The complex roles of cell surface modification in the biofilm formation of Campylobacter jejuni, a major cause of worldwide foodborne diarrheal disease, are poorly understood. In a screen of mutants from random transposon mutagenesis, an insertional mutation in the eptC gene (cj0256) resulted in a significant decrease in C. jejuni NCTC11168 biofilm formation (<20%) on major food contact surfaces, such as polystyrene and borosilicate glass, when compared with wild-type cells (p < 0.05). In C. jejuni strain 81-176, the protein encoded by eptC modified cell surface structures, such as lipid A, the inner core of lipooligosaccharide, and the flagellar rod protein (FlgG), by attaching phosphoethanolamine. To assess the role of eptC in C. jejuni NCTC11168, adherence and motility tests were performed. In adhesion assays with glass surfaces, the eptC mutant exhibited a 0.77 log CFU/츠<sup>2</sup> decrease in adherence compared with wild-type cells during the initial 2 h of the assay (p < 0.05). These results support the hypothesis that the modification of cell surface structures by eptC affects the initial adherence in biofilm formation of C. jejuni NCTC11168. In motility tests, the eptC mutant demonstrated reduced motility when compared with wild-type cells, but wild-type cells with the transposon inserted in a gene irrelevant to biofilm formation (cj1111c) also exhibited decreased motility to a similar extent as the eptC mutant. This suggests that although eptC affects motility, it does not significantly affect biofilm formation. This study demonstrates that eptC is essential for initial adherence, and plays a significant role in the biofilm formation of C. jejuni NCTC11168.

<P>Bacterial biofilm formation in foodservice facilities is a continuous cross-contamination risk through survival and persistence despite disinfectant treatments. In this study, we evaluated biofilm formation and disinfectant susceptibility of 178 strains obtained from a cafeteria kitchen and 70 foodborne pathogens and analyzed results by multivariate data analyses. A total of 23 areas in a cafeteria kitchen were selected for bacterial isolation and identification. The capacity for biofilm formation was tested using a crystal violet assay, and disinfectant susceptibility was examined using an agar well diffusion assay and resazurin reduction assay. The most frequently isolated genera were Bacillus (33%), Acinetobacter (17%), Kocuria (12%) and Staphylococcus (5%). The genus Bacillus showed the strongest capacity of the biofilm formation. The foodborne bacteria exhibited a wide range of susceptibility to disinfectants, such as sodium hypochlorite, hydrogen peroxide, benzalkonium chloride, lactic acid and citric acid. However, the susceptibilities changed after biofilm formation in a strain-dependent manner, and the relative resistance levels changed among the isolates. E. coli O157:H7에 의한 녹즙 저장 환경에서의 미생물학적 오염도 조사

본 연구는 건강한 식이에 대한 관심이 늘어나면서 그 수요량이 증가하고 있는 녹즙에 대한 미생물학적 품질평가를 목적으로 실시되었다. 또한 날것으로 섭취하는 과채류의 장출혈성 대장균의 오염으로 인한 집단식중독 사고가 많이 보고되고 있는 상황에서 E. coli O157:H7이 녹즙에 오염되었을 경우 보관온도, 보관시간별 미생물 커뮤니티의 변화를 정량 분석법과 DGGE분석을 통해 확인하여 잠재적인 식중독 위험성을 확인해 보고자 하였다. 정량 분석 결과 초기부터 총 호기성 세균수가 식품위생법상 기준치(1.0×10? CFU/mL)를 초과한 결과를 보였으며, E. coli O157:H7에 대한 세균수 측정결과를 보면 5℃와 10℃에서 균수가 감소하다가 증가하는 경향을 보였다. DGGE결과를 보면 보관시간이 길고 보관온도가 높을수록 더 많은 미생물 분포도를 보였고, 저장시간이 후반으로 갈수록 부패균인 젖산균이 다수 출현하는 것을 확인할 수 있었다. 이 외에 Rahnella aquatilis와 Citrobacter, Pseudomonas 등의 기회감염성 세균이 검출되었다. 이에, 위 녹즙시료는 잠재적인 식중독의 위험성을 보이므로 제품의 생산, 유통에 더욱 주의를 기울일 필요가 있다고 생각된다. 또한, 제품 구입 후 냉장저장을 하더라도 가능한 한 빠른 섭취가 권장되는 바이다. E. coli O157:H7에 위 제품이 오염되었을 경우 냉장과 실온조건 모두에서 세균수가 증가하였고, 특히 실온에 방치하였을 경우 그 증가폭이 크므로, 이에 특히 주의를 기울일 필요가 있다고 생각된다. The market for green vegetable juice (GVJ) is growing owing to the increasing demand for healthy food; however data on the safety and quality of GVJ are poorly reported. The objective of this study was to investigate the change in microbial community in GVJ during storage and its contamination by E. coli O157:H7. The microbial community was analyzed via culturable and non-culturable methods at 5, 10, and 25℃ for different storage times. In the non-culturable method, denaturing gradient gel electrophoresis (DGGE) was used. The initial bacterial concentration was 2.92×10? CFU/mL, which exceeded the limit prescribed by the Korean Food Hygiene law. The results of the DGGE analysis indicated that the microbial community during storage was diverse and the spoilage lactic acid bacteria were prevalent at a later stage. Other bacteria such as Rahnella, Citrobacter, Pseudomonas, and Cyanobacteria were identified. Thus, the results strongly emphasize the need to pay attention to GVJ production safety, especially with respect to temperature control, in order to prevent the growth of foodborne pathogens such as E. coli O157:H7 and other spoilage bacteria.

The inhibitory effects of Asparagus cochinchinensis against inflammatory response induced by lipopolysaccharide (LPS), substance P and phthalic anhydride (PA) treatment were recently reported for some cell lines and animal models. To evaluate the hepatotoxicity and nephrotoxicity of A. cochinchinensis toward the livers and kidneys of ICR mice, alterations in related markers including body weight, organ weight, urine composition, liver pathology and kidney pathology were analyzed in male and female ICR mice after oral administration of 150, 300 and 600 mg/kg body weight/day saponin-enriched extract of A. cochinchinensis (SEAC) for 14 days. The saponin, total flavonoid and total phenol levels were found to be 57.2, 88.5 and 102.1 mg/g in SEAC, respectively, and the scavenging activity of SEAC gradually increased in a dose-dependent manner. Moreover, body and organ weight, clinical phenotypes, urine parameters and mice mortality did not differ between the vehicle and SEAC treated group. Furthermore, no significant alterations were measured in alkaline phosphatase (ALP), alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), blood urea nitrogen (BUN) and the serum creatinine (Cr) in the SEAC treated group relative to the vehicle treated group. Moreover, the specific pathological features induced by most toxic compounds were not observed upon liver and kidney histological analysis. Overall, the results of the present study suggest that SEAC does not induce any specific toxicity in the livers and kidneys of male and female ICR mice at doses of 600 mg/kg body weight/day.

Apicidin, a histone-deacetylase inhibitor, has been successfully used to inhibit the growth of cancer cells. In this study, the apoptotic potential and mechanistic insights of apicidin were<br/> investigated in human myeloid leukemia U937 cells. Treatment of U937 cells with apicidin resulted in a decrease of cell viability with apoptotic characteristics, including chromatin condensation and ladderpattern fragmentation of genomic DNA. Apicidin converted the procaspase-3 protease to catalytically active effector protease, resulting in subsequent cleavage of poly (ADP-ribose) polymerase (PARP) and inhibitor of caspase-activated deoxyribonuclease (ICAD). In addition, apicidin induced the activation of caspase-9 protease and the cytosolic release of mitochondrial cytochrome c with mitochondrial membrane potential transition. Moreover, apicidin transiently increased the expression of Fas and Fas ligand proteins. Taken together, the results suggest that apicidin induces apoptosis of U937 cells through activation of intrinsic caspase cascades and Fas/FasL system with mitochondrial dysfunction.

Water temperature, salinity, transparency and phytoplankton in the eastern Korean coastal waters in May, August and October during the period of 2007-2010 were determinded. 12 offshore sites; Gangneung, Donghae, Samcheok, Imwon, Jubyun, Hupo, Chuksan, Ganggu, Wolpo, Pohang, Guryongpo and Gampo. Water temperature in the sea surface varied from 14℃ to 16℃, 18℃ to 25℃, 22℃ to 23℃ in May, August and October, but bottom water temperature had a significant fluctuation at different regions and each year. Unlike water temperature, salinity did not show significantly fluctuations. The temporal variations of the transparency did not show a great fluctuation. Chaetoceros spp., Leptocylindrus danicus and Pseudonitzschia pungens were most abundance phytoplankton species in the eastern coastal waters.
