Forensic Genetic Analysis for the PowerPlex-16 System in the Korean Population

We have analyzed 15 short tandem repeat (STR) markers included the Power Plex-16 (Promega) kit in a sample of 165 unrelated individuals from Korea. Allele frequencies and forensic parameters were calculated to provide an expanded and reliable forensic database. The Exact Test demonstrated that all loci surveyed here were found to be no deviation from Hardy-Weinberg equilibrium. Statistical analyses based on allele frequencies of the 15 STR loci show that twelve Eurasian populations tested can be classified into two distinct clusters; the Koreans appear to have the most genetic affinity with the Japanese of the East Asian cluster but tend to be different from the European cluster. The calculated parameters showed Penta E to be the most valuable marker from the STR loci studied. The combined probability of match calculated from 15 STR loci was 2.19 × 10-17, which is highly informative. Our data, therefore, can be used to extend the results obtained with other STRs, as well as provide valuable information for forensic and population genetic studies in the Korean population.

We analyzed variations at thirteen Combined DNA Index System (CODIS) short tandem repeat (STR) loci (CSF1PO, FGA, THO1, TPOX, vWA, D3S1358, D5S818, D7S820, D8S1179, D13S317, D16S539, D18S51, and D21S11) in a sample from 130 unrelated individuals in the Korean population. Allele and genotype frequencies were determined using commercial PCR amplification kits. The Exact Test demonstrated that all loci were found to be no deviations from Hardy-Weinberg expectations (P>0.05). The distribution pattern of allele frequencies of the 13 STR loci reflected a common genetic affinity in contemporary populations from east Asians, but the Koreans are more closely related with the Chinese than to the Japanese and Vietnamese in east Asians surveyed so far. Based on the results of allele frequencies of the 13 CODIS STR loci, Koreans studied here were the most significantly different from those of African-American and Caucasian. For forensic testing, the power of discrimination (PD) index ranged from 0.760 at TPOX to 0.961 at FGA. The combined probability of match (PM) calculated from all 13 core CODIS STR loci was 1.94×10 exp (-14), which is highly informative. Therefore, the Korean 13 CODIS STR data could be useful for the regional specific and prerequisite references to the forensic community.

<P><I>Lilium</I> bulbs have long been used as Chinese traditional medicines to alleviate the symptoms of various human inflammatory diseases. However, mechanisms of <I>Lilium</I> bulb-mediated anti-inflammatory activity and the bioactive components in <I>Lilium</I> bulbs remain unknown. In the present study, the anti-inflammatory activity of <I>Lilium</I> bulbs and the underlying mechanism of action were investigated in macrophages using <I>Lilium</I> bulb ethanol extracts (Lb-EE). In a dose-dependent manner, Lb-EE inhibited nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW264.7 cells and bone marrow-derived macrophages (BMDMs) without causing significant cytotoxicity. Lb-EE also down-regulated mRNA expression of inflammatory genes in LPS-stimulated RAW264.7 cells, which included inducuble nitric oxide synthase (iNOS), cyclooxygenase-2 (COX2), and tumor necrosis factor-<TEX>$ \alpha $</TEX> (TNF-<TEX>$ \alpha $</TEX>). Furthermore, Lb-EE markedly restored LPS-induced morphological changes in RAW264.7 cells to a normal morphology. HPLC analysis identified quercetin, luteolin, and kaempferol as bioactive components contained in Lb-EE. Mechanistic studies in LPS-stimulated RAW264.7 cells revealed that Lb-EE suppressed MyD88- and TRIF-induced NF-<TEX>$ \kappa $</TEX>B transcriptional activation and the nuclear translocation of NF-<TEX>$ \kappa $</TEX>B transcription factors. Moreover, Lb-EE inhibited IKK<TEX>$ \alpha $</TEX>/<TEX>$ \beta $</TEX>-induced activation of the NF-<TEX>$ \kappa $</TEX>B signaling pathway and IKK inhibition significantly reduced NO production in LPS-stimulated RAW264.7 cells. Taken together, these results suggest that Lb-EE plays an anti-inflammatory role by targeting IKK<TEX>$ \alpha $</TEX>/<TEX>$ \beta $</TEX>-mediated activation of the NF-<TEX>$ \kappa $</TEX>B signaling pathway during macrophage-mediated inflammatory responses.</P>
A Case of Pleural Paragonimiasis Confused with Tuberculous Pleurisy

Here, we report a case of pleural paragonimiasis that was confused with tuberculous pleurisy. A 38-year-old man complained of a mild febrile sensation and pleuritic chest pain. Radiologic findings showed right pleural effusion with pleural thickening and subpleural consolidation. Adenosine deaminase (ADA) activity in the pleural effusion was elevated (85.3 IU/L), whereas other examinations for tuberculosis were negative. At this time, the patient started empirical anti-tuberculous treatment. Despite 2 months of treatment, the pleural effusion persisted, and video-assisted thoracoscopic surgery was performed. Finally, the patient was diagnosed with pleural paragonimiasis based on the pathologic findings of chronic granulomatous inflammation containing Paragonimus eggs. This case suggested that pleural paragonimiasis should be considered when pleural effusion and elevated ADA levels are observed.
Laparoscopic Bile Duct Exploration with Glissonian Approach and Individual Dissection

Purpose: Laparoscopic Glossonian approach is very useful method not only in open hepatectomy but also in laparoscopic hepatectomy. Laparoscopic Glissonian approach followed by individual dissection makes it easy and safe for individualization of portal triad. Moreover, in the case of impacted bile duct stone in the hilar area, it makes easy bile duct exploration without blood loss. Method: Laparoscopic Glissonian approach and extracorporeal control of Glissonian pedicle make it easy and safe dissection of parenchyma and pedicle control. Hepatectomy including bile duct exploration by using combination of Glissonian approach followed by individual dissection could help bile duct exploration easy without bleeding. If IHD stone was located in the main bifurcation, bile duct exploration should be required. Glissonian approach made identification of left pedicle more easy. After isolation of left pedicle, individual dissection of hepatic artery and left portal vein was easy and safe. After dissection of liver parenchyma using Glissonian approach, bile duct exploration with ligation of left hepatic artery and portal vein performed. Result: This combination method has the advantages of Glissonian approach such as, short operation time and easy approach of hilum. Moreover, it makes bloodless and safe bile duct exploration in hilar type IHD stone. Conclusion: Combination of Glissonian approach and individual dissection is useful method in laparoscopic liver resection requiring bile duct exploration. We report the case of a 59-year-old female patient with Left IHD stone located from bifurcation to peripheral area in the left hepatic lobe. Laparoscopic left hepatectomy without bile duct exploration could result in remnant stone in bifurcation sometimes. Left hepatectomy including bile duct exploration by using combination of Glissonian approach and individual dissection could help bile duct exploration easy without bleeding. If IHD stone was located in the main bifurcation, bile duct exploration should be required. Glissonian approach made identification of left pedicle more easy. After isolation of left pedicle, individual dissection of left hepatic artery and left portal vein was easy and safe. After dissection of liver parenchyma using Glissonian approach, bile duct exploration with ligation of left hepatic artery and portal vein performed. The advantage of combination method is easy and safe hepatectomy and bile duct exploration could be achieved.

OBJECTIVES: The rates of asymptomatic infection with Middle East Respiratory Syndrome (MERS) coronavirus vary. A serologic study was conducted to determine the asymptomatic MERS infection rate in healthcare workers and non-healthcare workers by exposure status. METHODS: Study participants were selected from contacts of MERS patients based on a priority system in 4 regions strongly affected by the 2015 MERS outbreak. A sero-epidemiological survey was performed in 1,610 contacts (average duration from exposure to test, 4.8 months), and the collected sera were tested using an enzyme-linked immunespecific assay (ELISA), immunofluorescence assay (IFA), and plaque reduction neutralization antibody test (PRNT). Among the 1,610 contacts, there were 7 ELISA-positive cases, of which 1 exhibited positive IFA and PRNT results. RESULTS: The asymptomatic infection rate was 0.060% (95% confidence interval, 0.002 to 0.346). The asymptomatic MERS case was a patient who had been hospitalized with patient zero on the same floor of the hospital at the same time. The case was quarantined at home for 2 weeks after discharge, and had underlying diseases, including hypertension, angina, and degenerative arthritis. CONCLUSIONS: The asymptomatic infection was acquired via healthcare-associated transmission. Thus, it is necessary to extend serologic studies to include inpatient contacts who have no symptoms.
