To find antitumor components from higher fungi, the mycelia of Collybia confluens (Pers. ex Fr.) Kummer were cultured in artificial media. For efficient production of the mycelia, the influences of various modifications of culture conditions were examined. A water-soluble protein-bound polysaccharide fraction, Fr. A, was obtained from the mycelia by hot water extraction. When Fr. A was purified and fractionated by DEAE-cellulose and Sephadex G-200 gel filtration chromatographies into four fractions which were designated B, C, C-I and C-II. The tumor inhibition ratios of these fractions ranged from 46% to 75%, against the solid forms of sarcoma 180 in ICR mice at doses of 20 and 50 ㎎/㎏/day when given intraperitoneally. Especially, Fr. C which was named Collyban(CB) exhibited a marked life-prolonging effect of the mice against ascitic forms of sarcoma 180 at a dose of 50 ㎎ via i.p. administration. To extend spectra of the antitumor activities and eliminate the effects of allograft rejection, the characterization of antitumor effects of CB was performed in syngeneic host-tumor systems. It did not show any antitumor activity against L1210 murine leukemia in CD2F1 mice but prolonged their life span against ascitic forms of MM46 carcinoma in C3H/He mice. Also it exhibited antitumor activity against human cervical cancer HeLa cells that were xenografted into nude mice having BALB/c genetic backgrounds by the i.p. injection at a dose of 100 ㎎/㎏/day. In order to characterize the antitumor components, CB was examined by chemical analysis. It was acidic protein-bound polysaccharides composed of 31% polysaccharide, 27% protein and 3% hexosamine. CB was fractionated into two fractions, Fr. C-I(M.W.: 500 Kd) and Fr. C-II (M. W.: 30 and 8 Kd) by Sephadex G-200 gel filtration chromatography.

In order to detect poisonous or physiologically active constituents in the Higher Fungi growing in Korea, fortythree species including two varieties were collected in Kyung Ki and Chung Cheong Buk Do Provinces and subjected to systematic extraction with two kinds of organic solvents. The extracts were examined for alkaloid by adding alkaloid reagents. Of these fungi, three species, Geaster fimbriatus, Lampteromyces japonicus and Pseudocolus schellenbergiae, gave positive confirmatory tests.

A protein-polysaccaride fraction Kp(53.9% polysaccharide, 14.2% protein) was separated from the shake-cultured mycelia of a basidiomycetous fungus, Phellinus linteus, and its antitumor activity against sarcoma 180 in ICR mice was investigated. When administered after the tumor implantation, Kp exerted antitumor activity by inhibiting the growth of the sarcoma 180 solid tumor by 71.5% and increasing the life span of the sarcoma 180 ascitic mice by 51.5% at 100 mg/kg. In pretreatment tests, in which Kp was administered once daily for 9 days before the tumor implantation, Kp inhibited the growth of the solid and ascites form of sarcoma 180, respectively, by 35.4% and by 80.3% at 100 mg/kg. However, Kp showed no in vitro cytotoxic activity against a murine leukemia L1210 and a human gastric tumor SNU.1 upto the concentration of 200 mcg/ml. From these results, it is clear that the antitumor activity of Kp is exerted through its immunomodulating activity on the host''s immune system.

Bifidobacterium bifidum, one strain of medical preparations being on the market for human intestinal disorder, is very sensitive to rifampicin. If this preparation is taken with rifampicin, its therapeutic effect can't be expected. To develope rifampicin resistant mutants, B. bifidum was treated with N-methyl-N''-nitro-N-nitrosoguanidine(MNNG). All of thirty strains grown on the plates containing 10 mcg/ml rifampicin were over 1,000 times more resistant to rifampicin than parental strain and they were identified as B. bifidum by fructose-6-phosphoate phosphoketolase test. Three strains out of thirty, which produced almost same amount of organic acid as parental strain, were selected for further studies. They showed identical growth inhibition activity aganist E. coli compared with that of parental strain. And rifampicin was not inactivated.

To find antitumor components, a protein-bound polysaccharide fraction was obtained by extracting with hot water and precipitating with ethanol from the carpophores and the cultured mycelia of Laccaria laccata. The fraction was examined for antitumor activity against sarcoma 180 implanted in mice. The component extracted from the carpophores showed 75% and 60% tumor inhibition ratios in the doses of 20㎎ and 50㎎/㎏/day, respectively. The protein-bound polysaccharide fraction of the cultured mycelia of L. laccata also showed 57.8% tumor inhibition ratio in the dose of 20㎎/㎏/day. The chemical analysis of the protein-bound polysaccharide fraction showed that it contained a polysaccharide and a protein. The hydrolysates of the polysaccharide moiety contained six and one unknown monosaccharides. Fourteen amino acids were identified in the hydrolysates of the protein moiety.
알파-아밀라제 저해제 생산 방선균 Streptomyces minoensis DMCJ-144 의 균주개량

Strain of Streptomyces minoensis DMCJ-144 was tried to be improved so that it produces much more the α-amylase inhibitor. Streptomyces minoensis DMCJ-144 was treated with 1 ㎎/㎖ (pH 9.0) of N-methyl-N'-nitro-N-nitrosoguanidine at 30℃ for 60 min and irradiated with UV light distanced 30 cm for 20 min. After mutagenesis, surviving colonies were cultured on the CM contaning acriflavine (10㎍/㎖) three times in order to enhance the mutability. And then through multi-level screening, colonies that α-amylase inhibitor productibility was improved were selected by modified-blue value method. After third acriflavine treatment, α-amylase inhibitory activities of selected colonies were found to be much better as compared with that of parent strain. One mutant strain showed 5.4 time inhibitory activity than the parent strain.
