        This study was conducted to investigated the major characteristics of genetically modified rice of "Milyang 204" originated from Dongjinbyeo compared to a non-transgenic rice varieties Dongjinbyeo and Junambyeo. Basta resistant transgenic rice lines carrying bar gene produced by the Yeongnam Agricultural Research Institute were evaluated for their agronomic characters. The transgenic Japonica rice of "Milyang 204" showed inferior phenotypic traits compared to a non-transgenic rice variety Dongjinbyeo and Junambyeo. On the basis of UPOV (Union Internationale Pour la Protaection des Obtentions Vegetables) and NSMO(National Seed Management Office) the transgenic "Milyang 204" showed difference in some traits out of some agronomic traits, such as leaf color, angle of flag leaf, number of spikelets, culm length, white core and white belly compared to the nontransgenic varieties rice.

      • 정신분열병 및 정신분열형 장애에 대한 올란자핀의 효과와 안전성

        연구목적 : 본 다기관 공동임상연구는 국내 환자를 대상으로 새로운 항정신병약물인 올라자핀의 치료 효과와 안전성을 확인하고자 시행되었다. 방 법 : 1999년 7월부터 2000년 3월까지 국내 10개 병원에 방문한 정신분열병 및 정신분열형 장애 105명을 대상으로 비대조 개방 임상연구를 시행하였다. 중등도 이상의 정신병 증상을 지니거나, 부작용으로 약물교체가 필요한 경우 또는 기분장애 증상의 치료가 필요한 환자를 대상으로 하였다. 2∼7일간의 약물 배설기간 후에 1일 10mg의 올란자핀을 투여하였고, 이후로 3∼7일 간격으로 용량을 조절하여 총 8주간 투여하였다. 치료 효과는 PANSS(Positive and Negative Syndrome Scale), BPRS(Brief Psychiatric Rating Scale), CGI(Clinical Global Impression-Severity), MADRS(Montgomery-Asberg Depression Rating Scale), HAM-A(Hamilton Rating Scale for Anxiety)로 판정하였다. 안전성 평가는 자발적인 이상반응 보고, 활력징후와 혈액 검사 및 SAS(Simpson-Angus Scale), BARS(Barnes Akathisia Rating Scale), AIMS(Abnormal Involuntary Movement Scale)척도를 이용하였다. 결 과 : 105명중에서 85.7%인 90명이 8주간의 치료를 완료하였다. 평균 최빈 용량은 일일 16.1(±4.7)mg이었고, 종료시점에서의 BPRS전체 점수가 기저 상태에 비해 40%이상 감소된 반응군은 69.5%이었다. PANSS의 양성 증후군과 일반정신병리 뿐만 아니라 음성 증후군 소척도에서도 유의한 감소를 보였으며, 이 감소들은 치료 초기부터 나타나서 8주간 지속되었다. 43.8%의 피험자가 중등도 이상의 우울증상을 같이 지니고 있었으며, 올란자핀에 의해 MADRS와 HAM-A가 유의하게 감소하였다. 치료 기간 동안에 활력징후에는 뚜렷한 변화가 없었으나 체중은 8주간 지속적으로 증가하였다. SAS와 AIMS는 감소하였으며, 정좌불능증을 제외한 나머지 추체외로 증상의 발생비율도 낮았다. 임상적인 증상이나 징후를 동반하지 않는 ALT/SGPT의 상승을 보였지만 프로락틴을 포함하는 대부분의 혈액 검사상 뚜렷한 이상 변화는 없었다. 결 론 ; 비록 본 임상연구가 비대조 개방 임상연구로서 많은 제한점을 가지고 있지만, 올란자핀이 기존 약물에 내약성을 보이거나 기분증상을 동반하는 정신병에 치료 효과가 있었다. 그리고 기존 항정신병약물 치료시에 문제가 되었던 추체외로 증상과 고프르로락틴 혈증을 포함하는 대부분의 부작용면에서 안전성을 보여 주었다. Objective : This multicenter clinical trial was carried out to investigate the efficacy and the safety of olanzpine for the treatment of Korean patients. Mothod : 105 patients with schizophrenia and schizophreniform disorder, visited at 10 mental or university hospitals, had received an open and non-comparative treatment with olanzapine for 8 weeks. Patients had psychotic or depressive symptoms with the severity above moderate degree or intolerable side effects to previous antipsychotics. After a wash-out period of 2-7 days, 10mg olanzapine was prescribed initially to all the patients, and then the dosage could be adjusted within the range of 5-20mg/day of olanzapine by 3-7 days. Results : 90(85.7%) of 105 patients completed the 8-weeks trial and the mean modal dose of olanzapine was 16.1(±4.7%)mg/day. At the end of the trial, 73 patients(69.5%) were classified as responder, which was defined as 40% or more improvement in BPRS(Brief Psychiatric Rating Scale) score comparing to baseline. There was a significant reduction in the scores of PANSS(Positive and Negative Syndrome Scale)and subscales including negative symptom scores and CGI. Also weekly analysis showed that the reductions in scores were kept on for the whole period of the trial. 43.8% of all the patients had depressive symptoms at the baseline and total scores of MADRS(Montgomery-Asberg Depression Rating Scale) and HAM-A(Hamilton Rating Scale for Anxiety) were also reduced after the trials. Vital signs revealed no clinically significant changes but continuous weight gain was observed during the treatment with olanzapine. The scores of SAS(Simpson-Angus Scale) and AIMS(Abnormal Involuntary Movement Scale) for assessing the EPS(extrapyramidal symptoms)and tardive dyskinesia respectively were significantly decreased and only a few patients reported EPS as adverse events. Although mild and clinically non-significant of ALT/SGPT was observed, most laboratory parameters including plasma prolactin level showed to significant changes during the trial. Conclusions : Although this trial had many limitations because it was a non-comparative and open study, olanzapine showed high efficacy on the positive, negative and depressive symptoms in schizophrenia and schizophreniform disorder.In addition to that, olanzapine showed a substantially favorable safety profile, such as low incidence of EPS and hyperprolactinemia.

      • Generation and DNA Characterization of High-lysine Mutants by Biochemical Selection from Callus Culture of 'Hwayeongbyeo'

        Lysine is the first essential amino acid for optimal nutrient quality in rice grain. For the narrow genetic diversities of lysine contents in rice, somaclonal variation was the source of mutation in our breeding program. Biochemical selection was conducted using 1 mM S-(2-aminoethyl) cysteine followed by two passages of 5 mM lysine plus threonine in the callus subculture medium. The lysine contents in endosperm of all progenies recovered from the biochemical selection were higher than those of their donor cultivar 'Hwayeongbyeo'. These elevated lysine levels of mutants were successfully transmitted to $M_4$ generation. The lysine contents in endosperm varied 3.85 to $4.80\%$ compare to their donor cultivar 'Hwayeongbyeo' was $3.85\%$. Three of high-lysine germplasms, Lys-l, Lys-2 and Lys-7 were selected by biochemical selection and rapid screening methods. DNA analysis showed that a new insertion of Tos 17 which mapped to rice chromosome 11 on the high-lysine mutant, Lys-2.

        Comparison of Higher-order Aberrations between Eyes with Natural Supervision and Highly Myopic Eyes in Koreans

        Purpose: To describe the characteristics and investigate the differences of higher-order aberrations (HOAs) between the eyes with a natural, uncorrected visual acuity (UCVA) of 20/12 and eyes with highly myopic eyes in Korean adults. Methods: Thirty-one eyes of 20 subjects with UCVA of 20/12 (Group 1) and 54 eyes of 36 myopic patients with greater than -6 diopters (Group 2) were analyzed for type and magnitude of HOAs across a 6.0 mm pupil. HOAs were measured by Wavescan (VISX, Santa Clara, CA, USA) in natural scotopic conditions and were presented as root-mean-square (RMS:μm) in Belle aberration maps. Results: The mean spherical equivalent (SE) of manifest refraction was -0.15±0.25 D (range: +0.37 to -0.50 D) in Group 1 and -7.25±0.78 D (range: -6.00 to -9.25 D) in Group 2. The total root-mean-square (RMS) values of HOAs for Group 1 and Group 2 were 0.28±0.09 μm and 0.27±0.087 μm, respectively (P>0.05). The mean values of coma, trefoil, and spherical aberration were 0.14±0.091 μm, 0.14±0.089 μm, 0.091±0.059 μm in Group 1 and 0.16±0.077 μm, 0.14±0.073 μm, 0.082±0.059 μm in Group 2, respectively. Conclusions: This study helped establish ocular aberration standards for those with natural supervision and those with highly myopic eyes among Koreans. Individuals with natural supervision had significant amounts of HOAs, and there was no significant difference in the amount of HOAs between the two groups. The index of higher-order aberrations may not be a perfect predictor of the amount of refractive error. Korean Journal of Ophthalmology 21(2):79-84, 2007

        Cytological Characterization of Interspecific Hybrids in Rice (Oryza sativa L.)

        The wild species of Oryza are an important source of useful genes for resistance to biotic and abiotic stresses in rice. In this study, wide hybridization was used to widen the genepool of japonica rice cultivars. Interspecific crosses were made between three japonica rice cultivars and four wild species of the genus Oryza. Hybrids were produced normally through crosses between O. sativa (AA genome) and O. rufipogon (AA genome). However, hybrids between O. sativa and other distantly related wild species such as O. officinalis (CC genome), O. minuta (BBCC genome), and O. alta (CCDD genome) were produced following hybridization and embryo rescue. The hybrids were intermediate in their morphological characters, compared with the parents of cultivated and wild species. The hybrids between O. sativa and O. rufipogon were partially sterile but the hybrids between O. sativa and O. officinalis, O. sativa and O. minuta, O. sativa and O. alta were completely sterile. Cytological analysis of the interspecific hybrids, AA x CC, AA x BBCC, AA x CCDD showed irregular meiosis with predominant occurrence of univalents and unequal distribution of chromosomes at anaphase 1 of meiosis. Genomic In-situ Hybridization (GISH) analysis showed the presence of O. minuta chromosomes in the F₁ hybrids between O. sativa and O. minuta cross.

        미배양 태아제대혈액세포에서의 염색체이상 확인을 위한 FISH 의 이용

        목적: 미배양 태아제대혈액세포에서 FISH를 실시함으로써 신속하게 염색체 이상을 확인하고자 한다. 방법: 서울대학교병원 산부인과 및 함춘여성크리닉을 방문한 산모들 중 신속한 핵형분석이 필요한 경우 중 제대혈천자술을 시행한 14례의 환자들을 대상으로 염색체 18, 21, X, Y에 대해 FISH를 실시하였다. 결과 : 총 14례에서 염색체 18번과 21번에 대해 관찰한 결과, 1례의 에드워드증후군과 5례의 다운증후군을 관찰하였고 나머지는 정상으로 나타났다. 염색체 18번과 21번에 대한 정확도는 평균 91.0%와 92.4%로 나타났으며, 핵형분석과 비교한 결과는 모두 일치하였다. 1례의 45,X에서 염색체 X와 Y에 대한 FISH의 정확도는 평균 84.6%와 93.9%로 나타났으며, 핵형 분석 결과와 모두 일치하였다. 24시간 배양후 핵형분석에 실패한 9례는 48시간 이상 배양하여 핵형분석결과를 확인할 수 있었고, 3례의 다운증후군과 1례의 45,X, 3례의 46,XX 그리고 2례의 46,XY를 관찰할 수 있었다. 결론: FISH를 실시함으로써 미배양 태아제대혈액세포에서 48시간 이내에 염색체의 수적이상을 정확하게 확인할 수 있어 신속한 결과가 요구되는 경우와 배양이 용이하지 않아 핵형분석에 실패하는 경우 FISH가 매우 유용한 방법이 되리라 사료된다. Objective: To determine the fetal aneuploidy in fetal blood cells from cordocentesis. Methods: We analyzed their karyotype and performed fluorescence in situ hybridization(FISH) for chromosome 18, 21, X, and Y in 14 cases of fetal blood cells from cordocentesis at Department of Obstetrics & Gynecology, College of Medicine, Seoul National University and Hamchoon Women's Clinic. Results: In all cases we obtained the consistent results in both methods and were able to rapidly detect aneuploidy in uncultured fetal blood cells using FISH before karyotyping with culture for 48 hr. The averages for accuracy of FISH were from 84.6 % to 93.9%. Conclusion: In this study we suggest that the rapid detection in uncultured fetal blood using FISH is possible and that this diagnostic method will be clinically useful when rapid result would be demanded.
