        Prevalence and Subtypes of Blastocystis in Alpacas, Vicugna pacos in Shanxi Province, China

        Blastocystis, an enteric protist, has been reported to be an important cause of protozoal gastrointestinal manifestations in humans and animals worldwide. Animals harboring certain Blastocystis subtypes (STs) may serve as a potential source of human infection. However, information about the prevalence and genetic diversity of Blastocystis in alpacas is limited. In the present study, a total of 366 fecal samples from alpacas in Shanxi Province, northern China, were examined for Blastocystis by PCR amplification of the small subunit rRNA gene, followed by sequencing and phylogenetic analysis. The prevalence of Blastocystis in alpacas was 23.8%, and gender difference in the prevalence of Blastocystis was observed. The most predominant Blastocystis ST was ST10, followed by ST14 and ST5. The detection of ST5, a potentially zoonotic genotype, indicates that alpacas harboring ST5 could be a potential source of human infection with Blastocystis. These data provide new insight into the prevalence and genetic diversity of Blastocystis in alpacas.

      • RALY RNA Binding Protein-like Reduced Expression is Associated with Poor Prognosis in Clear Cell Renal Cell Carcinoma

        The molecular mechanisms involved in the progression of clear cell renal cell carcinomas (ccRCCs) are still unclear. The aim of this study was to analyse the relationships between expression of RALYL and clinical characteristics. In 41 paired samples of ccRCCs and adjacent normal tissues, we used real-time qPCR to evaluate the expression of RALYL mRNA. RALYL protein levels were determined in 146 samples of ccRCC and 37 adjacent normal tissues by immunohistochemistry. Statistical analysis was used to explore the relationships between expression of RALYL and the clinical characteristics (gender, age, tumor size, T stage, N stage, M stage, survival times and survival outcome) in ccRCC. In addition, these patients were follow-up period 64 months (range: 4~116months) to investigate the influence on prognosis. We found significantly differences between ccRCC tissues and normal tissues (p<0.001, paired-sample t test) in mRNA levels of RALYL. Immunohistochemistry analyses in 146 ccRCC samples and 37 adjacent normal tissues showed significantly lower RALYL protein levels in ccRCC samples (${\chi}^2$-test, p<0.001), inversely correlating with tumour size (p=0.024), T stage (0.005), N stage (p<0.001) as well as M stage (p=0.019), but not age (p=0.357) and gender (p=0.348). Kaplan-Meier survival analysis demonstrated that people with lower level of RALYL expression had a poorer survival rate than those with a higher level of RALYL expression, significantly different by the log-rank test (p=0.011). Cox regression analysis indicated that RALYL expression (p=0.039), N stage (p=0.008) and distant metastasis (p<0.001) were independent prognosis factors for the overall survival of ccRCC patients. We demonstrated that the expression of RALYL was significantly low in ccRCC and correlated with a poor prognosis in a large number of clinical samples. Our findings showed that RALYL may be a potential therapeutic target as well as a poor prognostic factor.

        Whole Cell-mediated Biocatalytic Synthesis of Helicid Cinnamylate and Its Biological Evaluation as a Novel Tyrosinase Inhibitor

        Tyrosinase inhibitors are clinically effective for treating some dermatological disorders related to melanin hyperpigmentation. Accordingly, the discovery and development of tyrosinase inhibitors have great value in the pharmaceutical and cosmetic industry. Here, a novel tyrosinase inhibitor, 6′-O-cinnamoyl-helicid (helicid cinnamylate) was successfully synthesized by a simple and effective biocatalytic approach with Aspergillus oryzae cells. Investigation of the effects of several key variables on helicid cinnamylate synthesis found that the reaction conversion, reaction rate and regioselectivity reached 99%, 9.40 mM/h and > 99%, respectively, at the optimal conditions with anhydrous acetone as the solvent, whole-cell concentration of 40 mg/mL, and the molar ratio of vinyl cinnamate to helicid of 10 at 45°C. The whole-cells retained 68.87% of its initial activity after reusing for seven batches, indicating a potent application potential in non-aqueous biocatalytic systems. It was worth noting that helicid cinnamylate demonstrated a more potent tyrosinase inhibitory activity with an IC50 value of 3.55 mM than helicid (IC50 = 4.48 mM) and arbutin (IC50 = 5.48 mM), which suggest that helicid cinnamylate could be developed as a more potential tyrosinase inhibitor. In conclusion, this study provides a novel whole-cell catalytic approach for the synthesis of helicid cinnamylate and insight into its application as a tyrosinase inhibitor.

        Multi-point displacement monitoring of bridges using a vision-based approach

        To overcome the drawbacks of the traditional contact-type sensor for structural displacement measurement, the vision-based technology with the aid of the digital image processing algorithm has received increasing concerns from the community of structural health monitoring (SHM). The advanced vision-based system has been widely used to measure the structural displacement of civil engineering structures due to its overwhelming merits of non-contact, long-distance, and high-resolution. However, seldom currently-available vision-based systems are capable of realizing the synchronous structural displacement measurement for multiple points on the investigated structure. In this paper, the method for vision-based multi-point structural displacement measurement is presented. A series of moving loading experiments on a scale arch bridge model are carried out to validate the accuracy and reliability of the vision-based system for multi-point structural displacement measurement. The structural displacements of five points on the bridge deck are measured by the vision-based system and compared with those obtained by the linear variable differential transformer (LVDT). The comparative study demonstrates that the vision-based system is deemed to be an effective and reliable means for multi-point structural displacement measurement.

        Strain-based structural condition assessment of an instrumented arch bridge using FBG monitoring data

        The structural strain plays a significant role in structural condition assessment of in-service bridges in terms of structural bearing capacity, structural reliability level and entire safety redundancy. Therefore, it has been one of the most important parameters concerned by researchers and engineers engaged in structural health monitoring (SHM) practices. In this paper, an SHM system instrumented on the Jiubao Bridge located in Hangzhou, China is firstly introduced. This system involves nine subsystems and has been continuously operated for five years since 2012. As part of the SHM system, a total of 166 fiber Bragg grating (FBG) strain sensors are installed on the bridge to measure the dynamic strain responses of key structural components. Based on the strain monitoring data acquired in recent two years, the strain-based structural condition assessment of the Jiubao Bridge is carried out. The wavelet multi-resolution algorithm is applied to separate the temperature effect from the raw strain data. The obtained strain data under the normal traffic and wind condition and under the typhoon condition are examined for structural safety evaluation. The structural condition rating of the bridge in accordance with the AASHTO specification for condition evaluation and load and resistance factor rating of highway bridges is performed by use of the processed strain data in combination with finite element analysis. The analysis framework presented in this study can be used as a reference for facilitating the assessment, inspection and maintenance activities of in-service bridges instrumented with long-term SHM system.

        Study on the Flavor Contribution of Phospholipids and Triglycerides to Pork

        For investigating the flavor contribution of phospholipids and triglycerides to pork, the longissimus muscle of Rongchang (RC) and PIC (bred by PIC Company of England) pig were selectively removed of intramuscular triglycerides or total intramuscular lipids. After cooking, the flavor of different samples was compared by sensory evaluation, gas chromatographymass spectrometry (GC-MS), and electronic nose. The results showed that removing triglycerides had little effect on the aroma of the cooked meat, but when total intramuscular lipids were removed a remarkable difference was observed. The control samples covaried strongly with some lipid-derived products, and the petroleum ether defatted samples also positively related to these compounds,but to a lesser extent, both of them negatively covaried with chloroform-methanol extracted samples which associated with some Maillard reaction products. Moreover the flavor difference between 2 breeds would decrease when triglycerides were removed, and reduce further when phospholipids and triglycerides were removed simultaneously. These results suggested that intramuscular lipids especially phospholipids contributed significantly to pork flavor, the flavor difference between 2 breeds resulted mainly from different intramuscular lipids content and composition.

      • Prognostic Significance of Beclin-1 Expression in Colorectal Cancer: a Meta-analysis

        Objective: Beclin-1 has recently been observed as an essential marker of autophagy in several cancers. However, the prognostic role of Beclin-1 in colorectal neoplasia remains controversial. Our study aimed to evaluate the potential association between Beclin-1 expression and the outcome of colorectal cancer patients. Materials and Methods: All related studies were systematically searched in Pubmed, Embase, Springer and Chinese National Knowledge Infrastructure databases (CNKI), and then a meta-analysis was performed to determine the association of Beclin-1 expression with clinical outcomes. Finally, a total of 6 articles were included in our analysis. Results: Our data showed that high Beclin-1 expression in patients with CRC was associated with poor prognosis in terms of tumor distant metastasis (OR=2.090, 95%CI=1.061-4.119, p=0.033) and overall survival (RR=1.422, 95%CI=1.032-1.959, p=0.031). However, we did not found any correlation between Beclin-1 over-expression and tumor differentiation (OR=1.711, 95%CI=0.920-3.183, p=0.090). In addition, there was no evidence of publication bias as suggested by Egger's tests for tumor distant metastasis (p=1.000), differentiation (p=1.000) and OS (p=0.308). Conclusions: Our present meta-analysis indicated that elevated Beclin-1 expression iss associated with tumor metastasis and a poor prognosis in patients with CRC. Beclin-1 might serve as an efficient prognostic indicator in CRC, and could be a new molecular target in CRC therapy.

        Intelligent On-demand Routing Protocol for Ad Hoc Network

        Ad hoc networks play an important role in mobile communications, and the performance of nodes has asignificant impact on the choice of communication links. To ensure efficient and secure data forwarding anddelivery, an intelligent routing protocol (IAODV) based on learning method is constructed. Five attributes ofnode energy, rate, credit value, computing power and transmission distance are taken as the basis ofsegmentation. By learning the selected samples and calculating the information gain of each attribute, thedecision tree of routing node is constructed, and the rules of routing node selection are determined. IAODValgorithm realizes the adaptive evaluation and classification of network nodes, so as to determine the optimaltransmission path from the source node to the destination node. The simulation results verify the feasibility,effectiveness and security of IAODV.

        Effects of controlled atmosphere on browning, redox metabolism and kernel quality of fresh in-hull walnut (Juglans regia L.)

        The green hull of fresh walnuts ( Juglans regia L.) is highly susceptible to browning and accompanied with deterioration ofthe walnut quality, and the currently available preservation technologies still do not meet the market threshold supply of freshwalnut. This study investigated the eff ect of controlled atmosphere (CA) storage alone and combination with 1-Methylcyclopropene(1-MCP) on hull browning and kernel quality of fresh in-hull walnut. Fresh in-hull walnuts were treated with air(Control), CA (5% O 2 , 7.5% CO 2 ), 1-MCP (0.5 μL L −1 ) + CA and stored at 0 ± 0.5 °C for 100 d. Browning incidence, hullredox metabolism, kernel color and walnut oil quality were analyzed. The results showed that CA promoted the activities ofsuperoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX), maintained total phenol (TP), γ-aminobutyricacid (GABA), ascorbic acid (ASA), and glutathione (GSH) contents, inhibited ethylene production, reduced reactive oxygenspecies (ROS) accumulation, and delayed the hull browning. Besides, CA attained the highest kernel quality throughinhibited the decrease of kernel color value and moisture content, and reduced acid value (AV) and peroxide value (POV)of walnut oil. While 1-MCP + CA treatment increased the activities of antioxidant enzymes, the combination weakened theother positive eff ects of CA. However, hull browning and kernel deterioration in 1-MCP + CA were less than that observedin air control. Overall, CA was an eff ective method for the preservation of fresh in-hull walnut at 0 ± 0.5 °C.
