      • NF‐AT5 is a critical regulator of inflammatory arthritis

        <P><B>Abstract</B></P><P><B>Objective</B></P><P>To investigate the role of NF‐AT5, an osmoprotective transcription factor, in synovial hyperplasia and angiogenesis in patients with rheumatoid arthritis (RA).</P><P><B>Methods</B></P><P>The expression of NF‐AT5 in synovial tissue and synoviocytes from RA patients was examined by immunohistochemistry and Western blot analysis, respectively. Messenger RNA (mRNA) in RA synoviocytes and human umbilical vein endothelial cells (HUVECs) transfected with dummy small interfering RNA (siRNA) or NF‐AT5 siRNA were profiled using microarray technology. Assays to determine synoviocyte apoptosis and proliferation were performed in the presence of NF‐AT5 siRNA. VEGF<SUB>165</SUB>‐induced angiogenesis was assessed by measuring the proliferation, tube formation, and wound migration of HUVECs. Experimental arthritis was induced in mice by injection of anti–type II collagen antibody.</P><P><B>Results</B></P><P>NF‐AT5 was highly expressed in rheumatoid synovium, and its activity was increased by proinflammatory cytokines, such as interleukin‐1β and tumor necrosis factor α. The mRNA profiling of synoviocytes and HUVECs transfected with NF‐AT5–targeted siRNA revealed 3 major changes in cellular processes associated with the pathogenesis of RA: cell cycle and survival, angiogenesis, and cell migration. Consistent with these results, NF‐AT5 knockdown in RA synoviocytes and HUVECs inhibited their proliferation/survival and impeded angiogenic processes in HUVECs. Mice with NF‐AT5 haploinsufficiency (NF‐AT5<SUP>+/–</SUP>) developed a very limited degree of synovial proliferation, as seen on histologic analysis, and decreased angiogenesis, and they exhibited a nearly complete suppression of experimentally induced arthritis.</P><P><B>Conclusion</B></P><P>NF‐AT5 regulates synovial proliferation and angiogenesis in chronic arthritis.</P>

        제조업 근로자에서 직무 스트레스가 자율 신경계 활성도에 미치는 영향

        목적 : 자율신경계 활성도와 직무 스트레스와의 연관성을 구명하기 위해 본 연구를 수행하였다. 직무 스트레스에 의한 심혈관계 질환 발생의 주요 기전으로 자율 신경계 활성도, 특히 교감신경계의 활성화가 기존의 연구에서 제시되고 이는 바, 이러한 자율신경계 활성도를 생리학적 지표(심박수 변동성; Heart rate variability, HRV) 및 생체시료(요중 카테콜아민)를 이용하여 정량적으로 평가하고 이들의 수준과 직무 스트레스와의 연관성을 구명하는 것을 목적으로 한다. 방법 : 천안시 소재 제조업체의 생산직 남성 근로자 145명을 대상으로 하여 단면연구를 실시하였다. 연구 대상자들은 동일한 근무조건(4조 3교대)로 한정하였다. 직무스트레스는 '직무 만족도 설문조사(Karasek's Job Conten Questionnaire, JCQ)'를 이용하여 평가하였고, 자율신경계 기능은 심박수 변동성과 요중 카테콜아민(에피네프린, 노에피네프린, 도파민)을로 평가하였다. 시료 수집 및 심박수 변동성 측정은 자율신경계의 일주기 리듬을 고려하여 각 연구 대상자들에게 각 교대근무대별로 각각 3회 반복 측정하였다. 직무 만족도 설문조사(49문항)와 함께 인구학적 사항, 직업력, 과거병력, 흡연, 음주력 등에 대한 자기 기입식 설문조사를 시행하였다. 결과 : 직무 만족도 설문조사에 의해 평가된 직무 스트레스 수준을 직무긴장모형(job strain model)에 의한 4개 집단으로 나누었을 때 연구 대상자들은 저긴장 집단(low strain) 24명, 고긴장 집단(high strain) 32명, 수동적 집단 65명, 능동적 집단 20명으로 분포하였다. 직무긴장모형 4개 집단간의 심박수 변동성 및 요중 카테콜아민의 유의한 차이는 없었으며, 심박수 변동성중 특히 심혈관계 질환의 주요 기전으로 제시되는 교감신경계의 활성을 나타내는 저주파 변동성도 4개 집단간에 유의한 차이는 없었다(p-value=0.71). 하지만, 직무기간을 사분위수로 나눈 후 직무 기간이 가장 짧은 군(직무기간<48.5개월, N=28)을 대상으로, 직무긴장모형의 4개 집단간의 저주파 변동성(교감신경 활성도 지시) 차이 비교를 위해 일원분산분석을 시행한 결과 집단-간(between group) 동질성검정이 p-값 0.092의 유의확률을 나타내었다. 결론 : 자율신경계 활성도, 특히 심박수 변동성을 이용한 정량적 평가를 통해 직무 스트레스와의 연관성을 평가한 연구라는 점에서 의의를 지닌다. 제한적이긴 하지만, 근무기간이 짧은 군에서 저긴장 집단과 고긴장 집단간의 저주파 변동성의 차이를 보여 심박수 변동성이 잠재적인 생리적 지표가 될 수 있음을 시사한다. Objective: A cross-sectional study was conducted to evaluate the relationships between autonomic nerve activity (i.e., heart rate variability and urinary catecholamines) and job stress. Methods: The study was conducted on 134 workers from a company producing consumer goods (i.e., diaper, paper towel) located in Cheonan, Korea. Job stress was assessed by Karasek's Job Content Questionnaire (JCQ), Autonomic function was assessed by heart rate variability (HRV) and urinary catecholamines (eponephrine, norepinephrine, and dopamine). Urine samplings, and measurings of HRV, were repeated three times for each shift. Information on demographic characteristics, previous job histories, past medical history, smoking and drinking were also collected. Results: No statistically significant differences were observed in ant HRV and urinary catecholamines among the four groups, which were categorized by the Job Strain Model. When data were stratified by work duration, low field HRV was borderline sigificantly higher in the high strain group in individuals with a sgorter work duration (<48.5 months, N=28, p-value of 0.92 by analysis of variance). Conclusions: Neither HRV nor urinary catecholamines are significantly associated with job stress, as assessed by the Karasek's JCQ. However, HRV seems to be a potential physiological incicator of jov stress only in the workers with a shorter work duration.

      • All-solid-state thin film battery based on well-aligned slanted LiCoO<sub>2</sub> nanowires fabricated by glancing angle deposition

        <P><B>Abstract</B></P> <P>We fabricated all-solid-state thin film batteries based on well-aligned slanted LiCoO<SUB>2</SUB> nanowires by glancing angle deposition, as a facile template-free method in order to increase the electrochemically active site, <I>i.e.</I>, the contact area between the solid electrolyte and the electrode. A highly porous thin film composed of well-separated slanted LiCoO<SUB>2</SUB> nanowires not only facilitates the penetration of solid electrolyte phase into the cathode, but also alleviates the thermally and mechanically induced stresses during post-annealing and electrochemical cycling. The all-solid-state thin film battery based on the well-aligned slanted LiCoO<SUB>2</SUB> nanowires, whose contact area between electrolyte and electrode was three times as high as that of a dense thin film, could provide additional migration pathways for lithium ion diffusion due to the enlarged reaction sites. This resulted in enhanced electrochemical kinetics, thereby leading to better rate capability and long-term cyclic stability as compared to the dense LiCoO<SUB>2</SUB> thin film.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Well-aligned slanted LiCoO<SUB>2</SUB> nanowires are fabricated by glancing angle deposition. </LI> <LI> One-dimensional nanostructured LiCoO<SUB>2</SUB> cathode enlarges the contact area. </LI> <LI> All-solid-state thin film battery exhibits enhances rate capability and cycling stability. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>We successfully fabricated well-aligned slanted LiCoO<SUB>2</SUB> nanowires as a one-dimensional nanostructured cathode by glancing angle deposition to enhance the electrochemical performance of all-solid-state thin film batteries.</P> <P>[DISPLAY OMISSION]</P>

      • Short Sleep Duration and Its Correlates among Cancer Survivors in Korea: the Korea National Health and Nutrition Examination Surveys

        Background: Though a large proportion of cancer survivors are assumed to be commonly affected by sleep disturbance, few studies have focused on short sleep problems and its correlates among Korean cancer survivors. The purpose of this study was to evaluate the prevalence of short sleep in adult cancer survivors from a nationwide population-based sample and to identify risk factors for short sleep duration. Materials and Methods: Based on the fourth and fifth Korea National Health and Nutrition Examination Surveys (2007-2012), 1,045 cancer survivors and 33,929 non-cancer controls were analyzed. The prevalence of short sleep was compared between these two groups. Associations between short sleep and its correlates were evaluated using multiple logistic regression among cancer survivors: odds ratios (ORs) and 95% confidence intervals (95%CIs) were estimated after adjusting for sociodemographic factors, lifestyle factors, psychological conditions, and cancer-related factors. Results: About 8.1% of cancer survivors slept for less than 5 hours per day (6.2% men and 9.3% women), whereas this was the case for only 3.7% of non-cancer controls. Cancer survivors who had the lowest household income level showed a significantly higher likelihood for short sleep (adjusted OR 2.82, 95%CI 1.06-7.54). Self-reported poor health and depressive symptoms were found to be associated with significantly increased likelihood for short sleep in cancer survivors (adjusted OR 3.60, 95%CI 1.40-9.26 and adjusted OR 2.00, 95%CI 1.17-3.42). Gastric cancer survivors had a 3.97-fold increased risk for short sleep (95%CI 1.60-9.90). Conclusions: The prevalence of short sleep occurs at a high rate among the Korean cancer survivors, which may indicate a poorer quality of life and a higher risk of future complications in survivorship. Targeted interventions that can assist cancer survivors to cope with sleep disturbances as well as ensuring psychological stability are warranted to reduce the latent disease burden.

      • Proteomic Analysis of Tumor Necrosis Factor-Alpha (TNF-α)-Induced L6 Myotube Secretome Reveals Novel TNF-α-Dependent Myokines in Diabetic Skeletal Muscle

        <P>There is a strong possibility that skeletal muscle can respond to irregular metabolic states by secreting specific cytokines. Obesity-related chronic inflammation, mediated by pro-inflammatory cytokines, is believed to be one of the causes of insulin resistance that results in type 2 diabetes. Here, we attempted to identify and characterize the members of the skeletal muscle secretome in response to tumor necrosis factor-alpha (TNF-α)-induced insulin resistance. To conduct this study, we comparatively analyzed the media levels of proteins released from L6 skeletal muscle cells. We found 28 TNF-α modulated secretory proteins by using separate filtering methods: Gene Ontology, SignalP, and SecretomeP, as well as the normalized Spectral Index for label-free quantification. Ten of these secretory proteins were increased and 18 secretory proteins were decreased by TNF-α treatment. Using microarray analysis of Zuker diabetic rat skeletal muscle combined with bioinformatics and Q-PCR, we found a correlation between TNF-α-mediated insulin resistance and type 2 diabetes. This novel approach combining analysis of the conditioned secretome and transcriptome has identified several previously unknown, TNF-α-dependent secretory proteins, which establish a foothold for research on the different causes of insulin resistance and their relationships with each other.</P><P>In the current study, we used a reliable proteomic approach to comparatively analyze skeletal muscle secretomes in the presence and absence of tumor necrosis factor-alpha (TNF-α) induced insulin resistance. Our results introduce a novel population of TNF-α dependent secretory proteins derived from skeletal muscle.         Improvement of Glucose Uptake Rate and Production of Target Chemicals by Overexpressing Hexose Transporters and Transcriptional Activator Gcr1 in <i>Saccharomyces cerevisiae</i>

        <P>Metabolic engineering to increase the glucose uptake rate might be beneficial to improve microbial production of various fuels and chemicals. In this study, we enhanced the glucose uptake rate in <I>Saccharomyces cerevisiae</I> by overexpressing hexose transporters (HXTs). Among the 5 tested HXTs (Hxt1, Hxt2, Hxt3, Hxt4, and Hxt7), overexpression of high-affinity transporter Hxt7 was the most effective in increasing the glucose uptake rate, followed by moderate-affinity transporters Hxt2 and Hxt4. Deletion of <I>STD1</I> and <I>MTH1</I>, encoding corepressors of <I>HXT</I> genes, exerted differential effects on the glucose uptake rate, depending on the culture conditions. In addition, improved cell growth and glucose uptake rates could be achieved by overexpression of <I>GCR1</I>, which led to increased transcription levels of <I>HXT1</I> and ribosomal protein genes. All genetic modifications enhancing the glucose uptake rate also increased the ethanol production rate in wild-type <I>S. cerevisiae</I>. Furthermore, the growth-promoting effect of <I>GCR1</I> overexpression was successfully applied to lactic acid production in an engineered lactic acid-producing strain, resulting in a significant improvement of productivity and titers of lactic acid production under acidic fermentation conditions.</P>

      • Understanding the Critical Role of the Ag Nanophase in Boosting the Initial Reversibility of Transition Metal Oxide Anodes for Lithium-Ion Batteries

        <P>The initial reversible capacity, a critical impediment in transition metal oxide-based anodes, is augmented in conversion-reaction-involved CoO anodes for lithium-ion batteries, by incorporating a chemically synthesized Ag nanophase. With an increase in the added amount of Ag nanophase from 5 to 15 wt %, the initial capacity loss decreases linearly up to 31.7%. The Ag nanophase maintains its pristine metallic nature without undergoing phase transformations, even during repeated vigorous electrochemical reactions of the active CoO phase. Complementary ex situ chemical/physical analyses suggest that the Ag nanophase promotes the catalytic generation of reversible gel-like/polymeric films wherein lithium ions are stored capacitively in the low-voltage region below 0.7 V during discharging. These scientific findings would provide a heretofore unrecognized pathway to resolving a major issue associated with the critical irreversibility in conversion-type transition metal oxide anodes.</P>

        강원 영서지역 산사태 및 강우특성 분석

        본 논문은 2006년 태풍 에위니아와 빌리스의 영향으로 강원 영서지역에서 발생한 집중호우에 의한 산사태 발생 원인과 특성에 대하여 분석한 결과이다. 강원 영서지역은 편마암, 화강암이 풍화된 화강풍화토가 주를 이루는 지역으로 집중호우 시 산사태 및 토석류의 피해에 취약한 것으로 나타났으며 강우특성 분석결과, 산사태 발생은 3일 이내의 누적강우량과 밀접한 관계가 있었고 지역별 발생빈도의 차이는 최대시우량의 영향으로 나타났다. 영서지역에서 발생한 산사태 860개소에 대한 자료의 분석 결과, 파괴형태는 전이형 슬라이드로 시작되어 유동성 슬라이드로 변화되는 양상을 보였으며, 사면 경사는 20~30°, 사면 길이는 11~20m 범위, 사면 폭은 6~10m 범위에서 발생빈도가 높은 것으로 나타났고, 영서지역의 산사태는 사면 폭이 좁은 소규모 형태로 편마암, 화강암 지대의 산사태 전형으로 분석되었다. This paper is the results of analysis for the causes and characteristics of landslide according to heavy rain occurred in west area of Gangwon province which is affected by typhoon such as Ewiniar and Bilis in 2006. West side of Gangwon province is topographically weak for the landslide and debris flow since it is covered by soil of weathered rock such as Gneiss and Granite. From the results of analysis for the rainfall characteristics, it was found that landslide occurrence is closely related to the accumulated rainfall amount less than 3 days. Furthermore, it was found that regional difference of occurrence frequency is effected by 1-hour maximum rainfall intensity. From the results of analysis for the landslide data of 860 locations occurred in west side, it was shown that failure mode was changed from transition slide to liquidity slide. Occurrence frequency was high at the slope angle of 20~30°, slope length of 11~20m, and slope width of 6~10m. Landslide of west side is the typical landslide of Gneiss and Granite and the type of small scale which has narrow slope width.
