Boehmeria nivea Extract Reduces TNF-α/IFN-γ-induced TARC and MDC Expressions in Human Keratinocytes

Boehmeria nivea leaf extract (BE) has been reported its anti-inflammatory properties. However, there are few studies on its activity for skin inflammatory disorders and the mechanism. Atopic dermatitis (AD) is one of chronic relapsing skin inflammatory condition. This study investigated the inhibitory effects of BE on AD-like inflammatory markers such as macrophage-derived chemokine (MDC/CCL22) and thymus and activation-regulated chemokine (TARC/CCL17) in human keratinocytes. While stimulation with TNF-α/IFN-γ induces TARC and MDC expressions, co-treatment with BE reduced the levels of the chemokines in HaCaT cells. The chemokine quantitation was determined by ELISA methods. To identify which signaling pathways are involved, we performed western blotting. BE inhibited the phosphorylations of STAT1 significantly as well as NF-κB and MAPK. Our results demonstrate that BE inhibits TNF-α/IFN-γ-induced TARC and MDC through regulating the activities of STAT1, NF-κB, and MAPK signaling pathways.

Pancreatic cancer is among the most difficult-to-treat tumors. More than half of patients with this cancer have very few symptoms at the early stages, allowing the development of distant metastases and resistance to cancer treatment. In this study, we found that Juniperus chinensis extract (JCX) decreased the cell viability and migration activity of PANC-1 and SNU-213 pancreatic cancer cells in a dose-dependent manner. JCX increased caspase- 3 activation and generation of reactive oxygen species (ROS). Nacetylcysteine treatment blocked JCX-induced ROS generation and the negative effects on pancreatic cancer cell viability. In addition, JCX down-regulated the levels of phospho-focal adhesion kinase (p-FAK) and phospho-extracellular signal-regulated kinase (p-ERK). Together, these results indicate that JCX induces apoptosis in human pancreatic cancer cell lines through ROS production, downregulating FAK/ERK signaling and activating caspase-3. We propose that JCX-derived compounds represent candidates for the development of alternative medicines for the treatment of pancreatic cancer.

In 2017, two new tomato mosaic virus (ToMV) isolates were collected from greenhouses in Buyeo, Chungcheongnam- do, South Korea. Full-length cDNAs of the new ToMV isolates were cloned into dual cauliflower mosaic virus 35S and T7 promoter-driven vectors, sequenced and their pathogenicities investigated. The nucleotide sequences of isolates GW1 (MH507165) and GW2 (MH507166) were 99% identical, resulting in only two amino acid differences in nonconserved region II and the helicase domain, Ile668Thr and Val834Ile. The two isolates were most closely related to a ToMV isolate from Taiwan (KJ207374). Isolate GW1 (Ile668, Val834) induced a systemic hypersensitive response in Nicotiana benthamiana compared with the isolate GW2, which a single residue substitution showed was due to Val834.

In this study, the essential oil (EO) and the major non-volatile metabolites in white flower buds of Magnolia denudata and violet flower buds of Magnolia liliiflora were analyzed using GC-MS and HPLC. The dominant EO compounds found in the flower buds were monoterpene hydrocarbons (MO), e.g. sabinene (15.8-14.6%), β-myrcene (17.2-9.9%), and β-pinene (12.9-7.7%). Univariate and multivariate analysis were subsequently performed to reveal the relationships between EO compounds and non-volatile metabolites in white and violet flower buds. The results revealed that most of monoterpenes and phenolic acids in the white flower buds, such as sabinene, β-pinene, and rutin, were found to be significantly higher compared to violet flower buds. Furthermore, hierarchical clustering analysis of correlation coefficients suggested a positive correlation between MO and phenolic compounds. Our findings revealed that the metabolic patterns of both EO and non-volatile compounds could reflect the metabolic characteristics of the different colored flower buds. Further studies will be conducted to investigate the biological activities derived from EO and non-volatile compounds.
