        Quantification of SPS mRNA Expression in Banana Fruit Ripened under Different Conditions Using Real-time RT-PCR

        Real-time reverse transcriptase (RT)-PCR using SYBR GreenI was applied to quantify sucrose phosphate synthase (SPS) mRNA expression at different storage days during the natural ripening (with no treatment), inhibited ripening with ethylene absorbent and accelerated ripening by treatment with propylene of banana fruit. The results showed that expression level of SPS mRNA were significantly different between 3 kinds of ripening fruit. Expression of SPS mRNA in fruit treated with propylene was dramatically promoted, the peak of SPS mRNA expression in these fruits appeared 16 days earlier than that in fruit with natural ripening. On the contrary, when fruit applied with ethylene absorbent, SPS mRNA expression was remarkably inhibited, the amount of SPS mRNA expression were significantly lower than that in naturally ripened fruit. These results indicated that the expression level of SPS mRNA of banana fruit can be impacted by regulating ethylene evolution, and that close relationships may exist between SPS mRNA expression and SPS activity, fruit softening, respiration rate, and ethylene production during ripening of banana fruit.

        Gene cloning and expression analysis of trehalose-6-phosphate synthase, glycogen synthase and glycogen phosphorylase reveal the glycometabolism in the diapause process of Aphidius gifuensis

        Aphidius gifuensis is an important biological control agent of aphid. It stores sugar as energy reserves before the onset of diapause, a form of dormancy that occurs when environmental conditions become unfavorable for development. In this study, cDNA of three relevant enzymes, trehalose-6-phosphate synthase (TPS), glycogen synthase (GYS) and glycogen phosphorylase (GP) were cloned, and their expression profiles in the preparation and maintenance of diapause, and post-diapause development were analyzed. The results showed that AgTPS, AgGYS and AgGP encoded proteins consisting of 807, 690 and 844 amino acids respectively. The expression profiles of three genes were significantly affected by diapause-inducing condition. The increase of transcription of AgGYS and AgGP were followed by the up-regulate of AgTPS expression in the preparation of diapause, expression of all three genes were inhibited while diapause is maintained, and expression of AgGYS was upregulated when diapause was terminated. These results suggested that trehalose is accumulated before initiation of diapause and glycogen may play an important role in post-diapause development.

        Effects of Supplementary Composts on Microbial Communities and Rice Productivity in Cold Water Paddy Fields

        Cold water paddy field soils are relatively unproductive, but can be ameliorated by supplementing with inorganic fertilizer from animal waste-based composts. The yield of two rice cultivars was significantly raised by providing either chicken manure or cow dung-based compost. The application of these composts raised the soil pH as well as both the total nitrogen and ammonium nitrogen content, which improved the soil’s fertility and raised its nitrification potential. The composts had a measurable effect on the abundance of nitrogencycling- related soil microbes, as measured by estimating the copy number of various bacterial and archaeal genes using quantitative real-time PCR. The abundance of ammonia oxidizing archaea and bacteria was markedly encouraged by the application of chicken manure-based compost. Supplementation with the composts helped promote the availability of soil nitrogen in the cold water paddy field, thereby improving the soil’s productivity and increasing the yield of the rice crop.

      • miR-200a Inhibits Tumor Proliferation by Targeting AP-2γ in Neuroblastoma Cells

        Background: MicroRNA-200a (miR-200a) has been reported to regulate tumour progression in several tumours but little is known about its role in neuroblastoma. Our aim was to investigate the potential role and mechanism of miR-200a in neuroblastomas. Materials and Methods: Expression levels of miR-200a in tissues were determined using RT-PCR. The effect of miR-200a and shAP-$2{\gamma}$ on cell viability was evaluated using MTS assays, and target protein expression was determined using Western blotting and RT-PCR. Luciferase reporter plasmids were constructed to confirm direct targeting. Results were reported as mean${\pm}$S.E.M and differences were tested for significance using the 2-tailed Students t-test. Results: We determined that miR-200a expression was significantly lower in neuroblastoma tumors than the adjacent non-cancer tissue. Over-expression of miR-200 are reduced cell viability in neuroblastoma cells and inhibited tumor growth in mouse xenografts. We identified AP-$2{\gamma}$ as a novel target for miR-200a in neuroblastoma cells. Thus miR-200a targets the 3'UTR of AP-$2{\gamma}$ and inhibits its mRNA and protein expression. Furthermore, our result showed that shRNA knockdown of AP-$2{\gamma}$ in neuroblastoma cells results in significant inhibit of cell proliferation and tumor growth in vitro, supporting an oncogenic role of AP-$2{\gamma}$ in neuroblastoma. Conclusions: Our study revealed that miR-200a is a candidate tumor suppressor in neuroblastoma, through direct targeting of AP-$2{\gamma}$. These findings re-enforce the proposal of AP-$2{\gamma}$ as a therapeutic target in neuroblastoma.

        Serum DKK-1 level in the development of ankylosing spondylitis and rheumatic arthritis: a meta-analysis

        To explore the association of serum Dickkopf-1 (DKK-1) levels with the development of ankylosing spondylitis (AS) and rheumatic arthritis (RA) in humans, databases including PubMed, EBSCO, Springerlink, Ovid, WANFANG and China National Knowledge Infrastructure (CNKI) were searched to identify relevant studies. On the basis of rigorous inclusion and exclusion criteria, case–control studies of the relationships between serum DKK-1 levels and AS and RA published before December 2014 were enrolled. Statistical analyses were performed using Comprehensive Meta-analysis 2.0 (CMA 2.0). Seven case–control trials with a total of 300 AS patients, 136 RA patients and 232 healthy controls were included in this study. Meta-analysis results revealed that DKK-1 serum levels were significantly higher in AS patients than in normal controls (standard mean differences (s.m.d.) = 0.301, 95% confidence interval (CI) = 0.094–0.507, P = 0.004), whereas no significant difference in DKK-1 serum levels was observed between RA patients and healthy controls (s.m.d. = 0.798, 95% CI = − 2.166–3.763, P = 0.598). Serum DKK-1 level may be closely related to the development of AS but not of RA.

      • Effects of Panax notoginseng, ginsenoside Rb1, and notoginsenoside R1 on proliferation of human breast carcinoma MCF-7 cells

        In this study, we evaluated the antiproliferative effects of Panax notoginseng, ginsenoside Rb1, and notoginsenoside R1 in the human breast carcinoma MCF-7 cell line. Our results indicated that both Panax notoginseng radix extract (NRE) and Panax notoginseng rhizoma extract (NRhE) possess significant antiproliferative activities in MCF-7 cells. Compared to control group (100%), at the concentrations of 0.05, 0.5, and 1.0 mg/ml NRE, cell growth was concentration-dependently reduced to 81.0 ${\pm}$ 6.1 (P < 0.01), 34.2 ${\pm}$ 4.8 (P < 0.001), and 19.3 ${\pm}$ 1.9 (P < 0.001), respectively. Similar results with NRhE at concentrations of 0.5 and 1.0 mg/ml were obtained in these MCF-7 cells. To identify the responsible chemical constituent, we tested the antiproliferation effects of two representative saponins, ginsenoside Rb1 and notoginsenoside R1, on the MCF-7 cells. The data showed that ginsenoside Rb1 was endowed with antiproliferative properties, while notoginsenoside R1 did not have an inhibitory effect in the concentrations tested. Our studies provided evidence that Panax notoginseng extracts and ginsenoside Rb1 may be beneficial, as adjuvants, in the treatment of human breast carcinoma.

        Genetic Linkage of Genes for a Thermo-sensitive Genic Male Sterility in Maize: The genetic analysis of an TGMS Line Qiong-6ms in maize

        Thermo-sensitive genic male sterile (TGMS) lines can provide new options for hybrid seed production using "two-line" system. A set of F2 and BC1 populations derived from the cross between Qiong-6ms and Dan958 were employed to analyze the inheritance of a TGMS line Qiong-6ms and map the TGMS genes in maize. The results demonstrated that the sterility of Qiong-6ms was governed by two duplicative recessive genes, named tms1 and tms2. The gene tms1 was mapped to chromosome 5 linked with the SSR markers umc1355, umc2302 and umc1784 at a distance of 3.0 cM, 1.3 cM and 0.9 cM respectively; while tms2 was localized on chromosome 3, linked with SSR markers bnlg1605 (0.5 cM) and umc2050 (4.2 cM). These markers, which are tightly linked with the tms1 and tms2 genes, will be helpful for marker assisted selection of TGMS lines in maize.

        Evaluation of a cutting simulation using a cupronickel B10 constitutive model considering the deformation temperature

        Cupronickel B10—an important material widely used in the aero industry—has excellent mechanical and physical properties, such as corrosion resistance and ductility. The present study conducted a quasi-static test and split Hopkinson pressure bar (SHPB) test to obtain the quasi-static and dynamic mechanical properties of cupronickel B10, respectively. In these two experiments, the parameters needed for material constitutive models in cutting simulation are derived. However, the impact deformation temperature in the SHPB experiment has long been ignored, resulting in insufficient simulation accuracy. Hence, the impact deformation temperature is referenced to modify material constitutive models. Simulation results of the models are validated in orthogonal cutting experiments. Validation shows that Johnson-Cook class models are superior in simulating the chip form while Drucker-Prager class models are suitable for forecasting the cutting force and temperature. In particular, modified Drucker-Prager models can reduce the error of cutting force F C to 18.01 %.

        Angelica sinensis Supercritical Fluid CO2 Extract Attenuates D-Galactose-Induced Liver and Kidney Impairment in Mice by Suppressing Oxidative Stress and Inflammation

        Angelica sinensis (AS, Danggui in Chinese) is an important herbal component of various traditional formulae for the management of asthenia and its tonic effects. Although AS has been shown to ameliorate cognitive damage and nerve toxicity in D-galactose (D-gal)-elicited senescent mice brain, its effects on liver and kidney injury have not yet been explored. In this work, mice were subjected to hypodermic injection with D-gal (200 mg/kg) and orally gavaged with AS (20, 40, or 80 mg/kg) once a day for 8 successive weeks. Results revealed that AS significantly improved liver and kidney function as assessed by organ index and functional parameters. In addition, AS pretreatment effectively ameliorated the histological deterioration. AS attenuated the MDA level and markedly enhanced the activities and gene expressions of antioxidative enzymes, namely Cu, Zn-SOD, CAT, and GPx. Furthermore, AS markedly inhibited the D-gal-mediated increment of expressions of inflammatory cytokines iNOS, COX-2, IκBα, p-IκBα, and p65 and promoted the IκBα expression level in both hepatic and renal tissues. In sum, AS pretreatment could effectively guard the liver and kidney of mice from D-gal-induced injury, and the underlying mechanism was deemed to be intimately related to attenuating oxidative response and inflammatory stress.

        Research on the Oxidation-Protective Coatings for Carbon/Carbon Composites

        Anti-oxidation coatings are the key technique for carbon/carbon (C/C) composites used as the thermal structural materials. The microstructure and oxidation behavior of several kinds of high-performance ceramic coatings for C/C composites prepared in Northwestern Polytechnical University were introduced in this paper. It showed that the ceramic coatings such as SiC, Si-$MoSi_2$, SiC-$MoSi_2$, $Al_2O_3$-mullite-SiC and SiC/yttrium silicate/glass coatings possessed excellent oxidation resistance at high temperatures, and some of these coatings were characterized with excellent thermal shock resistance. The SiC-$MoSi_2$ coating system has the best oxidation protective property, which can effectively protect C/C composites from oxidation up to 1973 K. In addition, the protection and failure reasons of some coatings at high temperature were also provided.
