      • CORBA 기반 SNMP 게이트웨이

        As telecommunicaton networks becomes more complex and intelligence, it is essential to develop an integrated network management system between heterogeneous telecommunication networks. To fulfil such demands, one solution is CORBA, which provides a distributed platform, in this paper, we present the design and implementation of a gateway between CORBA and SNMP, called as an Internet management protocol. The proposed scheme allows a CORBA managerto access the SNMP MIB without special transformation of SNMP managed objects and only but using SNMP API which the gateway provides. Also since managed objects are represented in CORBA IDL, a new SMI can be updated only by recompiling the interface. Consequently this gateway approach gives a great of advantages in a point that any changes of managed object implementation on managed agents MIBs are not required.

        알코올 의존과 후보 유전자들간의 연관성 연구

        연구목적 : 알코올 대사와 중추신경계내의 알코올 작용부위와 관련한 일부 후보유전자들이 알코올 의존과 관련이 있는지를 확인하고자 하였다. 방 법 : DSM-IV로 진단된 알코올 의존 환자 128명과 정상 대조군 128명을 대상으로 후보 유전자들의 다형성을 중합효소 연쇄반응과 제한효소처리법을 이용하여 동정하였다. 여기서 분리된 대립유전자와 유전자형에 따른 빈도의 차이를 유전자별로 각각 비교하였으며, 알코올 의존에 작용하는 서로 다른 유전자들에 의한 영향력을 배제하기 위하여 logistic regression 분석을 적용하였다. 결 과 : 알코올 의존군에서는 ALDH_2 유전자의 NN(wild type)형이 정상대조군에 비하여 유의하게 많았다(chi-square test, p<0.001). logistic regression analysis를 통하여 다른 유전자들에 의한 영향을 보정한 상태에서 ALDH_2가 NN(wild type)인 경우 다른 아형에 비하여 알코올 의존에 대한 odds ratio는 130.312이며 그 신뢰구간은 (17.22, 986.43)이었다. 한편, ASA는 N-glycosylation 부위에 변이형 대립유전자를 갖고 있는 경우(AG or GG)는 wild type에 비하여 알코올 의존에 대한 위험도는 2.344배이며 그 95% 신뢰구간은 (1.128, 4.871)이었다. 결 론 : 알코올 의존과 ALDH_2 유전자간에는 밀접한 연관성이 확인되었다. 또한 다른 유전자들은 개별 연관분석에서 의미있는 연관성이 관찰되지는 않았지만 logistic regression analysis를 통하여 N-glycosylation 부위에 다형성이 있는 ASA 유전자가 알코올 의존과 관련이 있음을 시사하였다. Objectives : This study was to explore the association between alcohol dependence and five candidate genes related to the metabolism of alcohol and the enzymes of the suspected sites in CNS. Methods : The genotype and allele frequencies of five candidate genes in 128 male hospitalized patients who met DSM-IV criteria for alcohol dependence were compared with 128 age-matched healthy male control subjects using polymerase chain reaction and restriction fragment length polymorphism. A logistic regression analysis was applied in order to exclude the reciprocal interactions among five candidate genes. Results : The NN genotype frequency of the ALDH_2 gene was significantly higher in alcoholic patients than in control subjects(chi-square test, p<0.001). No difference in frequency was found in the other four genes. In a logistic regression analysis, the odds ratio for alcohol dependence in the NN genotype of the ALDH_2 gene and AG or GG genotypes of the N glycosylation site on the ASA gene were 130.312(95% confidence interval, 17.22-986.43) and 2.344(95% confidence interval. 1.128-4.871), respectively. Conclusion : The result reiterates the association of the ALDH_2 gene polyporphism and the alcohol dependence. Logistic regression analysis additionally suggested that the N-glycosylation site on the ASA gene was associated with alcohol dependence.

      • Agrobacterium tumefaciens의 Ti plasmid 재조합에 관한 연구

        Crown gall의 병원체로 알려진 Agrobacterium tumefaciens가 함유하고 있는 Tumor inducing(Ti) plasmid가 식물 hormone과 opine의 합성원인이라는 것이 밝혀지고 식물 세포의 crown gall에는 Ti plasmid가 존재하며 Ti plasmid의 일부가 식물세포의 핵 genome에 삽입되어 형질 변화가 일어남이 보고 되었고 Ti plasmid의 식물체로의 gene도입 vector로서의 연구가 최근 활발히 진행되고 있으며 현재 PLGV vector와 PMON vector 등이 개발되고 있으나 조작이 복잡하고 아직은 초보적 단계인 것으로 알려져 있다. 본 연구에서는 원하는 식물 gene을 식물체에 도입하는 유용한 vector개발의 초보단계로서 Ti plasmid의 T-DNA의 일부인 nos gene을 분리하여 pBR322와의 재조합을 기도한 것이다. pBR 322를 포함하는 E. coli로 부터 Horwitz(1979)의 방법으로 pBR 322를 분리하였고 A. tumefaciens C58에서 Kado(1979)의 방법으로 Ti plasmid를 분리한 후 Ti plasmid 내의 nos gene을 분리하였다. 분리된 pBR 322를 Hind Ⅲ로 절단한 후 CIP로 처리하고 T_4 DNA ligase를 이용하여 Hind Ⅲ로 처리한 nos gene fragment와 재조합하였다. 이와같이 하여 얻은 recombinant DNA로 인한 E. coli HB101의 형질 변화를 Mandel과 Higa의 방법에 따라 조사한 결과 pBR 322의 tetracyclin gene 사이에 nos gene이 삽입된 것으로 확인되었다. 즉 ampicillin 배지에서는 92개의 colony가 형성되었는데 그중 59개는 tetracyclin 배지에서 colony를 형성하지 못하였다. 이러한 tetracyclin 감수성이면서 ampicillin 저항성인 colony를 배양하여 얻은 recombinant plasmid를 전기이동법으로 분리한 결과 예상대로 크기가 7.5kb 정도였고 Hind Ⅲ로 절단하였을 때 2개의 band가 확인되었다. 본 연구에서 얻은 nos gene-pBR 322 hybrid plasmid는 원하는 식물 gene을 이것에 삽입하여 A. tumefaciens 내에서 homologous recombination을 행한 후 식물체에 감염시켜 원하는 gene의 발현여부를 검출하는데 이용될 것으로 기대된다. It is mow realized that the tumor-inducing(Ti) plasmid in Agrobacterium tumefaciens is responsible for the induction of crown gall tumors in dicotyledonous plants and it has been demonstrated that a segment of the Ti plasmid, so called T-DNA, is stably integrated into and expressed in the genome of transformed plant cells. Recently, the use of A. tumefaciens Ti plasmid as a vector to introduce a foreign gene has been intensively studied and several vectors such as PLGV vector and PMON vector has been reported. However, their practical use is still at a primitive stage. It was attempted in the present study, therefore, to conduct the recombination of nos gene of T-DNA from A. tumefaciens Ti plasmid and E. coli pBR 322 as a vector it introduce foreign plant gene into dicotyledon plant cells. pBR 322 was isolated from E. coli containing pBR 322 accdrding to Horowitz(1979) and nos gene of T-DNA in Ti plasmid from A. tumefaciens C58 was obtained according to Kado(1979). The isolated pBR 322 was cut using Hind Ⅲ followed by the addition of CIP·pBR 322 fragments and Hind Ⅲ treated nos gene fragments were than recombined using T_4 DNA ligase and the E. coli cells were then transformed by recombinant DNA according to Mandel and Higa(1970). It was confirmed that nos gene was inserted into tetracyclin gene of pBR 322 by the finding that when the transformed cells were incubated in the ampicillin medium, 92 colonies were formed, in which 59 were tetracyclin sensitive. The above tetracyclin sensitive but ampicillin resistant cells were then cultured and the recombinant pBR-nos gene was isolated and subjected to electrophoresis. It was found that size of the recombinant DNA was approximately 7.5 Kb as expected and two bands were appeared on electrophoretogram when the recombinant DNA was treated with Hind Ⅲ. It might be possible to insert the foreign plant genes to the pBR-nos gene hybrid plasmid obtained in the present study and the resultant DNA could be then homolgously recombined with Ti plasmid of A. tumefaciens and the final recombined Ti plasmid could be used for the introduction of the foreign plant genes into dicotyledon plant cells.

        전착균일성에 미치는 보조음극의 영향

        A numerical simulation based on the finite element method is used to investigate the effect of auxiliary electrode on the uniformity of deposition rate at a patterned electrode. The calculated deposition rates agreed well with the measured ones. Influences of the width of auxiliary electrode, gap between cathode and auxiliary electrode, distance between the inner wall of bath and auxiliary electrode and shape of auxiliary electrode on uniformity of electrodeposit have been studied. The optimum geometric factor of auxiliary electrode was obtained. (Received January 2, 2004)

        백서 뇌에서 전기경련 충격에 의해 발현이 조절되는 새로운 유전자

        연구목적: 이 연구는 백서 뇌에서 전기경련 충격(Electroconvulsive shock, ECS)에 의해 발현이 조절되는 새로운 유전자를 검색하고, 발달단게 및 뇌 조직에 따른 발현양상을 관찰하기 위하여 시행되었다. 방법: 생후 7일, 21일된 어린 웅성 백서에 전기경련 충격을 가한 후 mRNS differential display-PCR(DD-PCR) 기법을 시행하여 발현의 차이를 나타내는 유전자를 검색한 후 서열분석을 실시하였다. 생후 7일 및 21일된 백서의 뇌와 성년 백서의 대뇌피질, 해마를 이용한 Northern blot 분석을 시행하여 검색된 유전자의 ECS 처치 후 시간경과에 따른 유전자 발현양상을 관찰하였다. 결과: ECS 처치군과 위충격군간에 발현의 차이를 나타낸 유전자 4개를 검색할 수 있었으며, 이중 ECS에 의해 발현이 증가되는 1개의 유전자를 선정하여 서열분석을 실시한 결과 EST 서열만이 보고된 새로운 유전자임이 밝혀졌다. 이 유전자는 생후 7일된 백서의 뇌에서는 ECS 처치 후 180분에 발현양의 증가가 최고치에 도달하였으나, 생후 21일된 백서의 뇌와 성년백서의 대뇌피질 및 해마에서는 360분 경과시까지 지속적으로 발현 양이 증가하였다. 결론: 이같은 결과는 ECS에 의해 생체 내에서 유전자 발현의 조절이 이루어짐을 시사하며, 향후 이러한 유전자의 세포 내 기능에 관한 연구를 시행함으로써, ECS에 의한 생체의 반응과 치료기전에 대한 이해를 넓힐 수 있을 것으로 기대된다. 중심단어:전기경련 충격·DD-PCR·유전자·백서 뇌. Objectives: This study was performed to identify genes regulated by electroconvulsive shock (ECS) and to observe the pattern of expression of genes according to different developmental stages and brain regions. Method: ECS(130V, 0.5 sec) was given to male Sprague-Dawley rats with age of postnatal day 7 and 21(P7, P21 respectively). After screening genes regulated by ECS with mRNA diffe-rential display-PCR(DD-PCR), we selected one clone among them and observed the induction of this gene after ECS by time-dependent Northern blot analysis of rat brain of P7, P21 and adult rat cortex and hippocampus. Results: By DD-PCR method, we have identified four clones whose expression was regulated by ECS. Among them, one(CP 10-2) was proved to be a new gene by sequencing and BLAST search. Its expression was increased after ECS in P7, P21, and adult rat brain. The expression of CP 10-2 reached peak level at 180 minutes after ECS in P7 rat brain, but was further increased until 360 minutes after ECS in P21 and adult rat brain. Conclusion: In this study, a new gene was identified in rat brain which showed up-regulated expression in response to ECS. Cloning and characterization of this new gene would be helpful to elucidate the effect of ECS in rat brain. KEY WORDS:Electroconvulsive shock·Differential display-PCR·Gene·Rat brain.

      • 인삼 사포닌이 지용성 비타민(A와 E)의 흡수에 미치는 영향

        본 연구에서는 人蔘 Saonin 이 不水溶性 Vitamin인 Vitamin A와 Vitamin E의 吸收에 미치는 영향을 쥐(albino rat, 100~120g ♂)를 使用하여 觀察하고 다음과 같은 結果를 얻었다. 1) Vit. A 를 人蔘 Saponin 과 함께 Stomach tubing法으로 投與한후 쥐의 血淸을 조사한 結果 試驗群의 Vit.A의 含量은 對照群보다 2倍나 된다. 그러나 3時間後부터는 過量의 Vitamin含量이 減少되기 시작하였다. 試驗群의 肝의 Vitamin A含量도 對照群의 1.5倍였으며 3時間後에는 正常 Vitamin量으로 回復되었다. 2) Vit.E의 경우도 Vitamin E를 投與하고 40分後에 血淸과 肝에서의 Vit. E含量을 調査한 結果 試驗群이 對照群에 比하여 1.4~1.5倍였다. 3) 위와같은 實驗結果는 人蔘 Saponin이 Vit. A와 Vit. E의 吸收를 促進한다는 것을 意味하는 것이다. 4) 한편 試驗群의 腎藏에서의 Vitamin含量이 對照群에 比하여 훨씬 높은 것은 人蔘 Saponin이 不水溶性 Vitamin의 吸收뿐 아니라, 移動 및 排泄에도 크게 영향을 미칠것으로 예상된다. It was attempted in this experiment to observe the effect of ginseng saponin on the absorption of water insoluble Vitamins such as Vitamin A and Vitamin E using albino rat (150-200g,♂) and the following result were obtained. 1) When the Vitamin A and ginseng saponin were fed by stomach tubing, Vit. A concentration of blood serum of test group was found as much as twice that of control, but the Vitamin A content started to decrease 3 hours after. The Vitamin A content of the liver of test animal was also found about 1.5 times that of control and its value returned to normal value three hours later. 2) The Vitamin E contents of the blood and lover of test animals were found as much as 1.4-1.5 times that of control 40 minutes after feeding. 3)The above results suggested tha saponin stimulated the absortion of water insolubl Vitamin A and E 4) On the other hand, the contents of the above Vitamins in the kidney of test animals were significantly higher suggesting that the ginseng saponin also affects the transport of the above water insoluble Vitamins.

        상악 결손부 수복을 위한 측두피판의 이용

        The functional deformity following removal of the maxilla is considerable, especially following maxilla cancer excision. Rehabilitation of oral and maxillofacial region used to be very difficult with prosthesis or traditional flaps. Temporal muscle and fascia flaps have been described for immediate reconstruction following hemi-maxillectomy, but not total maxillectomy. The muscle and superficial fascia of the temporal area differ in their physical characteristics, vascular supply and clinical applications. Both can be employed independently or simultaneously as regional flaps in the reconstruction of a variety of complex craniofacial defects. Four case is presented in which 3 case maxilla cancer, one case non-union after maxilla fracture, and all case showed successful use of this flap. Only one patient developed partial necrosis of the flap ; significant necrosis did not occur in any other patient. This present paper reviews the anatomy, surgical technique and utilisation of temporal muscle and fascia flaps.

        DC Arc로의 유동/열전달 해석 - 용강영역(2)

        Heat- and fluid-flow phenomena in the molten steel region in a DC arc furnace were investigated by simultaneous analysis of electromagnetic, flow and temperature fields. It was found that the flow pattern in molten steel was determined by electromagnetic and buoyancy forces rather than the shear force at the melt surface due to the high velocity plasma arc. While the short arc with high current made the hot region beneath the arc expanded, the long arc with high current drove the melt temperature high. And the low arc current led to the non-homogeneous temperature distribution of the melt because the bottom region of the melt was not stirred sufficiently.

      • 돌와(Gynostemma pentaphyllum)엑기스가 白鼠의 肝臟障碍에 미치는 影響

        This study was attempted to invesigate the effects of extract and crude saponins of Gynostemma pentaphyllum Makino on the activities of S-GPT, S-GOT, Al.P, LDH and the levels of total-bilirubin, total-cholesterol in the serum of nomal rats and experimentally CCl_4-intoxicated rats, and on the effect of body, liver and spleen weight in CCl_4-intoxicated rats. The results were shown as follows; The extract (GE.) and crude saponins were showed a significantly decrease in the activities of S-GPT, S-GOT, Al.P and the level of total bilirubin in the serum elevated by CCl_4, but crude saponins was more decreased than GE. The activities of LDH were significantly decreased of the 4th and the 6th days after administration of GE., and crude saponins was decreased in proportion to the number of days. The level of total cholesterol were significantly decreased in all sample groups. when compared with control group, but remarkably decreased in GE. 500㎎/㎏ and crude saponins 100㎎/㎏. The liver and spleen weight were decreased on the administration of GE. and crude saponins. According to the above results, it is thought that GE. and crude saponins exhibited significant antihepatotoxic activity in the CCl_4-intoxicated rats.
