효모 세포벽 분해효소 생산균의 탐색 및 효소생산 최적조건의 조사

담자효모 세포벽을 분해하는 효소 생산균의 탐색을 위하여 50여 토양시료와 방선균 1000여개의 미생물을 가지고 3차에 걸친 탐색작업을 실시한 결과 1차적으로 autolysed washed yeast 배지 위에서 투명한 생성크기로 493개 균주를 선발하였으며, 이 균주들을 2차적으로 생효모 한천배지 위에서의 투명한 생성 크기로 117개 균주를 선발 하였다. 선발된 균주들을 가지고 용해활성도의 측정방법을 이용하여 최종적으로 St-1702 균주가 선발되었고, St-1702 균주는 동정 실험결과 잠정적으로 Streptomyces eurythermus 또는 그 유연균으로 동정되어졌다. St-1702 균주를 이용하여 효소생산 최적조건을 조사한 결과 최적배지 및 배양조건은 다음과 같았다 : freeze dried yeast 2% glucose 1%, K_2HPO_4 1%, MgSO_4·7H_2O, 1% peptone 0.5% (NH_4)_2CO_3 0.2% 초기 pH 값 7.0의 배지 및 30℃에서 3일간 배양. 최적배지에서 선발균주를 배양한 배양액을 이용하여 담자효모들의 원형질체 형성 실험을 한 결과 시중에서 구입된 효모 세포벽 분해 효소들보다 더 높은 원형질체 형성률을 보여 주었다. Thousand actinomycetes and 50 soil samples were used for the isolation of microorganisms producing yeast cell wall lytic enzymes. Among 493 strains producing large clear zones on autolysed washed yeast (AWY), 117 strains were selected on living yeast cell agar plates. With the method of lytic activity, one strain (St-1702) was selected, which was temporarily identified as Streptomyces eurythermus. The optimal condition for enzyme production of this strain was partially determined as follows: incubation of the strain for 3 days at 30℃ in the medium containing 2% freeze dried yeast cell, 1% glucose, 1% K_2HPO_4, 0.01% MgSO_4·^7H_2O, 0.5% peptone, and 0.2% (NH_4)_2CO_3 with pH 7.0. The protoplast formation of yeast by using the enzyme produced by this strain was compared with commercial enzymes.

국내에서 재배가 장려되고 있는 콩 19품종의 형태학적 특성조사로 콩의 부위별 중량, 길이, 색 등을 측정하였고 이들 형태학적 인자와 함유성분과의 상관성을 분석하였으며 콩단백질이 화학적 특성과 콩에 함유된 영양저해인자들의 함량도 조사하였다. 19품종 콩의 백립중은 12.28∼25.54g이었으며 날콩 무게에 대한 종피 및 배아의 중량 비율은 각각 6.99∼9.29%, 1.66∼3.35%이었다. 이들 콩의 길이, 단폭 및 장폭은 5.95∼7.95㎜, 5.03∼6.95㎜, 6.44∼8.387㎜이었으며 Hunter Lab. Color system에 의한 종피 및 자엽의 황색도는 각각 15.9∼21.7, 17.7∼23.1이었다. 한편 상관성 분석 결과 날콩의 무게는 자엽 무게 및 장폭과, 자엽의 무게는 장폭과 각각 0.97 이상의 높은 점의 상관성을 보였고 조단백질의 함량은 날콩의 무게, 자엽의 무게, 길이, 장폭 및 배꼽의 길이와 상관관계가 있지만 상관계수는 비교적 낮은 것으로 나타났다. 장려 품종 콩 19종의 단백질 특성을 비교키 위해 용해도에 따른 단백질 분획의 분포, 전기영동 특성 및 아미노산 조성을 조사하였다. 이들 콩에 함유된 글리시닌은 콩단백질의 48.19∼58.86%를 차지하였으며 다른 분획에 비해 프를아민의 비율이 콩품종에 따른 차이가 컸다. 콩단백질의 전기영동 양상은 팔달, 장백, 장엽, 단엽, 남천 및 S-138이 21.5∼37.0kd 범위에서 보인 단백질 띠의 이동도 차이를 제외하고서는 큰 차이를 보이지 않았다. 콩에 소량 함유된 아미노산류는 시스테인, 메티오닌, 타이로신 및 트레오닌이었으며 타이로신의 비율은 품종간에 차이가 가장 큰 것으로 나타났다. 국내 재배장려품종 콩 19종에 함유된 몇가지 영양저해인자의 함량과 리폭시게나아제의 활성을 조사하였다. 복부팽만인자인 라피노오스와 스타치오스의 함량은 각각 0.88∼1.58%와 3.34∼5.30% 범위였는데 힐, 백운, 장백 등의 품종이 이 두 종류 당의 함량이 높았다. 콩에 함유된 트립신 저해제는 29.2∼37.0 ㎎ TI/g, 피트산태로 존재하는 인은 건물 100g 당 336∼605㎎ 정도로 품종에 따라 큰 차이를 보였으며, 리폭시게나아제의 활성은 163.6∼403.5 unit로 팔달, 장백, S-133 등의 품종이 높은 값을 보였다. As a work the investigation of physico-chemical characteristics of the recommended 19 Korean soybean varieties, size, proximate compositions, protein characteristics and antinutritional factors of the soybean were analyzed. The weights of whole, seed coat and germ of each soybean were ranged from 0.1228 to 0.255g, from 0.0093 to 0.0162g and from 0.0034 to 0.0056g, respectively. Length and wide and narrow widths of the whole soybeans were 6.44∼8.38, 5.95∼7.95㎜ and 5.03-6.95㎜, respectively. Yellow value(b in Hunter Lab. Color System) of seed coat and lipid contents, of the dried soybeans were 37.42∼44.37 and 17.22∼21.17%, respectively. From the statistical analysis, it was found that the whole weight was highly correlated with cotyledon weight, and wide and narrow widths of soybeans. The higher correlation coefficient was also marked in the relationship between protein and nitrogen free extract contents. The average contents of water soluble sugars in dried soybeans such as sucrose, raffinose and stachyose were 5.21±1.02%, 1.13土0.20% and 4.30±0.51%, respectively. From the fractionation of soybean protein the glycinin content was ranged from 48.19 to 58.86% and the content variation of this fraction was the least in comparison with the case of the other fractions. The electrophoresis profiles of 19 varieties were not significantly differed except 6 varieties involving Paldal in molecular weight range of 21.5∼37.0Kd. The composite ratios of amino acids of 19 soybean varieties were almostly same except tyrosine, cystein, proline and methionine. The range of trypsin inhibitor content was 21.2∼37.0㎎ TI/g and Bangsa, Baekun, Nam cheon, Eunha, Danyeob and Hill were the varieties with high level of the inhibitor. The lipoxigenase activity was 163.6∼403.5 unit/g in the recommended 19 soybean varieties in Korea. Phytate phosphorus was highly contained in Baekcheon, Danyeob and Paldal.

A study of a family with a hereditary deficiency of antithrombin III (AT III), as measured by an immunoassay, is reported. The propositus was a 37-year-old male with a 3-year history of recurrent attacks of deep vein thrombosis of the lower legs. A study of his family revealed a disposition to thromboembolism on the parental side and an autosomal dominant pattern of inheritance, involving five of ten siblings. They have suffered from frequent episodes of venous thrombosis of the lower legs.
두개골 결손시 경막 대체물질 삽입에 의한 연부조직 유착 방지의 실험적 연구

In a series of rabbits, two craniectomies were made in both side of the parietal hone. Each lesion was filled with gelfoam, lyodura, fat and dural film. Five rabbits were left empty as a control group. The animals were killed in one month after the operation. The degree of adhesion between the soft tissue of the scalp and various dural substitute was evaluated at the time of operative dissection and studied histopathologically. The results were as follows. 1) The insertion of dural substitued substances can prevent the adhesion between the scalp and dura mater. 2) The dural film was most effective in prevention of adhesion. 3) The dural film inserted group exhibited inflammatory reaction. 4) The gelfoam, lyodura and fat inserted groups were though to have had a moderate prevention effect of adhesion. With these results, we could conclude that the insertion of material between the dura and scalp can prevent adhesion. However the ideal inserted material which can pevent inflammatory reaction and adhesion has not been determined. At this point we have not found and ideal substitute. To get an ideal material for prevention of adhesion, further intensive study is required.
원외부 후하소뇌 동맥류에 의한 뇌실내 출혈 : 증례 보고 Case Report

A case of ruptured aneurysm of the distal PICA presenting with isolated intraventricular hemorrhage is presented. The diagnosis of distal PICA aneurysm should be considered if isolated intraventricular hematoma is found without obvious parenchymal hemorrhage or subarachnoid blood in the basal cisterns. Complete vertebral arteriography is a requiste for the recognition of this condition. The outcome in patients with these aneurysms should be good if surgical repair is performed before rebleeding occurs.
