 연구방법 : RPMI 1640으로 배양한 NIHOVCAR-3 세포주에 30㎍/㎖ 의 cisplatin을 먼저 투여 후 1시간 배양하고 이어서 100nM의 taxol을 투여하고 24시간 동안 배양한 A군과, 반대로 100 nM의 taxol을 투여하고 24시간 동안 배양한 후 이어서 30㎍/㎖의 cisplatin을 투여 후 1시간 배양한 B군으로 나누어 실험을 진행하였다. A군과 B군에서 각각 투여 전부터 98시간까지 계획된 시간에 각각의 flask들로부터 세포들을 수확하여 propidium iodide로 DNA를 염색한 후 flow cytometry를 이용하여 세포주기 분기의 변화, apoptosis 및 좌멸세포 (cellular debris)의 출현 정도를 관찰하였으며, 이와 동시에 trypan blue에 의한 염색으로 사멸세포(dead cell)의 백분율을 구하였다. 결과 : Cisplatin을 taxol보다 먼저 투여한 경우에는 초기에는 세포들이 cisplatin에 의해 주로 S 분기에 정지된 후 이어서 taxol에 의해 G2M 분기에 정지되었으며, 반면 taxol을 cisplatin보다 먼저 투여한 경우에는 대부분의 세포들이 S 분기에 정지되지 않고 G2M 분기에 정지되었다. Apoptotic peak은 cisplatin을 먼저 투여한 경우 보다 taxol을 먼저 투여한 경우에 조기에 출현하였으며 장시간에 걸쳐서 지속적으로 관찰되었다. 좌멸세포의 백분율도 taxol을 먼저 투여한 경우가 cisplatin을 먼저 투여한 경우에 비하여 증가되었다. 결론 : cisplatin과 taxol의 병합투여시에 taxol을 cisplatin보다 먼저 투여하는 것이 항암효과의 향상을 이룩할 수 있을 것으로 생각되었다. The overt effects of the anticancer drugs such as cisplatin and taxol appear to be DNA modification and microtubule stabilization respectively. But the mechanism by which these drugs affect tumor cell cycle perturbation and their correlation to apoptosis and cytotoxicity are not well understood, especially in combined sequential treatment of cisplatin and paclitaxel (taxol). In this study, to elucidate the action mechanisms as a function of cell cycle changes and cytotoxicities and to determine the adequate treatment sequence of cisplatin and taxol to acquire more enhanced cytotoxic effects when they are combined, we evaluated the cell cycle perturbations and its correlation to cytotoxic effects, which is measured by the extents of apoptosis and the fractions of cellular debris and live cells after combination treatment of cisplatin and taxol changing their treatment sequences in NIHOVCAR-3 ovarian cancer cell line. Our results were as follows; (1) The accumulation in S phase inhibited the entrance of tumor cells to G2M phase when the cisplatin treatment was preceded to taxol in their combination. (2) The tumor cells were not accumulated in S phase but most of them entered to and accumulated in G2M phase and they were leading to cell death when the taxol treatment was preceded to cisplatin in their combination. (3) Apoptotic peaks in taxol pretreatment group were detected earlier and persisted longer than that of cisplatin pretreatment group. (4) The cytotoxicities represented by the decreased fractions of live cells and the increased fractions of cellular debris were higher in taxol pretreatment group than those of cisplatin pretreatment group. These results suggested that the taxol pretreatment is more effective in combination of cisplatin and taxol and the relative decrease in the cytotoxicity in cisplatin pretreatment group was considered to be derived from the inhibition of entrance of tumor cells to G2M and protected them from the action by taxol. From these results, we concluded that the taxol pretreatment will enhance the cytotoxic effects to tumor cells when cisplatin and taxol will be administered and it indicates that correlations between cell cycle perturbation, apoptosis and cell death have to be considered in the future combination treatment of other drugs and in the development of new treatment regimens.

        리기다소나무의 수용추출액이 무 종자의 발아과정에서 단백질과 동위효소 패턴에 미치는 영향

        Aqueous extracts of Pinus rigida changed the electrophoretic patterns of total proteins and of hydrolytic enzymes such as peroxidase, esterase and amylase during the germination of radish (Raphanus sativus var. hortensis for. acanthiformis). When the extract treatment was finished, at the late stage of radish germination, aqueous extracts of P. rigida had suppressed the expression of 24 KD and 60 KD proteins. the extract induced new isozyme bands, indicating concomitant activity of peroxidases, esterase activities were stimulated in the cathodic region. The activity of amylase was enhanced by the extract.

        Pinus rigida Allelochemicals가 차풀종자의 발아과정에서 동위효소의 활성에 미치는 영향

        Eleven phenolic compounds including caffeic acid were identified through analyzing the aqueous extracts of Pinus rigida by HPLC. Among them, protocatechuic acid was the maximum amount of 6.84 ppm. Seed germination of Cassia mimosoides var. nomame was significantly stimulated by the extract of P. rigida leaves in the proportion ot concentration. However, root growth was elevalted at a threshold concentration below 25%, but it was inhibited at high concentrations. In 50% extract of P. rigida, upward root tip of C. mimosoides var. nomame showed negageotropism which the root end showed necrosis. New isozyme bands were induced indicating concentration activity of peroxidase from the extract of C. mimosoides var. nomame, especially in the cathodic region. Although it reduced the mumber of isozyme bands of esterase, esterase activities were stimulated in the anodic region of C. mimosoides var. nomame. The activity of amylase was not remarkably different between control and treatment.

      • 버어리종 열처리에 의한 수분과 화학성분의 상관성 분석

        This study was carried out to investigate relationship between contents of moisture and chemical components in burley tobacco after heat treatment. Initial moisture contents of burley tobacco was 32% after cutting and casing. Burely cut tobaccos were treated at various temperature(10$0^{\circ}C$, 13$0^{\circ}C$, 145$^{\circ}C$ and 16$0^{\circ}C$) and time(0, 5, 10, 15, 20 and 30 min) in mechanical convection oven. Moisture contents of burley tobacco after heat treatment were in the range of 1.9% to 18.4%. Relationship between moisture contents and total sugar, pH, total volatile base, ammonial, L value of color, nicotine, nornicotine, and $\beta$-nicotyrine contents were positively correlated. Correlation between moisture contents and crude ash, ether extracts, 2,6-deoxyfructosazine, 2,5-deoxyfructosazine, a value of color, citric acid, m-xylene, ${\gamma}$-butyrolactone and 5-methyl-2-furfural were negative.
