        만성 알코올 유발 마우스 간손상 및 지방 축적에 대한 제주조릿대잎 에틸 아세테이트 분획물의 간 보호 효과

        Sasa (S.) quelpaertensis Nakai (Korean name, Jeju-Joritdae), which has anti-oxidative and anti-inflammatory activities, is a type of bamboo grass distributed widely in Jeju Island, Korea. S. quelpaertensis leaves are used for therapeutic purposes in traditional Korean medicine. This study examined the hepatoprotective effects of the S. quelpaertensis ethyl acetate fraction (SQEA) in a mouse model to mimic alcoholic liver damage. The mice were administered orally with 30% alcohol (5 g/kg) once per day with or without SQEA treatments (100 and 200 mg/kg) for 14 days consecutively. Alcohol consumption increased the serum alcohol content and histopathological changes but reduced the liver weight. Moreover, the livers of the alcohol group exhibited the accumulation of malondialdehyde and cytochrome P450 2E1 (CYP2E1), and lipid droplet coating protein perilipin-2. On the other hand, SQEA dose-dependently attenuated the alcohol-induced serum ethanol content and liver histopathological changes but increased the liver weight. Moreover, SQEA attenuated the level of CYP2E1 and inhibited alcohol-induced lipogenesis in the liver via decreased perilipin-2 expression. These results suggest that SQEA can provide a potent way to reduce the liver damage caused by alcohol consumption.

        Consumption of Jeju Ground Water Containing Vanadium Components Enhances Hepatic Antioxidant Defense Systems in ob/ob Mice

        비만 마우스의 산화적 스트레스에 대해 바나듐 함유 제주 지하수에 대한 연구로서, 제주 지하수는 수도수 처리군보다 비만 마우스 간의 과산화 지질을 감소시켰으며, superoxide dismutase, catalase, 그리고 glutathione peroxidase의 단백질 발현 및 활성 그리고 환원형 glutathione 양을 증가 시켰다. 제주 지하수는 항산화 효소의 전사인자인 erythroid transcription factor NF-E2 (Nrf2)의 인산화형 단백질 발현을 증가시켰다. 이로서 제주 지하수는 Nrf2의 활성화를 통하여 항산화 효소시스템을 증가시켰다. The present study examined the effects of consumption of Jeju ground water containing vanadium components on oxidative stress in obese (ob/ob) mice. Intake of Jeju ground water decreased the generation of oxidative stress induced-lipid peroxidation in the liver of ob/ob mice It also enhanced the enzymatic antioxidant defense system by increasing the protein expression and activity of superoxide dismutase, catalase, and glutathione peroxidase in liver tissues. Jeju ground water intake also upregulated the intracellular content of reduced glutathione. The induction of antioxidant enzyme expression by consumption of Jeju ground water was mediated by the erythroid transcription factor NF-E2 (Nrf2). Increased nuclear expression of phospho Nrf2 was observed in ob/ob mouse liver cells following intake of Jeju ground water. These results suggest that consumption of Jeju ground water stimulated the antioxidant defense system in the livers of ob/ob mice via induction of Nrf2.

        방사선을 조사한 마우스에서 비장세포에 대한 톳의 보호 작용

        The immune system is specifically sensitive to oxidative stress induced by ionizing radiation because of its rapid proliferative activity. For this reason, an instructive immune system is one of the best ways to minimize side effects, such immunodeficiency, of gamma radiation. Over the past few decades, several natural plants with antioxidant and immunomodulatory properties have been identified as adjuncts for nontoxic and successful radiotherapy. Hizikia fusiforme extract (HFE) containing plentiful dietary fiber and fucoidan is known for its instructive antioxidant capacity, immunomodulation abilities, and immune activation. In this study, we determined whether HFE protects radiosensitive immune cells from gamma radiation-induced damage. C57BL/6 mice were irradiated with gamma-ray. The effect of HFE on the ionizing radiation damage of immune cells was then evaluated with an MTT assay, 3H-thymidine incorporation assay, and PI staining. We found that HFE stimulated the proliferation of gamma-ray irradiated immune cells without cytotoxic effects. We also observed that HFE not only decreased DNA damage but also reduced gamma radiation-induced apoptosis of the immune cells. Our results suggest that HFE can protect immune cells from gamma-ray damage and may serve as an effective, non-toxic radioprotective agent.

        방사선으로 인한 산화적 손상에서 phloroglucinol의 모낭 보호 효과

        When exposed to gamma-rays, hair follicular cells immediately go through apoptosis, which hampers their rapid differentiation essential for the regeneration of hair. Phloroglucinol (PG) is a phenolic compound of Ecklonia cava, brown algae abundant in Jeju island, Korea. Containing plentiful polyphenols, PG is known for its instructive effects by inhibiting apoptosis, scavenging oxygen radicals, and protecting cells against oxidative stress. In this study, we demonstrate that PG rescues radiosensitive hair follicular cells from gamma radiation-induced apoptosis and DNA damage. To identify protective capacity of PG on hair follicles, we irradiated with 8.5 Gy (1.5 Gy/min) of gamma-rays to the whole body of C57BL/6 mice at day 6 after depilation with or without PG. In mice exposed to radiation, the expression of proapoptotic molecule p53 was downregulated in the skin of PG treated group. On immunohistochemical observation of the skin, PG inhibited the immunoreactivity of p53 and cleaved caspase-3. PG treatment protected hair follicular cells from cell death due to gamma-radiation. Our results suggest that PG presents radioprotective effects by inhibiting apoptosis of radiosensitive hair follicular cells and can protect hair follicular cells from gamma-ray induced damage.

        당유자 과피 추출물에 의한 HL60 세포의 Apoptosis 유도

        당유자 과피(GGP) 80% 에탄올 추출물을 4종의 암세포에(피부암, 대장암, 유방암 및 혈액암) 처리하여 증식 억제 활성을 측정한 결과, 혈액암 HL60 세포에서 높은 증식 억제 활성을 보였다. 이에 CGP 추출물이 HL60 세포에 대한 apoptosis 유도에 따른 세포 증식 억제 활성을 조사하였다. Apoptosis 유도의 첫 단계인 막 투과성을 측정한 결과, confocal image와 flow cytometry에서 CGP를 처리하였을 때 탈분극 현상에 따른 막 투과성이 증가하였고 세포내 핵을 hoechst 33342를 이용하여 염색하였을 때 apoptosis가 일어났을 때 나타나는 전형적인 형태의 apoptotic body가 농도 의존적으로 증가하는 것을 확인할 수 있었으며 flow cytometry를 통하여 세포 주기를 측정하였을 때 DNA-hypodiploid 형태의 sub-G1가 CGP 농도 의존적으로 증가하는 것을 확인할 수 있었다. Apoptosis 유도 기전을 western blot으로 측정한 결과를 보면, CGP 추출물을 혈액암 HL60 세포에 처리하였을 때 Bcl family의 anti-apoptotic Bcl-2 단백질의 감소와 pro-apoptotic Bax 단백질의 증가로 인하여 하위기전인 caspase-3가 활성화되었으며, 이 활성화로 인하여 apoptosis 유도에 직접적으로 관여하는 PARP 단백질을 활성화시키면서 apoptosis를 유도하였다. 따라서 당유자 과피는 항암과 관련되어진 기능성식품 및 소재 개발 원료로서 개발이 가능하리라고 사료된다. In the present study, we investigated the anti-proliferation activity of Citrus grandis Osbeck peel (CGP) in HL60 (human promyelocytic leukemia) cells. It was found that 80% ethanol extract of CGP could inhibit the cell growth in a dose-dependent manner (250~1,000 ㎍/mL), which was associated with morphological changes and apoptotic cell death such as depolarized mitochondrial membrane, formation of apoptotic bodies and increased populations of apoptotic sub-G1 phase. The results indicate that CGP extract inhibits the growth of HL60 cancer cells by the induction of apoptosis, which may be mediated by its ability to change the Bcl family proteins and increase the activation of caspase-3 and PARP. Therefore, it is suggested that CGP has the potential to provide a remarkable natural defense against the proliferation of HL60 cells.

        김대중 정권의 ‘대북 포용 정책’과 ‘북일 관계 개선’의 상관 관계

        The Koizumi regime and the Kim Jong-il regime, which pursued negotiations to normalize diplomatic relations, achieved some success despite structural obstacles including the Bush administration’s opposition to improving the relations. How did they pursue the normalization negotiations? It is also necessary to analyze the positions they were taking in the negotiations. This study found that despite the structural obstacles to the improvement of the North Korean relations by the Bush administration, it was possible to make some achievements in the negotiations between Japan and North Korea by establishing the Korea - Japan cooperation system. The involvement of the Kim Dae-Jung administration acted as an important factor in the promotion of Japan-North Korea negotiations. In particular, at the Japan-ROK Summit, President Kim Dae-Jung recommended Prime Minister Koizumi Junichiro to hold the Japan-North Korea Summit Meeting and sent the envoy to North Korea’s Chairman of the National Defense Commission Kim Jong-Il. Such encouragement from the Kim Dae-Jung administration was a crucial factor in the progress of Japan - North Korea negotiations. In other words, this involvement of the Kim Dae-jung regime influenced the fundamental factors in the progress of the negotiations. Those are Japan’s recognition of negotiations with North Korea and the recognition of North Korea’s policies and negotiations with Japan. US policy toward North Korea has been influential on the North Korea policy of South Korea and Japan as a structural factor, but by establishing the Korea - Japan cooperation system, Korea and Japan could pursue different policy positions from the US policy.

        아베 정권의 개헌 정책 결정 메커니즘

        본 연구는 1950년대 후반의 하토야마 정권과 기시 정권 때만해도 ‘개헌 및 재군비’에 냉담했던 여론과 정치계가 어떠한 과정을 거쳐 변했으며, 수십 년간 이어져온 ‘보수 본류’를 누르고 ‘전후 총결산’과 같은 논조의 정책을 추진할 수 있었는가에 대한 문제 의식에서 출발하였다. 분석 결과, 정권과 관료에 의해 규정된 정책 실패는 변화를 넘어 개혁의 필요성과 위기 인식을 공유하는 계기가 되었다는 것을 알 수 있었다. 특히 1990년의 걸프전 사례는 일본의 외교 실패로 규정되었고, 이러한 사태는 ‘위기’로 인식되었다. 그리고 이런 과정을 통해 일본의 안보 정책에 새로운 전환점을 맞이했다. 즉, 안보 정책의 변화를 꾀하던 각 정권은 걸프전 쇼크를 내세워 ‘정치적 정당성’을 확보했고, 이를 통해 평화유지활동(PKO) 협력법이나 자위대 파견과 같은 안건을 보다 수월하게 처리하였으며 정책 결정 과정에서 ‘절차적 정당성’도 확보했다. 이와 더불어 아베 정권은 ‘개헌’과 관련된 정책을 추진하는데 있어서 세이와 정책 연구회와 같은 조직화된 ‘공부 모임’을 활용했다. 특히, 이를 통해 정책과 관련된 정보와 추구하는 가치를 공유 및 확산하는 과정들 속에서 정계 지지 기반을 확보할 수 있었다. 정리하면, 장기간에 걸쳐 규정되고 공유된 시대 인식과 가치의 확산을 통해 확보한 정치적·절차적 정당성을 바탕으로 일본의 개헌과 관련된 정책 및 안보 체제는 변해왔다. In this study, I will analyze how the opinions of public and political circles, who bitterly opposed amending the constitution and rearmament during the regimes of Hatoyama and Kishi in the late 1950s, have changed and through what process. As a result of the analysis, the policy failures prescribed by the government and bureaucrats became an opportunity to share the necessity for reform following a shared perception of crisis. In particular, the case of the Gulf War in 1990 was defined as a diplomatic failure on the part of Japan, and this situation was recognized as a “crisis” in Japan. Through this process, a new turning point was reached in Japan’s security policy. In other words, each Japanese government that was trying to change its security policy secured the political legitimacy by using the Persian Gulf Crisis as an example of a failure in Japan. Using this mechanism, it has handled matters such as the Act on Cooperation with United Nations Peacekeeping Operations and Other Operations and the dispatch of the Self-Defense Forces with greater ease, as well as the policy decision process. ‘Procedural justification’ was also secured. In addition, securing political legitimacy either helped form a new organization and/or strengthened the organization"s cohesion. On the other hand, the recognition of the era which was defined and shared by the Japanese government greatly helped to secure political legitimacy in the political world. In particular, the Abe administration was able to secure a foundation for political support in the process of sharing and spreading information and values through an organized study group such as the Seiwaken in promoting policies related to the constitutional reform. In summary, the spread of recognition and values of the era defined and shared over a long period of time helped to more easily change the policy of specific regimes.

        금융시장 안정을 고려한 금융회사 지배구조의 개선

        The corporate governance of a financial company should be designed not only based on the financial soundness of the firm and the protection of its shareholders and other stake-holders but on the stability of financial markets. The financial crisis in 2008 well illustrates that if the corporate governance of a financial company is not appropriate to control the management’s rent seeking, it may put the long-term growth of the firm and the interests of its shareholders and stake-holders into trouble and further lead to a crisis in financial markets. In this context, major countries have made efforts to renovate the corporate governance of financial companies in the light of the protection of the interest of shareholders and stake-holders as well as its influence on the financial market stability. This essay, in this regard, examines whether the current laws effectively control the rent seeking by the management and take good account of its potential influences on the financial market stability. Currently, the commercial law does not impose special restrictions on financial firms, but under the commercial law listed companies are subject to strengthened conditions in the case of independent directors and an auditing committee. And these conditions shall be applied to the financial companies which means they are under stricter controls than others. The recently enacted law on corporate governance is considered to be well corresponding to global standards. However, the independency of the board of directors, remunerating system of the management, internal control, and risk management should be improved further for the financial market stability. In the consideration of the influences of financial companies on financial market stability, the corporate governance of financial companies should be able to supervise rent seeking by the management with efficiency and effectiveness. To this end, it is critical to ensure transparent and independent work of supervision on corporate management activities. For transparency the scope of responsibility of among internal inspection agencies should be clarified and for independency from the clout of the management and dominant stockholders should be secured during the nominating process of directors. More specifically, first the board of directors shall be responsible for comprehensive decision-making in supervising the management and related risk management. Second, a remunerating system should be associated with longer term achievement, thus not alluring short-term rent seeking by the management. Third, an auditing committee should secure higher independency in their work of audit and accounting. Four, compliance officers who are responsible for the observance of internal rules of a company should perform their duties by clear standards. Five, the decision-making by the management in regard of risk management should be checked and balanced by a risk management committee.
