        Disruption of rsmA Gene of Pectobacterium carotovorum subsp. carotovorum LY34 and Effect on Pathogenicity

        The rsmA gene was cloned from soft-rot bacterium Pectobacterium carotovorum subsp. carotovorum LY34 (Pcc LY34), and its role in pathogenicity was investigated by marker exchange mutagenesis. From a cosmid library of Pcc LY34 genomic DNA, a positive clone carrying the rsmA gene was selected, and the gene was cloned by polymerase chain reaction (PCR) amplification. The gene is 186 bp in size and encodes a protein of 62 amino acids with a predicted molecular mass of 6,839 Da. The calculated pI of the RsmA is 8.16. The phylogenetic tree showed that the RsmA of Pcc LY34 appeared genetically identical to the CsrA of Pectobacterium atrosepticum SCRI1043 (100% identity) and similar to the CsrA of Yersinia pestis KIM10+(98.3%). The gene was disrupted by the $Km^r$ gene, and the cells became mutated (i.e., $RsmA^-$ mutant). The pathogenicity test revealed that the disease rating of the $RsmA^-$ mutant only differed slightly from that of the wild type on a slice of potato tuber and a Chinese cabbage stalk. These results suggest that RsmA is not an essential factor for the pathogenicity of Pcc LY34 and that the rsmA gene of Pcc LY34 is not completely derepressed in the $RsmA^-$ mutant for virulence-related genes, contrary to the results of Erwinia carotovora subsp. carotovora $RsmA^-$ mutant, which proved hypervirulent for celery petioles. These results showed that the microenvironmental conditions of the host and/or strain of pathogen are important for the coordination of virulence gene expression.

        목재가 선적된 선창에서 발생한 산소결핍에 의한 질식사

        Oxygen deficiency has been frequent in a clouted space. Wood consumes oxygen and discharges carbon dioxide Instead of photosynthesis in closed space without light, so do some microorganisms on the surface. We experienced a case that a healthy Insect-proofer fell down and died of asphyxia on stair-board at 7 m below the hatch of the cargo-hold shipping wood Analysis of gases in cargo-hold revealed O2; 12.3%, CO; 105 ppm, CH4; 2.7%, and H2S; 1.9% at 1 m below the hatch, and then O2; 6.1%, CO; 220 ppm, CH4; 2.9%, and H2S, 2.3 ppm at 2.5 m below the hatch. Autopsy findings were unremarkable. We justiced the cause of death asphyxia due to oxygen deficiency. As seen in this case, the serious oxygen deficiency was accounted for oxygen consumption by wood and microorganisms.

        Misorientation Characteristics at the Growth Front of Abnormally-Growing Goss Grains in Fe–3%Si Steel

        Penetrating morphologies at the growth front of abnormally-growing Goss grains in Fe–3%Si steel was examined in view ofthe possibility that they are the two-dimensional section of the solid-state wetting (SSW) along the triple junction line. Toobtain the statistically meaningful data, 102 penetrated grain and 204 penetrating abnormally-growing Goss grain morphologieswere examined. The misorientation angles of penetrating and penetrated grains were measured by electron backscattereddiffraction. Among the 102 examined penetrated grain boundaries, none has low misorientation angles less than 15°,whereas 17.2% of the 204 penetrating grain boundaries have low misorientation angles, and 23.5% of them have coincidencesite lattice boundaries. Besides, boundary energies of penetrating Goss grains, which were estimated from misorientationangles of the three grains in the penetrating morphology, satisfied the energetic condition for SSW along the triple junctionline. These results imply that the abnormal grain growth of Goss grains in Fe–3%Si steel occurs by the mechanism of subboundaryenhanced SSW.

        AHP를 이용한 CT 및 MRI의 비용편익분석

        The imports of used medical equipment such as CT, MRI have been rapidly increased every year. How to manage those medical equipment has been one of critical issues in the government sector. In this study, we demonstrate how benefit/cost analysis using the Analytic Hierarchy Process (AHP) can be applied to the comparison of used CT/MRI and new CT/MRI. The final results show that the new CT/MRI is more attractive than the used CT/MRI.

      • 유리체절제술중 발생한 의인성망막열공

        To determine the current incidence, location and treatment of intraoperative iatrogenic retinal breaks occuring during pars plana vitrectomy, we reviewed the records of 208 consecutive eyes after pars plana vitrectomy. Thirty seven of 208 eyes (18%) developed 68 intraoperative tears. Of the 68 tears, 14 (21%) were located in the anterior to equator and 54 (79%) were located in the posterior to equator. Thirty six retinal tears (43%) were located in the upper quadrants and in the lower quadrants, we detected 47 retinal tears (57%). Patients with proliferative diabetic retinopathy and proliferative vitreoretinopathy had a higher incidence of iatrogenic retinal breaks than those with other diagnoses. Methods used to treat the retinal tears included cryotherapy, laser photocoagulation, scleral buckling, internal tamponade of gas, air and silicone oil. Thirty three of 37 eyes (89%) were reattached. According to the use of endolaser photocoagulation, we divided the period of study that the first half : 1989. 5. - 1991. 6. and the second half : 1993. 1 - 1993. 12. The results of chorioretinal adhesion in the 1st half were higher than those of the second half. These results show that vitreoretinal surgery for proliferative diabetic retinopathy and proliferative vitreoretinopathy requires careful managements for prevention of iatrogenic retinal tears. Despite improvements in instrumentation and surgical techniques, iatrogenic retinal tears continue to be an important complication of pars plana vitrectomy. However, with careful intraoperative examination, most iatrogenic retinal tears can be identified and treated successfully.

        The applicability of a small hydro-power system for water pollution treatment facilities is investigated at the fish farm in Nam Jeju. For this study, the present situations of the fish farm are examined, and the running problem of screen is found out. As a means of solving this, the small hydro-power system which runs with seawater of head tank is considered. In order to apply this system, the field examination is carried out by sampling methods. The flux and the gross head are measured from selected site at the fish farm, and available generating electric power is estimated by these data. From this result, it is turned out that there is a set up possibility of this system at the fish farm.

      • 날씨 마케팅 적용을 위한 기후 데이터의 군집 분석

        최근 환경오염으로 인한 날씨의 변화, 자원 고갈에 따른 국제 유가의 상승 등 날씨 및 에너지 문제가 기업이나 국가 심지어 개인의 일상생활, 경제활동에 크나큰 영향을 미치고 있다. 이와 같은 이유로 대체에니지 중 태양 에너지 개발에 필요한 일사량 관리와 기후 데이터의 변화 특성 등을 근거로 지역성 규명에 관한 연구가 많이 이루어지고 있다. 그리나, 아직까지는 데이터 마이닝을 통한 지역적 특성에 따른 군집 및 체계적인 분석 데이터 검색 서비스가 효과적으로 제공되지 않고 있다. 이 논문에서는 국내에서 측정된 기후데이터를 저장 및 관리하기 위한 데이터를 모델링하고, κ-means 기법을 이용하여 국내 기후 데이터를 지역적 특성에 따라 군집함으로써 체계적인 데이터 정보를 제공한다. 그리고 이 정보를 날씨 마케팅에 적용될 수 있는 방법을 제시한다. 제안 시스템은 기업의 날씨 마케팅 연구 및 이에 영항을 미치는 요소와 분석 정보를 제공할 수 있는 기본 데이터베이스 구축에 유용하게 활용될 것이다. Recently, the weather is influenced by the environmental pollution and the oil price is rising because of the lack of resources. So, the weather and energy are influencing not only enterprise or nation, but also individual daily life and economic activity very much. Because of these reasons, there are so many researches about management of solar radiation needed to develope solar energy as alternative energy. And many researchers are also interested in identifying the area according to changing characteristics of climate data. However, so far, the researches have not been developed how to apply the cluster analysis, retrieval and analytical results according to the characteristics of the area through data mining. In this paper, we modeled the data for storing and managing the climate data tested in twenty cities in the domestic area. And we provided the information according to the characteristics of the area after clustering the domestic climate data, using κ-means clustering algorithm. And we suggest the way how to apply to the department store and amusement park as an applied weather marketing. The proposed system is useful for constructing the database about the weather marketing and for providing the elements and analysis information, influencing to the weather change.

      • 유도경기상황의 심판의식 및 판단장애에 관한 연구

        The purpose of this study is to produce fair judo judges by examining and analyzing the judicial consciousness during the authentic judo games as well as to attribute to consistent and clear judgment. In accordance with the result of the study I suggest: First, we must try to make judo judges participate continuously in the training program for judo judges and offer them some helpful programs to judge modern judo which is changing right now and to adapt to the new skills or the changed judicial rules so that they may enhance their accurate positions. Second, the judo judges are required to perceive their concrete judicial criteria by repeated practice considering when to give the positive scores and when to give the negative scores after being given the information. Third, more organized training project is needed especially before judo competition, in addition, simulated judicial practice by analyzing some real games on video is helpful to relieve the judo judges' anxiety. Fourth, the judo judge who commited misjudgment or made mistakes on account of his/her circumstances such as his/her academical, regional connections and the teal leaders must be got cold and definite punishment. Lastly, more qualified judo judges, who are distributed fairly in order to be offered more experience are the resources toward he equally high-leveld-judo judges-state. Therefore, we need to study more on the specific elements which influence on the judo judges' judgment and the giving scores.
