The mechanism of apoptosis induced by eugenol in human osteosarcoma cells

Eugenol is commonly used in dentistry for the sedation of toothache, pulpitis, and dental hyperalgesia. This study was performed to investigate the apoptotic effect of eugenol to human osteosarcoma (HOS) cells and the potential use of this compound in osteosarcoma cells. Eugenol showed the apoptotic effect in HOS cells in dose- and time-dependent manner. Fragmentation and condensation of DNA were showed by TUNEL assay, Hemacolor stain and Hoechst stain. In the DNA electrophoresis analysis, cells showed DNA degradation characteristic of apoptosis with a ladder pattern of DNA fragments. Apoptosis-related factors were analyzed by western blotting. Cells treated with eugenol showed caspase-3, PARP, lamin A and DFF-45 cleavage. Eugenol treatment induced caspase-3 cleavage and activation. Cleavages of PARP, DFF-45 and lamin A were accompanied with activation of caspase triggered by eugenol in HOS cells. Though this study needs more investigations, these results suggest that eugenol induce apoptosis via caspase dependent pathway in HOS cells and eugenol may constitute a potential antitumor compound against osteosarcoma cells.

<P>With the recent increase of the rotational speed of hard disk drives, nanoparticle contamination can cause serious problems including thermal asperity. We designed a comb-shaped clamp and spacer to improve the efficacy of the recirculation filter in the operation of the hard disk drive (HDD). We calculated flow characteristics by a commercial computational fluid dynamics (CFD) code for six cases of the comb-shaped clamp and spacer. The calculation results showed that the outward velocity was the highest when the comb-shape angle was 45deg. We measured temporal variations of particle concentration during the operation of the HDD by experiments. Considering the decay rate and the percent clean-up of particle concentration, we found the case of 45deg to be the best design.</P>
경찰공무원의 리더십이 혁신의지에 미치는 영향에 관한 연구

우리나라 경찰조직에서 혁신활동을 시도하는 목적은 경쟁력 강화라는 국가적인 목표에 대한 공공기관의 부응 이라는 측면도 있으나, 무엇보다 우선적으로 요구되는 것은 국민에 대한 치안서비스의 만족을 위한 자구책이라고 볼 수 있다. 이러한 관점에서 경찰은 계속적으로 혁신을 통한 변화를 노력하고 있다. 경찰조직내에서 추진하고 있는 혁신은 일방적인 지시와 권위주의를 탈피하고 경청과 설득의 자세를 견지함으로써 달성해야 한다. 소통은 조직경쟁력과 치안성과를 좌우하는 핵심요소로, 이를 통해 대내적으로는 각종 불신과 마찰을 해소하고 대외적으 로는 정책의 수립과 실천 그리고 평가 모든 과정에 걸쳐 국민 참여를 확대하고 소통채널을 다각화하여 쌍방향 치 안의 내실을 도모할 수 있다. 따라서 이 연구에서는 경찰조직의 리더십이 혁신의지에 어떠한 영향을 미치는지를 검증하고자 한다. 이를 위해 첫째, 경찰공무원을 대상으로 리더십과 혁신의지에 대한 인식수준을 분석하고, 둘째, 리더십이 혁신의지에 미치는 영향을 분석하여 경찰혁신 활동의 주체가 되는 구성원이 혁신의지를 가지고 혁신활 동에 적극참여 할 수 있는 방안을 모색하는데 목적이 있다. A communication is a key part of these, it affects organization competitive power and police performance. Through communications, Internally, they can resolves various conflicts and distrusts. Externally, they also can expand citizen participations all over steps that are policy makings, practices and evaluations, and diversify communication channels. As a result, they can ensure a internal stability of interactive security. Therefore, this study aimed to verify how the leadership of police organization has effects on the innovative willingness. For this, the first is to analyze the recognition level of police officer's leadership and innovative willingness, the second is to analyze effects of leadership on innovative willingness and find ways that can make police officers who is the main agents of police innovation activity participate in the innovative activity with willingness.

<P><B>Abstract</B></P><P>NF-κB activation plays a crucial role in anti-apoptotic responses in response to the apoptotic signaling during tumor necrosis factor (TNF)-α stimulation. TNF-α induces apoptosis sensitive to the hepatitis B virus (HBV) infected cells, despite sustained NF-κB activation. Our results indicate that the HBV infection induces sustained NF-κB activation, in a manner similar to the TNF-α stimulation. However, these effects are not merely combined. Computational simulations show that the level of form of the IKK complex activated by phosphorylation (IKK-p) affects the dynamic pattern of NF-κB activation during TNF-α stimulation in the following ways: (i) the initial level of IKK-p determines the incremental change in IKK-p at the same level of TNF-α stimulation, (ii) the incremental change in IKK-p determines the amplitudes of active NF-κB oscillation, and (iii) the steady state level of IKK-p after the incremental change determines the period of active NF-κB oscillation. Based on experiments, we observed that the initial level of IKK-p was upregulated and the active NF-κB oscillation showed smaller amplitudes for a shorter period in HepG2.2.15 cells (HBV-producing cells) during TNF-α stimulation, as was indicated by the computational simulations. Furthermore, we found that during TNF-α stimulation, NF-κB-regulated anti-apoptotic genes were upregulated in HepG2 cells but were downregulated in HepG2.2.15 cells. Based on the previously mentioned results, we can conclude that the IKK-p-level changes induced by HBV infection modulate the dynamic pattern of active NF-κB and thereby could affect NF-κB-regulated anti-apoptotic gene expressions. Finally, we postulate that the sensitive apoptotic response of HBV-infected cells to TNF-α stimulation is governed by the dynamic patterns of active NF-κB based on IKK-p level changes.</P>
