        Two New Species of Placolecis (Lichenized Ascomycota) from China

        Two new species of the lichen genus Placolecis are discovered in China, namely P. kunmingensis An. C. Yin & Li S. Wang and P. sublaevis An. C. Yin & Li S. Wang. The new combination P. loekoesiana (S.Y. Kondr., Farkas, J.J. Woo & Hur) An. C. Yin is proposed. Placolecis kunmingensis is characterized by having simple, spherical or ellipsoid, hyaline spores, and pear-shaped pycnidia; while P. sublaevis can be distinguished by its thallus forming larger aggregations with slightly flattened lobes at the thallus margin, and urn-shaped pycnidia. Descriptions, a phylogenetic tree and a key are provided for all the known Placolecis species in China.

        A small hairpin RNA targeting myeloid cell leukemia-1 enhances apoptosis in host macrophages infected with Mycobacterium tuberculosis

        Myeloid cell leukemia-1 (Mcl-1) plays an important role in various cell survival pathways. Some studies indicated that the expression of Mcl-1 was upregulated in host cells during infection with the virulent Mycobacterium tuberculosis strain, H37Rv. The present study was designed to investigate the effect of inhibiting Mcl-1 expression both in vivo and in vitro on apoptosis of host macrophages infected with M. tuberculosis using a small hairpin (sh)RNA. Mcl-1 expression was detected by the real time-polymerase chain reaction, western blotting, and immunohistochemistry. Flow cytometry and transmission electron microscopy were used to measure host macrophage apoptosis. We found elevated Mcl-1 levels in host macrophages infected with M. tuberculosis H37Rv. The expression of Mcl-1 was downregulated efficiently in H37Rv-infected host macrophages using shRNA. Knockdown of Mcl-1 enhanced the extent of apoptosis in H37Rv-infected host macrophages significantly. The increased apoptosis correlated with a decrease in M. tuberculosis colony forming units recovered from H37Rv-infected cells that were treated with Mcl-1-shRNA. Reducing Mcl-1 accumulation by shRNA also reduced accumulation of the anti-apoptotic gene, Bcl-2, and increased expression of the pro-apoptotic gene, Bax, in H37Rv-infected host macrophages. Our results showed that specific knockdown of Mcl-1 expression increased apoptosis of host macrophages significantly and decreased the intracellular survival of a virulent strain of M. tuberculosis. These data indicate that interference with Mcl-1 expression may provide a new avenue for tuberculosis therapy.

      • Taxol Produced from Endophytic Fungi Induces Apoptosis in Human Breast, Cervical and Ovarian Cancer Cells

        Currently, taxol is mainly extracted from the bark of yews; however, this method can not meet its increasing demand on the market because yews grow very slowly and are a rare and endangered species belonging to first-level conservation plants. Recently, increasing efforts have been made to develop alternative means of taxol production; microbe fermentation would be a very promising method to increase the production scale of taxol. To determine the activities of the taxol extracted from endophytic fungus N. sylviforme HDFS4-26 in inhibiting the growth and causing the apoptosis of cancer cells, on comparison with the taxol extracted from the bark of yew, we used cellular morphology, cell counting kit (CCK-8) assay, staining (HO33258/PI and Giemsa), DNA agarose gel electrophoresis and flow cytometry (FCM) analyses to determine the apoptosis status of breast cancer MCF-7 cells, cervical cancer HeLa cells and ovarian cancer HO8910 cells. Our results showed that the fungal taxol inhibited the growth of MCF-7, HeLa and HO8910 cells in a dose-and time-dependent manner. IC50 values of fungal taxol for HeLa, MCF-7 and HO8910 cells were $0.1-1.0{\mu}g/ml$, $0.001-0.01{\mu}g/ml$ and $0.01-0.1{\mu}g/ml$, respectively. The fungal taxol induced these tumor cells to undergo apoptosis with typical apoptotic characteristics, including morphological changes for chromatin condensation, chromatin crescent formation, nucleus fragmentation, apoptotic body formation and G2/M cell cycle arrest. The fungal taxol at the $0.01-1.0{\mu}g/ml$ had significant effects of inducing apoptosis between 24-48 h, which was the same as that of taxol extracted from yews. This study offers important information and a new resource for the production of an important anticancer drug by endofungus fermentation.

        Notes : First Report of Heterodermia squamulosa (Lichenized Ascomycota, Physciaceae) in South Korea

        Heterodermia squamulosa (Degel.) W.L. Culb. was found in the mountain of Gariwang, Gangwon province, in 2008. It is characterized by numerous squamules along the margin, decorticate and white lower surface, rhizines along the margin, black and densely squarrosely branched, usually forming a dense mat under the thallus. Apothecia margins densely squamulose, ascospores 12~15 × 25~30 ㎛. Atranorin and zeorin contained in thallus. This is the first record of this species in South Korea.

        A Comparative Study of Camouflage Printing Color Matching Based on Monitor and Paper Card

        To improve the accuracy of color reproduction for camouflage printing, a new color matching method, namedmonitor-paper-fabric, was proposed by importing color management into textiles, which matched the printing pastesaccording to the color of the paper card printed by the ink-jet printer after color management. Not like the traditional colormatching method, it matched the printing pastes according to the color from computer monitor after color correction. Twocolor matching methods were analyzed by comparing the color difference. It was found that the “monitor-paper-fabric” colormatching method could be considered as a convenient and feasible color matching method. Most color differences betweenthe monitor and the fabric in the camouflage pattern were reduced to lower than 4, except for color blocks 5 and 6. Inaddition, the color differences of six color blocks between the paper and the fabric were all less than 3.5, and were lower thanthose between the monitor and the fabric. The color consistency between the paper and the fabric was better than thatbetween the monitor and the fabric.

        New Species and New Records of Buellia (Lichenized Ascomycetes) from Jeju Province, South Korea

        A new species and 2 new records of lichen genus Buellia were discovered from Chuja-do Island in Jeju Province during a recent floristic survey: B. chujana X. Y. Wang, S. Y. Kondr., L. $L\ddot{o}k\ddot{o}s$ & J.-S. Hur sp. nov., B. halonia (Ach.) Tuck., and B. mamillana (Tuck.) W. A. Weber. The new species is characterized by a brown, areolate thallus, the presence of perlatolic acid, and a saxicolous habitat. Together with previously recorded species, 10 Buellia species were confirmed from Jeju-do Island. Among these species, 3 growing in the exposed rocky area contained xanthone (yellowish lichen thallus, UV + orange), indicating that production of xanthone in this genus might be a defense strategy against the harm of UV light. Although the genus Buellia has been thoroughly studied in Korea before, novel species have been discovered continuously, and large species diversity has been found in this crustose genus, even from a small rocky island. This study indicates that the coastal area harbors a vast number of crustose lichen species, and there is great potential to discover unknown lichens in the coastal rocky area in Korea.

        Notes : Cladonia peziziformis (Lichenized Ascomycota, Cladoniaceae) New to Korea

        Cladonia peziziformis (With.) J.R. Laundon was collected from Baega mountain, Jeonnam Province, Korea in 2008. It is characterized by short and slender podetia with verruculose surface, split along the sides. Apothecia large, pale brown, always growing on the top of the podetia. Primary squamules shell-like, thick, and convex. Fumarprotocetraric acid contained in thallus. This is the first record of this species in Korea.

        Effect of Lonicera edulis polysaccharide on reducing oral dyeing of lonicera edulis juice

        Fluorescence spectroscopy, particle size determination, and potential analysis were exploited to elucidate the effect of Lonicera edulis polysaccharide on polyphenol protein. The results revealed that Lonicera edulis polysaccharides mediated the binding of polyphenols and proteins through competition and formation of ternary complexes and were also able to enhance the stability of the polyphenol-protein complex solution system. A certain electrostatic effect was also present in the process simultaneously. As confirmed by the dyeing test, to improve oral dyeing, the optimum conditions of adding polysaccharide, pectin, and casein were as follows: the dosage of the polysaccharide group was 1.2 mg/mL, coloring time was 100 min, pH value was 4.0. Pectin group added 0.8 mg/mL with coloring time 80 min, pH 5.0. The addition of casein was 1.2 mg/mL; the coloring time was 100 min with pH 5.0. The sample juice substantiated a significant improvement in the dyeing of porcine tongue mucosa. Under the optimal conditions, microscopic observation validates that the mucosal color of the porcine tongue epidermis was closer to that of unstained porcine tongue epidermis, which significantly improved astringency and oral staining.
