Gram-positive bacteria, Bacillus subtilis BR151 and Staphylococcus aureus RN4220 were transformed with high efficiency by electroporation. The cells were incubated until late log phase, washed three times with 10% glycerol, 1mM HEPES, 12% sucrose and resuspended to $10^{10}{\sim}10^{11}cfu/ml$, then stored at -$70^{\circ}C$. Transformation efficiency of B. subtilis BR151 was $1.03{\times}10^7cfu/{\mu}g$ with cells washed with 10% glycerol and electroporated by 15KV/cm, 0.7msec pulse with pUB110. Transformation efficiency of S. aureus RN4220 was $4{\times}10^6cfu/{\mu}g$ with cells washed with 1mM HEPES + 10% glycerol and electroporated by 15KV/cm, 2.5msec pulse. The number of total transformants was 1000 when B. subtilis BRI51 was transformed with 100ng pUB110 DNA and the number of total transformants was 9000 when S. aureus RN4220 was transformed with 10ng pUB110 DNA

Clinically isolated bacterial strains resistant to almost of all the clinically superior beta-lactam antibiotics can be used to screen the promising ones among the newly synthesized beta-lactam antibiotics. To select the resistant strains, the susceptibility of 389 strains of S. aureus, 144 strains of coagulase negative staphylococci, 509 strains of E. coli, 115 strains of E. cloacae and 187 strains of P. aeruginosa to methicillin, ampicillin, piperacillin and gentamicin was determined. The susceptibility of 19 bacterial strains selected through the first screening to cefixime, cefotiam, cefotaxime, flomoxef, cepfirome, cefdnir, SCE-2787, panipenem and imipenem was determined. Four strains of S. aureus finally selected have high degree of resistance to almost of all bata-lactam antibiotics used and also produce beta-lactamases. These 4 strains of S. aurues can be used to screen effectively the promising beta-lactam antibiotics among the numerous numbers of the newly synthesized beta-lactam antibiotics.

To find active components in Ganoderma Iucidum. their free amino acid and free amino acid derivatives were analyzed. After extracting with hot water, the extracts were filtrated by three steps. So, supernatants below 10,000 dalton were obtained. Filtrates were derivativated with PITC (phenylthiocarbamyl) derivative reagent and PITC amino acid was obtained. Then, they were analyzed by RP-HPLC. 13 amino acids were analyzed in cultured Korean Nok-kak ji (one of Ganoderma Iucidum), 15 amino acids in cultured Korean Ganoderma Iucidum, 12 amino acids wild Ganoderma Iucidum, 16 amino acids in cultured Taiwan Ganodenna Iucidum.

To find amylase inhibitors produced by microorganisms from soils, a strain which had a strong inhibitory activity against bacterial α-amylase was isolated from the soil sample collected in Seoul. The morphological and physiological characteristics of this strain on several media and its utilization of carban sources showed that it was one of Streptomyces species according to the International Streptomyces Project method. The amylase inhibitor of this strain was purified by means of acetone precipitation, adsorption on Amberlite XAD-2, and column chromatography on Amberlite CG-50 and SP-sephadex C-25. The inhibitor was stable at the pH range of 1-10 and at 100℃ for half an hour, and had inhibitory activities against other amylases such as salivary α-amylase, pancreatic α-amylase, fungal α-amylase and glucoamylase.The kinetic studies of the inhibitor showed that its inhibitory effect on starch hydrolysis by α-amylase was non-competitive.

It has been known that Ganoderma lucidum and Lentinus edodes have anticancer activity and immune enhancing activity. These two mushrooms were grown in liquid culture and harvested. From these<br/> mycelia, DNA was isolated and EtBr-CsCl density gradient ultracentrifugation was performed to purify it further. Then mitochondrial DNA was isolated by bisbenzimide-CsCl density ultracentrifugaton. Mitochondrial DNA of Ganoderma lucidum was digested by restriction enzymes, EcoR I, Hind III, and Pst I, then electrophoresed. It showed 12, 22, 4 fragments. Mitochondrial DNA of Lentinus edodes was digested by EcoR I. Electric pattern showed 6 fragments. 4 fragments had appeared by Pst 1 digested mitochondrial DNA. Hind III couldn't digest mitochondrial DNA of Lentinus edodes. Mitochondrial DNA of fusants was isolated to compare to those of parents. The results showed that fusant P₂S₂ has new, recombined mitochondrial DNA. But P₂S₂ had the same<br/> DNA that Ganoderma lucidum had.
새로운 퀴놀론 항균제 DW-116의 Post-Antibiotic Effect

The post-antibiotic effects (PAE) of DW-116 were evaluated against Bacillus subtilis ATCC 6633, Bacillus cereus ATCC 27348, Staphylococcus aureus ATCC 25923, Escherichia coli ATCC 25922 and Pseudomonas aeruginosa MB4-16, repectively. Against gram-positive bacteria, PAEs of DW-116 were longer duration (20-35 min) than those of rufloxacin(1O-20 min), and shorter than those of ciprofloxacin (50-90 min). Especially, against E. coli, DW-116 and ciprofloxacin obtained approximately 3 hrs of PAES.
