      • Studies on the Mode of Action of HC-toxin(I)

        Helminthosporium carbonum 이 생산하는 host-specific HC-toxin의 작용기작이 부분적으로 연구되었다. 말단 epoxide group에 의한 DNA의 alkylation이 fluorescence spectrometry를 이용한 binding assay에 의해 증명되어 옥수수 DNA의 guanine residue가 HC-toxin의 primary target site 중의 하나일 수 있는 가능성이 밝혀졌다. The mode of action of host-specific HC-toxin was studied. The alkylation ability of HC-toxin to a bionucleophile, such as DNA, was proved by fluorescence spectrometry. However, the alkylation was not specific for isolated DNA from susceptible genotype of maize.

        Hygromycin 의 식물생장조절 효과

        A plant growth inhibitor has been isolated from culture filtrate of strain 80-8, a soil isolate. This compound was identified as hygromycin, an antibiotic against gram-positive and gram-negative bacteria. This is the first report that hygromycin inhibits the growth of plant.

        Bacillus sp. SKU31-1가 생산하는 α-Glucosidase 저해제 분리 및 특성 조사

        미생물 기원의 ${\alpha}$-glucosidase 저해제를 탐색하는 과정에서 토양에서 분리한 균주인 Bacillus sp. SKU31-1 배양액에서 강력한 저해제 compound K을 일련의 크로마토그래피 방법에 의해 분리 정제하였고 $^1H$ NMR, $^{13}C$ NMR, $^1H-^1H$ COSY spectra 분석과 문헌조사를 통해 5-amino-1-hydroxymethyl-1,2,3,4-cyclohexanetetrol로 동정되었다. Compound K의 ${\alpha}$-glucosidase 저해 활성은 $IC_{50}$값이 maltose 기질에서는 $1.9{\mu}M$이고, sucrose 기질 사용시 4.9 mM이었다. Lineweaver Burk plot에 의해 $K_i$값이 0.15 mM 로 강력한 경쟁적 저해제 임이 밝혀졌다. In the course of screening for ${\alpha}$-glucosidase inhibitor produced by microorganism, the active compound was isolated from the culture filtrate of Bacillus sp. SKU31-1 using a series of chromatography procedures. The structure of the active compound was elucidated as 5-amino-1-hydroxymethyl-1, 2, 3, 4-cyclohexanetetrol on the basis of spectroscopic evidence obtained and comparison with data from the literature. The active compound showed potent inhibitory activity against ${\alpha}$-glucosidase with an $IC_{50}$ value of $1.9{\mu}M$ for maltose and 4.9 mM for sucrose. A Lineweaver-Burk plot indicated that its inhibition of ${\alpha}$-glucosidase was competitive, with a $K_i$ value of 0.15 mM.

      • Genetic Analysis of Syringomycin and Syringopeptin production by Pseudomonas syringae

        Two types of necrosis-inducing lipodepsipeptide toxins, called syringomycin and syringopeptin, are major virulence factors of Pseudomonas syringae. The phytotoxins syringopeptin and syringomycin are synthesized by nonribosomal peptide synthetases which are encoded by the syringomycin (syr) and syringopeptin (syp) genomic island of Pseudomonas syringae. With the insertional inactivation of rpoS, the rpoS null mutant strain was constructed, and it showed no effect on the production of toxins. Complementation with the wild type ropS gene in trans restored wild-type phenotypes. Expression of the translational fusions syl`-`lacZ and syp`-`lacZ confirmed that rpoS does not affect in syringomycin and syringopeptin biosynthetic operons.

        풋마름병균의 길항세균 Bacillus amyloliquefaciens SKU-78의 대량 배양 조건 확립

        길항균을 생물 농약으로 개발하기 위해서는 저렴한 산업용 배지를 이용한 대량 생산 체계를 확립하는 것이 중요하다. 본 연구에서는 풋마름병 방제 효과가 뛰어난 Bacillus amyloliquefaciens SKU-78 균주의 배양조건을 확립하였다. 저가의 산업용 기질로는 콩가루와 옥수수 전분 배지가 균 생장에 가장 효과적이었고, 최초 pH 5.5, 배양 온도 $30^{\circ}C$, 교반속도 150-250 rpm의 조건으로 30 L fermenter를 이용한 배양에서 20 시간째에 최대 생균수($1.2{\times}10^{11}$ CFU/ml)에 도달하였다. 저가의 산업용 배지로 배양한 배양액을 관주 처리하였을 때 65%의 발병 억제 효과를 나타냄으로써 SKU-78 균주의 산업용 배지를 이용한 대량배양의 기초가 마련되었다. Mass production of biocontrol agent is an essential step for its commercial use. Media composition and culture conditions for production of Bacillus amyloliquefaciens SKU-78, a potential biocontrol agent against bacterial wilts, were optimized by a flask culture. Low cost media combining nitrogen and carbon sources were tested. Maximum cell growth (> $2{\times}10^9$ CFU/ml) was obtained in a medium of 5% soy flour combined with 3% corn starch after 24 h cultivation. The optimum initial pH, temperature and shaking speed was 5.5, $30^{\circ}C$ and 150-250 rpm, respectively. Fermentation of SKU-78 was scaled up in 30 L fermenter and the profiles of cell density, pH, dissolved oxygen and spore formation were recorded. After 8 h lag phase, exponential growth occurred and reached at maximum viable cell number ($1.2{\times}10^{11}$ CFU/ml) after 20 h. The SKU-78 strain grown in a low cost medium exhibited the high suppression of bacterial wilts. The results indicate that SKU-78 strain can be produced in a low cost medium and provide a basis for scaling up to industrial level.

      • 성능 확장형 공유메모리구조의 병렬시스템 설계

        An approach for designing a architectural basis is presented. To achieve balanced high performance computing capability, a computer system should support both the system throughput and execution speedup of a unit task. The goal of this research is to design an architecture that can support both the system throughput and the execution speedup of a given task. In general, any single parallel architecture is not ideal for the broad computational spectrum of algorithm characteristics. The underlining architectureis constructed as combining the throughput oriented multiprocessing architecture for the baselime engine with the function oriented parallel architecture for the speedup engine. The function oriented parallel subsystem(FOPS) can be customized for any specific application domain and is optimally tuned for the specific group of parallelism. In this approach, a FOPS is utilized via parallel function calls from any process unning on a processor of the throughput engine and parallel function call supported is the call of both the parallel hardware and software execution. Single or multiple function calls can be supported for any specific application area depending on needs. Also the multiple function calls within each FOPS and inter-function calls across the FOPS are supported. The research is to design the universial base architecture that can be optimally and economically configured for any application purpose to attain ultimate performance increase.

        제주첨단과학기술단지 개발사업이 인근지역의 토지가격변화에 미친 영향에 관한 연구

        This paper analyzes the effects of the Jeju Science Park, one of the six core projects being promoted by the Jeju Development Center (JDC), on nearby land prices. We created a hedonic model using distance from the Jeju Science Park as an explanatory variable. Other explanatory variables which were considered were distance from the Ara Development District, Jeju National University commercial zone, Namgwang Elementary School and Jeju National University Hospital in addition to zoning category and land type. The rate of increase in land prices is treated as the dependent variable. Two different models were created. In Model 1, only land prices within a 2 km radius of the Jeju Science Park was considered. In Model 2, land prices within a radius of 2 to 3 km was analyzed. In Model 1, distance from the Jeju Science Park exerted an influence on nearby land prices at the 10% significance level, but in Model 2, there was no significant effect on land prices within a radius of 2 to 3 km. We therefore conclude that the Jeju Science Park exerts an effect on land prices within a 2 km radius but fails to exert an effect on land prices outside this area. 본 연구는 국제자유도시개발센터에서 추진하고 있는 6대 핵심프로젝트 중 첨단과학기술단지 조성사업을 선택하여 동 사업이 인근지역의 토지가격에 미친 영향을 분석하였다. 이를 위해 첨단과학기술단지로부터의 거리 변수를 설명변수로하여 토지가격모형을 구성하였다. 그리고 그 외에 아라택지 개발지구까지의 거리, 용도지역, 제주대학교 상권, 지목, 행정구역, 남광초등학교 권역, 제주대학교 병원을 설명변수로 고려하고 종속변수는 토지가격 상승률로 설정하였다. 그리고 본 연구는 두 개의 모형을 구성하였는데, 모형1은 첨단과학기술단지로부터 2㎞ 이내의 공간적 범위를 설정하였고, 모형2는 첨단과학기술단지로부터 2㎞초과 3㎞이내를 공간적 범위로 설정하였다. 분석결과 모형1에서는 첨단과학기술단지로부터의 거리변수가 인근 토지가격에 미치는 영향이 10% 유의수준에서 유의하였지만 모형2, 즉 2㎞초과 3㎞이내에서는 유의하지 않았다.이와 같은 결과를 통해 첨단과학기술단지의 영향이 인근 토지에 2㎞ 반경이내에만 영향을 미치고 그 이상으로는 영향을 미치지 못한다는 것을 발견 할 수 있었다.

        Bacillus subtilis JW-1 균주가 생산하는 bacilysin의 풋마름병 억제 효과 및 특성

        풋마름병 균에 대해 강한 저해활성을 갖는 활성물질 Compound S와 conversion product인 compound S'이 Bacillus subtilis JW-1 배양액에서 일련의 크로마토그래피 방법으로 분리 정제되었고, $^1H$ NMR, $^{13}C$ NMR, $^1H-^1H$ COSY와 HMBC 등의 spectra 분석에 의해 구조가 alanyl-L-${\beta}$-(2,3-epoxycyclohexyl-4-one)alanine와 alanyl-L-${\beta}$-(2,3-dihydroxycyclohexyl-4-one) alanine로 각각 동정되었다. Compound S는 $G^+$, $G^-$ bacteria, yeast와 Candida albicans 등에 대해 광범위한 antimicrobial activity를 나타내며, conversion product의 활성 상실을 통해 intact oxirane ring이 Compound S의 활성에 필수적임이 밝혀졌다. The inhibitory compound (Compound S) against Ralstonia solanacearum and its conversion product (Compound S') were isolated from the culture filtrate of Bacillus subtilis JW-1 using a series of chromatography procedures. The structures were elucidated as alanyl-L-${\beta}$-(2,3-epoxycyclohexyl-4-one)alanine and alanyl-L-${\beta}$-(2,3-dihydroxycyclohexyl-4-one)alanine, respectively on the basis of nuclear magnetic resonance spectral data, including $^1H$, $^{13}C$, $^1H-^1H$ correlation spectroscopy and heteronuclear multiple bond correlation spectroscopy. The compound S exhibited a broad antimicrobial activity against $G^+$, $G^-$ bacteria, Saccharomyces cerevisiae and Candida albicans. The activity loss of the conversion product revealed that the epoxy function was essential for activity of Compound S.
