        환경스트레스 처리에 의한 개똥쑥 artemisinin 생합성 증진

        Artemisinin is a secondary metabolite of Artemisia annua that shows potent anti-malarial, anti-bacterial, antiviral, and anti-tumor effects. The supply of artemisinin depends on its content in Artemisia annua, in which various environmental factors can affect the plant’s biosynthetic yield. In this study, the effects of different light-emitting diode (LED)-irradiation conditions were tested to optimize the germination and growth of Artemisia annua for the enhanced production of artemisinin. Specifically, the ratio between the red and blue lights in the irradiating LED was varied for investigation as follows: [Red : Blue] = [6 : 4], [7 : 3], and [8 : 2]. Furthermore, additional stress factors like UV-Birradiation (1,395 µW/cm2 ), low temperature (4°C), and dehydration were also explored to induce hormetic expressions of ADS, CYP, and ALDH1, which are essential genes for the biosynthesis of artemisinin. Quantitative polymerase chain reaction (qPCR) was used to analyze the expression levels of the respective genes and their correlation with the specified conditions. [8 : 2] LED-irradiation was the most optimal among the tested conditions for the cultivation of Artemisia annua in terms of both fresh and dry weights post-harvest. For the production of artemisinin, however, [7 : 3] LEDirradiation with dehydration for six hours pre-harvest was the most optimal condition by inducing around twofold enhancement in the biosynthetic yield of artemisinin. As expected, a correlation was observed between the expression levels of the genes and the contents of artemisinin accumulated.

        류마티스 관절염에 있어 종양괴사인자 다형성에 대한 연구

        Objectives : Tumor necrosis factor-${\alpha}{\cdot}$(TNF-${\alpha}{\cdot}$) is a proinflammatory cytokine involved in the pathogenesis of rheumatoid arthritis. This study was designed to investigate the relation between TNF-${\alpha}{\cdot}$ gene polymorphism and rheumatoid arthritis in Korean population. Methods : This study was carried out on 103 rheumatoid arthritis patients who fulfilled the American College of Rheumatology 1987 revised criteria for rheumatoid arthritis and 208 healthy control subjects. Blood samples from all subjects were obtained for DNA extraction. The extracted DNA was amplified by polymerse chain reaction(PCR). PCR products were visualized by 2% agarose gel electrophoresis. We investigated the genotyping of TNF-${\alpha}{\cdot}$ by using Pyrosequencing. Results : The genotypes of TNF-${\alpha}{\cdot}$ gene were GG, AG and AA. While the distribution of TNF-${\alpha}{\cdot}$ polymorphism in control subjects was 92.31%, 7.21%, 0.48% respectively, in rheumatoid arthritis patients was 93.20%, 6.80%, 0.00%(GG, AG. AA). There was no statistical significant allelic frequency difference between control and rheumatoid arthritis groups. Conclusion : We concluded that there was no significant association between TNF-${\alpha}{\cdot}$ gene polymorphism and rheumatoid arthritis. However, the findings of this study need to be confirmed in more patients and further studies.

        친사회적 조력자 모델을 활용한 데이트 폭력 예방 프로그램 효과에 관한 연구

        In some countries, such as the United States, crime prevention programs that emphasize the social responsibility of bystanders to prevent dating violence crimes and induce change in their attitudes and helpful behaviors for crime prevention are being actively conducted. They has theoretical foundations such as prosocial behavior theory, community readiness model, and elaboration likelihood model. Bystanders can be said to be helpers to prevent crime, and according to previous studies, bystanders' intervention plays a decisive role in crime prevention and damage recovery. When bystanders' involvement increases, social norms against crime are strengthened and the community's capacity increases, contributing to the reduction of dating violence crimes. This study verified whether a crime prevention program that is being conducted abroad is effective in Korea, targeting college students. After randomly selecting 95 university students from three universities located in Gyeonggi-do, dividing them into a treatment group and a control group, a crime prevention program was conducted for the treatment group. As a result, students who participated in the crime prevention program increased knowledge related to dating violence, decreased acceptance of rape myths affecting dating violence crimes, and increased bystander attitudes to help prevent crimes. Based on previous studies that knowledge and attitude predict behavior, it can be predicted that a crime prevention program that promotes the intervention of a bystander will be effective in preventing actual crime. 미국 등 일부 국가에서는 데이트 폭력 범죄 예방을 위해 가해자 또는 피해자 주변인들의 사회적 책임감을 강조하며 범죄 예방을 위한 태도 변화와 도움 행동을 유도하는 범죄예방 프로그램이 활발하게 진행되고 있다. 친사회적 행동 이론, 지역사회 변화모델, 정교화 가능성 모델 등 이론적 기반을 두고 있다. 주변인들은 범죄를 막기 위한 조력자라고 할 수 있는데, 선행연구에 의하면 조력자의 개입은 범죄예방과 피해회복에 결정적 역할을 한다. 구성원들의 개입이 증가하면 범죄에 대한 사회적 규범이 강화되고 공동체의 역량도 높아져 데이트 폭력 범죄 감소에 기여할 수 있다. 본 연구는 외국에서 진행되고 있는 범죄예방 프로그램이 국내에서도 효과가 있는지 대학생들을 대상으로 검증하였다. 경기도에 소재한 3개 대학 95명 대학생들을 무작위로 선정하여 처치집단과 통제집단으로 나눈 후 처치집단을 대상으로 범죄예방 프로그램을 진행하였다. 그 결과 범죄 예방 프로그램에 참여한 학생들은 데이트 폭력과 관련된 지식이 증가했고, 데이트 폭력 범죄에 영향을 미치는 강간 통념 수용도가 낮아졌고, 범죄를 예방하기 위해 도움을 주고자 하는 조력자 태도가 증가했다. 지식과 태도가 행동을 예측한다는 선행연구에 기초하면 조력자의 개입을 촉진하는 범죄 예방 프로그램이 실제 범죄예방에 효과적일 것으로 예측할 수 있다.

      • Expression of Human Galβ1 , 3GalNAcα2 , 3-Sialyltransferase (hST3Gal II) in E. coli and Production of Polyclonal Antibody Using Recombinant Protein

        Gal β 1,3GalNAc α 2,3-sialyltransferase (ST3Gal I and II) is the enzyme that transfers CMP-NeuAc with an α 2,3-linkage to a galactose residue O-linked to a threonine/serine of a glycoprotein, and to the terminal galactose residues of glycolipids. Compared to ST3Gal I with the same acceptor substrate specificity, ST3Gal II exhibits much more acceptor substrate preference for glycolipids than for O-linked oligosaccharide of glycoproteins. For expression of human Galβ1,3GalNAc α 2,3-sialyltransferase (hST3Gal II) in E. coli, the expression plasmid (pET-hST3Gal II) was constructed by PCR and cloning into a plasmid (pET-3d) containing a phage T7 RNA polymerase promoter. The recombinant protein expressed by induction with IPTG was accumulated as inclusion bodies, which correspond to molecular mass of approximately 31-kDa on SDS-PAGE, in the insoluble fraction of cells. The isolation and washing of inclusion bodies with Triton X-100 and EDTA yielded a high proportion of the recombinant hST3Gal II with a large increase in purity. To investigate the subcellular localization of ST3Gal 11, polyclonal antibody to the recombinant hST3Gal II (rhST3Gal II) was produced. By immunoblot analysis this antibody showed the specificity for the 48-kDa protein which probably corresponds to ST3Gal II in the Golgi fraction of rat liver. This result shows that ST3Gal II is localized in the Golgi apparatus.

      • Genomic Structure Analysis of Human Galβ1 . 3GalNAc α2 . 3-Sialytransferase (hST3Gal II)

        The human Galβ1,3GalNAcα 2,3-sialyltransferase (hSTSTGal II) genomic gene was isloated from human genomic library. The gene was found to span about 16 kb and to be composed of six exons. The sequences of all the exon/intron splice junctions conformed to GT-AG rule, and the sizes of exon and intron ranged from 46-460 by and 10-6.4 kb, respectively. Exon 1 contained the enrite 5´-untranslated region and the begining of the coding region to amino acid residue 113, including a cytoplasmic domain (6 residues), a hydrophobic signal anchor sequence (21 residues), a stem region (36 residues), and a part of the active domain (50 residues). Exons 2-6 encoded the remaining part of the active domain, and exon 6 contained a translation termination codon and the 3´-untranslated region. Sialylmotif L, a putative nucleotide sugar binding domain, was encoded by exons 2 and 3, and sialylmotif S was encoded by exons 5 and 6. Compared with genomic orgainzations of five other sialyltransferase genes reported so far, the genomic structure of the hST3Gal Il gene was fairly similar to that of the hST3Gal I.

        말기암환자가 보완대체요법을 선택한 이유

        목적: 말기암환자들이 보완대체요법을 선택하게된 주관적 경험을 파악하여, 대상자 입장에서의 당위성을 이해하고 이용과정에서 발생할 수 있는 현상들을 이해함으로써 좀 더 바람직하고 효율적인 이용을 위한 호스피스 완화의료 중재를 하는데 기여하고자 실시하였다. 방법: 심층면담한 내용을 Colaizzi(1976)의 방법을 이용하여 귀납적이고 서술적인 기술과 분석을 실시하였다. 결과: 말기암환자와 가족들이 보완대체요법을 선택한 4가지 범주는 첫째 현대의학의 한계 인식, 둘째 보완대체요법의 효과에 대한 신뢰, 셋째 가족의 정서적 욕구 충족 넷째, 의사의 부정적인 태도로 인한 불신이었다. 주제모음은 9가지로 '불가능한 완치에 대한 기대', '병원치료에 대한 불확실성', '항암치료의 부작용을 해결하기 위한 보조적 수단', '증상완화와 생명연장을 위함', '암치료의 부작용에 대한 두려움', '체험과 정보에 대한 신뢰', '타인의 권유', '가족으로서 도리를 다함', '의사의 부정적 태도에 불만족'이었다. 결론: 암환자의 치료 결정과정에서 대상자와 의사가 바람직한 의사소통과정을 통하여 현대의학 치료에 대한 설명이 충분히 이루어져서 대상자가 일반적인 호스피스 완화의료 서비스를 이용하고 과학적인 근거에 바탕을 둔 보완대체요법에 대한 정확한 정보 제공이 필요하다고 본다. Purpose: The purpose of this study was to analyze the reason to select complementary and alternative therapy for terminally ill cancer patients. Methods: The data were collected from 21 terminal cancer patients and families through the in-depth interview. Data analysis were performed by the Colaizzi's phenomenological method (1976). Results: The reason to select complementary and alternative therapy for terminal cancer patients and families was then categorized with 4 elements; Awareness of limitations in contemporary medical treatments, Belief in effectiveness of the CAM, Satisfaction with emotional needs of family members, and Disbelief due to negative attitudes of physicians. The result indicated the following 9 themes expectation for a complete cure, uncertainty in hospital treatments, complementary method for management of side effect of chemotherapy, alleviation of symptoms and life-sustaining, fear for side effects of cancer treatments, belief in earned information, referrals by other, responsibility of family, and dissatisfaction with negatine attitudes of physicians. Conclusion: Physicians should provide a sufficient explanation and try to effectively communicate with clients about hospice and palliative service and the CAM. We strongly realized that concerns about patients' best care and satisfactions with family's needs should be understood.
