      • Expression of the Na<sup>+</sup>-HCO<sub>3</sub><sup>−</sup>cotransporter and its role in pH<sub>i</sub>regulation in guinea pig salivary glands

        <P>Patterns of salivary HCO3<SUP>−</SUP>secretion vary and depend on species and gland types. However, the identities of the transporters involved in HCO3<SUP>−</SUP>transport and the underlying mechanism of intracellular pH (pHi) regulation in salivary glands still remain unclear. In this study, we examined the expression of the Na<SUP>+</SUP>-HCO3<SUP>−</SUP>cotransporter (NBC) and its role in pHiregulation in guinea pig salivary glands, which can serve as an experimental model to study HCO3<SUP>−</SUP>transport in human salivary glands. RT-PCR, immunohistochemistry, and pHimeasurements from BCECF-AM-loaded cells were performed. The amiloride-sensitive Na<SUP>+</SUP>/H<SUP>+</SUP>exchanger (NHE) played a putative role in pHiregulation in salivary acinar cells and also appeared to be involved in regulation in salivary ducts. In addition to NHE, NBC also played a role in pHiregulation in both acini and ducts. In the parotid gland, NBC1 was functionally expressed in the basolateral membrane (BLM) of acinar cells and the luminal membrane (LM) of ducts. In the submandibular gland, NBC1 was expressed only in the BLM of ducts. NBC1 expressed in these two types of salivary glands takes up HCO3<SUP>−</SUP>and is involved in pHiregulation. Although NBC3 immunoreactivity was also detected in submandibular gland acinar cells and in the ducts of both glands, it is unlikely that NBC3 plays any role in pHiregulation. We conclude that NBC1 is functionally expressed and plays a role in pHiregulation in guinea pig salivary glands but that its localization and role are different depending on the type of salivary glands.</P>

        Phage Ф FSV가 Lactobacillus casei의 증식에 미치는 영향

        Phage Φ FSV가 유산종균의 배양에 미치는 영향을 조사하기 위하여 prophage Φ FSV를 보유한 L. casei YIT 9018 균주나 prophage cured strain인 L. casei HYM 1213 균주에 phage Φ FSV(Lac S21)나 wild type phage Φ J-1(Lac J-1)을 임의의 농도로 각각 감염시켜 이들 균주의 증식을 비교하였다. Lac S21 phage를 L. casei YIT 9018 균주에 감염시켰을 경우, 초기 감염농도가 6.0×10 exp(6) pfu/㎖ 미만인 시료에서는 균주의 성장 및 유산생성능이 정상적이었으며, 오히려 phage수는 감소하였다. 그러나 L. casei HYM 1213균주는 Lac S21 phage에도 민감하게 반응하여, 감염된 균주는 감염시킨 농도에 상관없이 모두 2일균에서 phage가 10^9∼10^10 pfu/㎖로 급격히 증가하였으며, 계속 배양함에도 phage는 감소되지 않고 6일균에서 10^7∼10^9 pfu/㎖로 유지되었고, 이로 인하여 유산균은 정상 발효를 못하였다. 따라서 phage Φ FSV는 모균주인 L. casei YIT 9018 균주에는 어떠한 영향도 미치지 못하고, 오히려 prophage cured strains에는 민감하게 작용하여 비정상 발효를 유도함을 알 수 있었다. 또한, Lac J-1 phage는 prophage Φ FSW를 보유한 L. casei YIT 9018 균주와 prophage cured strain인 L. casei HYM 1213 균주에 모두 민감하게 감염되어 phage의 농도가 급증하고, 이로 인해 균주의 이상발효를 야기시켰다. In order to study effect of the phage Φ FSV on the growth of lactic acid bacteria, Lactobacillus casei YIT 9018 (wild type strain with prophage) or L. casei HYM 1213 (prophage cured strain) was infected with various concentrations of phage Φ FSV (called Lac S21) or wild type virulent phage (called Lac J-1). When L. casei YIT 9018 was infected with Lac S21 under the concentration of 6.0×10 exp(6) pfu/㎖, the growth and lactic acid production of the strain were normal and the number of phages decreased. But L. casei HYM 1213 was susceptible to Lac S21. Regardless of the concentration of the phage infection, the number of phages increased rapidly into 10^9 to 10^10 pfu/㎖ at 2 day cultures and was maintained 10^7 to 10^9 pfu/㎖ phages until 6 day cultures. The lactic acid production of L_n casei HYM 1213 infected with Lac S21 was abnormal. Therefore, phage Φ FSV had an evil effect on growth of L. casei HYM 1213, but not L. casei YIT 9018. On the other hand Lac J-1 caused abnormal fermentation to either L. casei YIT 9018 or L. casei HYM 1213.

        Response Prediction after Neoadjuvant Chemotherapy for Colon Cancer Using CT Tumor Regression Grade: A Preliminary Study

        Purpose To investigate whether CT-based tumor regression grade (ctTRG) can be used to predict the response to neoadjuvant chemotherapy (NAC) in colon cancer. Materials and Methods A total of 53 patients were enrolled. Two radiologists independently assessed the ctTRG using the length, thickness, layer pattern, and luminal and extraluminal appearance of the tumor. Changes in tumor volume were also analyzed using the 3D Slicer software. We evaluated the association between pathologic TRG (pTRG) and ctTRG. Patients with Rödel’s TRG of 2, 3, or 4 were classified as responders. In terms of predicting responder and pathologic complete remission (pCR), receiver operating characteristic was compared between ctTRG and tumor volume change. Results There was a moderate correlation between ctTRG and pTRG (ρ = -0.540, p < 0.001), and the interobserver agreement was substantial (weighted к = 0.672). In the prediction of responder, there was no significant difference between ctTRG and volumetry (Az = 0.749, criterion: ctTRG ≤ 3 for ct- TRG, Az = 0.794, criterion: ≤ -27.1% for volume, p = 0.53). Moreover, there was no significant difference between the two methods in predicting pCR (p = 0.447). Conclusion ctTRG might predict the response to NAC in colon cancer. The diagnostic performance of ctTRG was comparable to that of CT volumetry.

      • Evaluation of the Efficacy of Kochiae fructus Extract in the Alleviation of Carbon Tetrachloride-induced Hepatotoxicity in Rats

        Hepatoprotective effects of the extract of Kochiae fructus (KF), a traditional oriental medicinal plant, were evaluated against carbon tetrachloride($CCl_4$)-induced liver damage in rats. Male Sprague-Dawley rats were divided into control, $CCl_4,\;CCl_4$ plus methanol extract of KF (KFM-$CCl_4$), and $CCl_4$ plus butanol extract of KF (KFB-$CCl_4$) groups. KFM and KFB were orally administered once a day (200 mg/kg body weight) for 14 days. A mixture of 0.2 mL/100 g body weight of $CCl_4$ in olive oil was injected at 30 minutes after the final administration of KFM and KFB. The KFB pretreatment resulted in a significant decrease in the serum transaminase and lactic dehydrogenase levels in the $CCl_4$-treated rats. The $CCl_4$ treatment significantly lowered the activities of glutathione, glutathione reductase (GR), glutathione-S-transferase (GST), superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH-Px). However, pretreatment with KFM and KFB resulted in a significant increase in the glutathione, GR and GST levels. KFB increased the activities of SOD, catalase and GSH-Px, but KFM did not alter them. Pretreatment with KFM and KFB resulted in a significant decrease in the production of aminopyrine N-demethylase in the $CCl_4$-treated rats. KF extract would appear to contribute to alleviate the adveISe effect of $CCl_4$ treatment by enhancing the hepatic antioxidant defense system.

        In this paper, a comparative study on the performance of two-phase and three-phase randonuzed pulse position PWM scheme for mitigation of audible switching acoustic noise in motor drives is done in the randomized pulse position PWM, each of two-phase or three-phase PWM pulses is located randomly in each switching interval. Simulation and experimental efforts were executed to investigate the spread effects of power spectra of inverter output voltage, waveforms of ripple current and audible switching acoustic noise.

      • Synthesis, structures and electrochemical characterization of ferrocene-substituted porphyrin and porphodimethene

        Condensation of ferrocenecarboxaldehyde and dipyrrolylmethane gave 5,15-diferrocenyl-2,8,12,18-tetraethyl-3,7,13,17-tetramethyl-5,15-dihydroporphyrin (H21), and 5,15-diferrocenyl-2,8,12,18-tetraethyl-3,7,13,17-tetramethylporphyrin (H22) in 31 and 49% yield, respectively. Structures of H21·CHCl3 and H22·0.5C6H5Me have been determined by X-ray diffraction. Compound H21 has syn-axial configuration and H22 has syn configuration of two ferrocenyl groups. Compound H21 was only proven to be an intermediate species of H22 in the photooxidation pathway. The large steric bulkiness of the ferrocenyl group permitted the isolation of compounds H21 and H22 directly from the oxidation reaction of 5,15-diferrocenyl-2,8,12,18-tetraethyl-3,7,13,17-tetramethylporphyinogen. Porphodimethene H21 has exceptional stability toward air and light in the absence of acid. Electrochemically, ferrocene subunits of H21 are oxidized with a reversible two-electron transfer process (0.33 V), while those of H22 are oxidized with two reversible one-electron transfer processes (0.06 and 0.27 V). ⓒ 2000 Elsevier Science B.V.         Fluid–Structure Interaction Based on Meshless Local Petrov–Galerkin Method for Worm Soft Robot Analysis

        The purpose of this study was to develop a two-way fl uid–structure interaction (FSI) method using the meshless local Petrov–Galerkin (MLPG) method for both the structure and the fl uid to accurately predict the nonlinear behavior of a worm soft robot. Previous research on soft robots has been mainly performed by fi nite element analysis (FEA). However, the nonlinear behavior of a soft robot causes element distortion and discontinuous stress between the adjacent elements, even when adaptive mesh is employed in the FEA. Therefore, MLPG was employed here to precisely predict the nonlinear behavior of a soft robot without using fi nite elements. In addition, a pneumatic soft robot simulation requires two-way FSI analysis that can transmit and receive data between the fl uid and the structure, and the structure and the fl uid, in sequence. To improve accuracy for the interface, the arbitrary Lagrangian–Eulerian method and the level set method were applied here. It was verifi ed that the maximum errors of the fi nite element method FSI and the developed MLPG FSI method were 4.69%, and 0.77%, respectively, and the latter method required fewer nodes than FEM.

        말굽버섯 추출물이 Streptozotocin으로 유발된 당뇨 흰쥐의 혈당, 지질대사 및 면역세포에 미치는 영향

        말굽버섯(Fomes fomentarius)이 당뇨에 미치는 영향을 연구할 목적으로 실험동물에 STZ를 투여하여 당뇨를 유발시킨 후 말굽버섯 물추출물과 메탄올 추출물을 2주간 투여하여 혈당, 지질함량 및 면역에 미치는 영향을 검토한 결과는 다음과 같다. 혈당은 당뇨로 인해 증가하였으나 말굽버섯 추출물 투여군에서 유의적으로 감소를 보였다. 혈청과 간의 총콜레스테롤과 중성지방 함량은 말굽버섯 추출물 급여로 DM군에 비하여 현저한 감소를 나타내었다. HDL-콜레스테롤 함량과 GSH-Px의 활성은 말굽버섯 추출물 급여 시에는 DM군과 비교하여 유의한 증가가 나타났다. 혈청 보체 C3의 함량은 대조군에 비하여 DM군에서 유의한 감소를 보였고, DM-FM 200군에서 대조군 수준으로 회복이 관찰되었다. B cell과 T cell은 DM군에서 유의한 감소를 보였고, 말굽버섯 추출물 투여로 유의적으로 증가되었다. Helper T cell과 suppressor T cell의 분포는 DM군에서 유의하게 감소되었으나, 말굽버섯 추출물 투여로 유의한 증가를 나타내었다. 이상의 결과를 종합하여 볼 때 말굽버섯의 물 추출물과 메탄올 추출물은 혈당조절 기능, 지질대사 개선 및 면역작용을 활성화시킴으로서 STZ의 투여로 인한 당뇨의 증상을 완화시키는 것으로 사료된다. This study was conducted to investigate the effect of extract of Fomes fomentarius (FF) on blood glucose, lipid profile, antioxidant enzymes and immune cells in streptozotocin (STZ)-induced diabetic rats. Male Sprague- Dawley rats were divided into control, STZ-induced diabetic group (DM), STZ plus FF water extract treated group (DM-FW 200) and STZ plus FF methanol extract treated group (DM-FM 200). FW (200 ㎎/㎏ BW) and FM (200 ㎎/㎏ BW) were orally administered once a day for 14 days. Admdinistering FW and FM to STZ-induced diabetic rats lowered the blood glucose level. The supplementation of FW and FM suppressed the increase in the total cholesterol and triglyceride levels in the serum and liver of the diabetic rats. The high density lipoprotein-cholesterol level and glutathione peroxidase activity were higher in the FF-supplemented group compared to the diabetic group. Administering FW and FM increased the suppress in the serum complement component C3, whole blood B-cell, T-cell, helper T cell and suppressor T cell of the diabetic rats. Therefore, it could be suggested that FW and FM are alleviated the diabetic complication through enhancing the hyperglycemia and preventing diabetic complications.
