Machine tools that are capable of large-scale complex processing are in high demand. However, the drum-type ATC, currently being used, has difficulty in maintaining rigidity due to the increased weight of the tool. Thus, a ring-type ATC that would provide rigidity while reducing costs is considered. The tool replacement shaft of the cam box, one of the main components of ATC, is greatly affected by the increase in tool weight and load acting in various directions. Thus, its fatigue life should be considered. A life test can be conducted to determine its fatigue life, but because the required life is long, the test takes a considerable amount of time. This problem can be solved by using the accelerated life test, but the parameter data required to perform the test are insufficient. In this study, we present a structural analysis of the cambox shaft, derive the shape parameters required for an accelerated life test, and confirm that the parameters derived using data bagging techniques are accurate.

Objectives: This study was designed to investigate the effects of horse bone powder extract on the growth of longitudinal bone in adolescent male rats. Methods: Longitudinal bone growth was measured by fluorescent microscopy. To examine the effects on the growth plate metabolism, the total height of growth plate, the induction of local insulin-like growth factor-1 (IGF-1), and bone morphogenetic protein-2 (BMP-2) were measured. Results: Horse bone powder extract enhanced longitudinal bone growth and total height of the growth plate. Also, it promoted the induction of local IGF-1 and BMP-2 of the growth plate. Conclusions: This study shows that the horse bone powder extract effects longitudinal bone growth in adolescent rats and might be used for both stunted adolescents and inherent growth failure patients.

In the present study, the anti-cancer activity of 80% ethanol extracts from 120 kinds of medicinal herbs and native plants were investigated. Among them, the barks of Melia azedarach L. var. japonica Makino showed the highest cytotoxicity in HCT-15 human colon cancer cell. With this result, we carried out hollow fiber (HF) assay and anti-metastasis study to confirm the anti-cancer effects of M. azedarach var. japonica. In MTT assay, M. azedarach var. japonica.inhibited the proliferation of HCT-15 cells in dose-dependent manner. HF assay was carried out using A549 human adenocarcinoma cell, HCT-15 and SK-Hep1 human liver cancer cell via intraperitoneal (IP) and subcutaneous (SC) site. As a results, SK-Hep1 implanted in IP site showed the highest cytotoxicity. The result from metastatic model using B16/BL6 mouse corresponded to that of HF assay. These results suggest that the ethanol extract from M. azedarach var. japonica. might have a potent anti-cancer activity and advanced study is needed for the development of novel natural anti-cancer drug.

The acute toxicity of Paecilomyces sinclairii was tested in beagle dogs. We daily examined clinical signs, body weights, and hematological/ biochemical examinations for 14 days after<br/> administration of Paecilomyces sinclairii with different dose levels (0, 0.4, 2 and 10 g/kg). There were no clinical signs and no significant changes in hematological and biochemical analysis. These results showed that Paecilomyces sinclairii did not induce any remarkable acute toxic response and the LD50 was greater than 10 g/kg in beagle dogs.

Objectives : To investigate the effects of DL-HGF to hair growth activity in mouse, various kinds of ethosome and liposome formulations entrapped DL-HGF were produced and this study was carried out. Methods : The B16/BL6 mice were classified into five groups: vehicle control (Con) group, Et-1-applied group, Et-2-applied group, LP-1-applied group, LP-2-applied group. Active hair growth (anagen) was induced in the back skin by application of a waxosin mixture with subsequent depilation and the activity of hair growth was measured by macroscopic observation and histology. Results : In vehicle control group, there was no hair growth activity during experiment period. In Et-1-applied group, the rate of hair growth was about 100%, LP-1-applied group and Et-2-applied group showed 70-80% and 40-50% of hair growth rates, respectively. The rate of hair growth of LP-2-applied group was lower than other applied groups (20-30%). In H/E staining, Numerous hair folicles and hair shafts were observed in Et-1-applied group and other groups showed lower level of hair folicles and hair shafts formation than Et-1-applied group, Conclusion : Et-1 formulation showed highest hair growth activity than other ethosome and liposome formulations entrapped DL-HGF.

Amyloid β peptide (Aβ) still best known as a molecule to cause Alzheimer"s disease (AD). AD is characterized by the accumulation and deposition of Aβ within the brain, leading to neuronal cell loss and perturbation of synaptic function by causing free radical formation, inflammation and apoptosis. We investigated the inflammatory action of Aβ on two types of brain cells, neuronal cells (SH-SY5Y) and neuroglia cells (C6), and its mechanism. We measured the production of NO-iNOS, TNF-α, and ICAM-1 using RT-PCR and Western blot analysis less than the concentration of cytotoxic effects (> 70% survivability). Aβ had no effect on the production of NO and TNF-α, but significantly increases of iNOS and ICAM-1. Based on this, we suggest that the inflammatory effect of Aβ results from the action of ICAM-1 in neuronal cells, rather than the release of inflammatory mediators such as NO and TNF-α in neuroglia cells. In addition, we confirmed whether p53 was related to the action of Aβ by using SH-SY5Y (p53<SUP>-/-</SUP>) dominant cells. Neither the expressionof p53 nor the cytotoxicity of SH-SY5Y (p53<SUP>-/-</SUP>) cells were directly affected by Aβ. However, ICAM-1 was not expressed in SH-SY5Y (p53<SUP>-/-</SUP>) cells. This means that p53-independent pathway exists in the expression of ICAM-1 by Aβ while p53 plays a role as an on-and-off switch.

With the constant development of industrial technology, precision processing systems are required to be diversified, accelerated, and enlarged. Consequently, interest in machine tools for the high-speed processing of large workpieces is increasing. automatic tool changer (ATC) is an automatic tool change device for automatically changing tools during work and is important part of the machining center. The ring-type automatic tool exchange device is advantageous in terms of the number of tools and the size of the magazine owing to its faster operating time and higher space efficiency than before, and ability to use large-sized tools. In this study, seven cases improved according to the thickness of the ring magazine selected using finite element analysis (FEA), and the optimal design was evaluated by static structural and dynamic analysis and multibody dynamics analysis for each case.

본 연구는 에탄올 농도별(0, 30, 50, 70, 100 %)로 추출한 사매추출물의 항산화 효과를 Oxygen Radical Absorbance Capacity(ORAC)로 평가한 결과 30 % 에탄올 추출물에서 가장 우수하였다. 따라서 30 % 에탄올 추출물의 주름완화에 관련한 효능 및 효과를 확인하기 위하여 마우스 유래 섬유아세포에 대한 세포 독성, MMP-8 collagenase 억제활성 검사를 실시하였다. 그 결과 30 % 에탄올 사매추출물을 섬유아세포에 처리한 결과 세포 독성이 없었고, 20 %의 MMP-8 collagenase 억제 활성을 보였다. 사매 추출물 1 %가 함유된 Di-Wrinkle Free Cream (DWFC)으로부터 35 ~ 50세의 21명 여성을 대상으로 하루에 2번 도포하여 8주간 인체 안전성과 주름개선 임상평가를 수행하였다. 주름 평가 방법은 3차원 입체 측정기(PRIMOS)를 이용하여 평가를 하였다. DWFC에 대한 안전성 평가는 피험자 방문 시 마다 이상반응, 부종, 인설, 가려움, 자통, 작열감, 뻣뻣함, 따끔거림이나 다른 이상반응이 발생하는지 육안평가와 문진으로 평가하였다. PRIMOS(GFM, Germany)를 이용하여 눈 주변의 주름을 분석한 결과 시험제품 사용 8주 후 통계적으로 유의하게 주름이 13 % 감소하였으며, 시험기간 동안 특별한 피부 이상반응은 관찰되지 않았다. 이와 같은 결과로 보아 사매추출물은 인체에 부작용이 없는 주름개선에 효과 있는 화장품으로 개발이 가능할 것으로 사료된다. In this study, we investigated anti-oxidative effects of Duchesnea indica extracts by using Oxygen Radical Absorbance Capacity (ORAC). The extracts were prepared with 0 %, 30 %, 50 %, 70 % and 100 % aqueous ethanol respectively. The 30 % EtOH D. indica extract showed higher ORAC activity than the other extracts. Therefore, we performed in vitro studies on cytotoxicity of NIH-3T3 cells and MMP-8 collagenase inhibition using by the 30 % EtOH extract. The 30 % EtOH extract showed no cytotoxicity and significant inhibition on MMP-8 collagenase. And we performed clinical studies for the anti-wrinkle effect of the Di-Wrinkle Free Cream. The cream formula was prepared with 2 % arbutin and 1 % D. indica extract. Twenty one healthy women volunteers, ages of 35 and 50, applied the cream on their faces twice a day for 8 weeks. The skin was evaluated with PRIMOS (phaseshift rapid in vivo measuring of human skin) system and analyzed by the student's paired t-test. The wrinkles on the eye region were reduced by 13 % based on the PRIMOS system after 8 weeks. In the safety study of the Di-Wrinkle Free Cream, no symptoms were observed such as erythema, edema, scaling, itching, stinging, burning, tightness and prickling by visual observation and medical examination of volunteers for 8 weeks. Moreover, there was no noticeable skin disorder during experience period. These results suggested that D. indica extracts could be applied as cosmeceuticals effective for anti-wrinkle.
